Modifying Thermostability and Reusability of Hyperthermophilic Mannanase by Immobilization on Glutaraldehyde Cross-Linked Chitosan Beads.

ABSTRACT: In the current study, the purified β-mannanase (Man/Cel5B) from Thermotoga maritima was immobilized on glutaraldehyde cross-linked chitosan beads. The immobilization of Man/Cel5B on chitosan beads was confirmed by Fourier-transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) analysis. After immobilization, the protein loading efficiency and immobilization yield were found to be 73.3% and 71.8%, respectively. The optimum pH for both free and immobilized enzymes was found to be pH 5.5. However, the optimum temperature of immobilized Man/Cel5B increased by 10 °C, from 85 °C (free Man/Cel5B) to 95 °C (Immobilized). The half-life of free and immobilized enzymes was found to be 7 h and 9 h, respectively, at 85 °C owing to the higher thermostability of immobilized Man/Cel5B. The increase in thermostability was also demonstrated by an increase in the energy of deactivation (209 kJmol^-1) for immobilized enzyme compared to its native form (92 kJmol^-1), at 85 °C. Furthermore, the immobilized Man/Cel5B displayed good operational stability as it retained 54% of its original activity after 15 repeated catalytic reactions concerning its free form.

Project description:The use of dyes at an industrial level has become problematic, since the discharge of dye effluents into water disturbs the photosynthetic activity of numerous aquatic organisms by reducing the penetration of light and oxygen, in addition to causing carcinogenic diseases and mutagenic effects in humans, as well as alterations in different ecosystems. Chitosan (CS) is suitable for removing anionic dyes since it has favorable properties, such as acquiring a positive charge and a typical macromolecular structure of polysaccharides. In this study, the optimization of CS beads crosslinked with glutaraldehyde (GA) for the adsorption of reactive blue dye 4 (RB4) in an aqueous solution was carried out. In this sense, the response surface methodology (RSM) was applied to evaluate the concentration of CS, GA, and sodium hydroxide on the swelling degree in the GA-crosslinked CS beads. In the same way, RSM was applied to optimize the adsorption process of the RB4 dye as a function of the initial pH of the solution, initial concentration of the dye, and adsorbent dose. The crosslinking reaction was investigated by scanning electron microscopy (SEM), Fourier transformed infrared spectroscopy (FTIR), and X-ray diffractometry (XRD). The design described for the swelling degree showed an R2 (coefficient of determination) adjusted of 0.8634 and optimized concentrations (CS 3.3% w/v, GA 1.7% v/v, and NaOH 1.3 M) that were conveniently applied with a concentration of CS at 3.0% w/v to decrease the viscosity and facilitate the formation of the beads. In the RB4 dye adsorption design, an adjusted R2 (0.8280) with good correlation was observed, where the optimized conditions were: pH = 2, adsorbent dose 0.6 g, and initial concentration of RB4 dye 5 mg/L. The kinetic behavior and the adsorption isotherm allowed us to conclude that the GA-crosslinked CS beads' adsorption mechanism was controlled mainly by chemisorption interactions, demonstrating its applicability in systems that require the removal of contaminants with similar structures to the model presented.

Project description:Owing to the great demand for using sustainable, renewable, and widely available materials in catalytic systems for the conversion of waste/toxic material to high value-added and harmless products, biopolymers derived from natural sources have demonstrated great promise as an alternative to state-of-the-art materials that suffer from high costs and limitations. These have encouraged us to design and fabricate a new super magnetization of Mn-Fe3O4-SiO2/amine-glutaraldehyde/chitosan bio-composite (MIOSC-N-et-NH2@CS-Mn) for advanced/aerobic oxidation process. The morphological and chemical characterization of the as-prepared magnetic bio-composite was assessed using ICP-OES, DR UV-vis, BET, FT-IR, XRD, FE-SEM, HR-TEM, EDS, and XPS techniques. The PMS + MIOSC-N-et-NH2@CS-Mn system was capable of degrading methylene orange (98.9% of removal efficiency) and selectively oxidizing ethylbenzene to acetophenone (conversion 93.70%, selectivity 95.10% and TOF 214.1 (103 h-1) within 8.0 min and 5.0 h, respectively. Moreover, MO was efficiently mineralized (TOC removal of ∼56.61) by MIOSC-N-et-NH2@CS-Mn with 60.4%, 5.20, 0.03 and 86.02% of the synergistic index, reaction stoichiometric efficiency, specific oxidant efficiency, and oxidant utilization ratio in wide pH ranges, respectively. An understanding of its vital parameters and relationship of catalytic activity with structural, environmental factors, leaching/heterogenicity test, long-term stability, inhibitory effect of anions in water matrix, economic study and response surface method (RSM) were evaluated in detail. Overall, the prepared catalyst could be employed as an environmentally friendly and low-cost candidate for the enhanced activation of PMS/O2 as an oxidant. Additionally, MIOSC-N-et-NH2@CS-Mn exhibited great stability, high recovery efficiency, and low metal leaching, which eliminated the harsh condition reaction and supplied practical application performance for water purification and selective aerobic oxidation of organic compounds.
