Light-Fueled Submarine-Like Droplet.

ABSTRACT: Flexibly and precisely manipulating 3D droplet transportation is a fundamental challenge for broad implications in diagnostics, drug delivery, bioengineering, etc. Herein, a light method is developed for manipulating a droplet to make it behave like a submarine. This light method enables flexible 3D transportation, stable suspension, and floating of a droplet, which can be freely altered. It is demonstrated that the localized photothermal effect induced thermocapillary flow in the water droplet/oil phase is responsible for energizing and manipulating the droplet. With such remarkable motility, the light-fueled submarine-like droplet successfully realizes various functions such as the acid-base detection, particle capture and transportation, and target crystal collection, dissolution and transportation. It is demonstrated that the light-fueled submarine-like droplet shows promising perspective for long-sought precise droplet manipulation in various applications.

p53 suppresses lipid droplet-fueled tumorigenesis through phosphatidylcholine.

Project description:Choline deficiency causes disorders including hepatic abnormalities and is associated with an increased risk of multiple types of cancer. Here, by choline-free diet-associated RNA-Seq analyses, we found that the tumor suppressor p53 drives the Kennedy pathway via PCYT1B to control the growth of lipid droplets (LDs) and their fueling role in tumorigenesis. Mechanistically, through upregulation of PCYT1B, p53 channeled depleted choline stores to phosphatidylcholine (PC) biosynthesis during choline starvation, thus preventing LD coalescence. Cells lacking p53 failed to complete this response to choline depletion, leading to hepatic steatosis and tumorigenesis, and these effects could be reversed by enforcement of PCYT1B expression or restoration of PC abundance. Furthermore, loss of p53 or defects in the Kennedy pathway increased surface localization of hormone-sensitive lipase on LDs to release specific fatty acids that fueled tumor cells in vivo and in vitro. Thus, p53 loss leads to dysregulation of choline metabolism and LD growth and couples perturbed LD homeostasis to tumorigenesis.

These properties derive from careful engineering of the energy maxima and minima along the threading coordinate and their light-triggered modulation. Unlike its precursor, this second-generation system is amenable to functionalization for integration into more complex devices.

A Soft Transporter Robot Fueled by Light.

Project description:Mobile organisms with ability for locomotion and transportation, such as humans and other animals, utilize orchestrated actuation to perform actions. Mimicking these functionalities in synthetic, light-responsive untethered soft-bodied devices remains a challenge. Inspired by multitasking and mobile biological systems, an untethered soft transporter robot with controlled multidirectional locomotion with the ability of picking up, transporting, and delivering cargo driven entirely by blue light is created. The soft robot design is an ensemble of light-responsive liquid crystalline polymers that can harness motion either collectively or individually to obtain a high degree of motion control for the execution of advanced tasks in a dry environment. Through orchestrated motion of the device's "legs", single displacement strides, which exceed 4 mm and can be taken in any direction, allow for locomotion around objects. Untethered cargo transportation is demonstrated by a pickup and release mechanism using the device's "arms". This strategy demonstrates the constructive harnessing of orchestrated motion in assemblies of established actuators, performing complex functions, mimicking constructive behavior seen in nature.

Pigmented Structural Color Actuators Fueled by Near-Infrared Light.

Project description:Cuttlefish can modify their body shape and both their pigmentary and structural colors for protection. This adaptability has inspired the development of appearance-changing polymers such as structural color actuators, although in most cases, the original shape has been confined to being flat, and pigmented structural color actuators have not yet been reported. Here, we have successfully created a pigmented structural color actuator using a cholesteric liquid crystal elastomer with a lower actuation temperature where both actuation and coloration (structural and pigmental) are tunable with temperature and NIR light. The shape, structural color, and absorption of the NIR-absorbing dye pigment of the actuator all change with temperature. Light can be used to trigger local in-plane bending actuation in flat films and local shape changes in a variety of 3D-shaped objects. A cuttlefish mimic that can sense light and respond by locally changing its appearance was also made to demonstrate the potential of pigmented structural color actuators for signaling and camouflage in soft robotics.

Light-Fueled Nanoscale Surface Waving in Chiral Liquid Crystal Networks.

Project description:Nano- and micro-actuating systems are promising for application in microfluidics, haptics, tunable optics, and soft robotics. Surfaces capable to change their topography at the nano- and microscale on demand would allow control over wettability, friction, and surface-driven particle motility. Here, we show that light-responsive cholesteric liquid crystal (LC) networks undergo a waving motion of their surface topography upon irradiation with light. These dynamic surfaces are fabricated with a maskless one-step procedure, relying on the liquid crystal alignment in periodic structures upon application of a weak electric field. The geometrical features of the surfaces are controlled by tuning the pitch of the liquid crystal. Pitch control by confinement allows engineering one-dimensional (1D) and two-dimensional (2D) structures that wave upon light exposure. This work demonstrates the potential that self-organizing systems might have for engineering dynamic materials, and harnessing the functionality of molecules to form dynamic surfaces, with nanoscale precision over their waving motion.

Light-fueled transient supramolecular assemblies in water as fluorescence modulators.

Project description:Dissipative self-assembly, which requires a continuous supply of fuel to maintain the assembled states far from equilibrium, is the foundation of biological systems. Among a variety of fuels, light, the original fuel of natural dissipative self-assembly, is fundamentally important but remains a challenge to introduce into artificial dissipative self-assemblies. Here, we report an artificial dissipative self-assembly system that is constructed from light-induced amphiphiles. Such dissipative supramolecular assembly is easily performed using protonated sulfonato-merocyanine and chitosan based molecular and macromolecular components in water. Light irradiation induces the assembly of supramolecular nanoparticles, which spontaneously disassemble in the dark due to thermal back relaxation of the molecular switch. Owing to the presence of light-induced amphiphiles and the thermal dissociation mechanism, the lifetimes of these transient supramolecular nanoparticles are highly sensitive to temperature and light power and range from several minutes to hours. By incorporating various fluorophores into transient supramolecular nanoparticles, the processes of aggregation-induced emission and aggregation-caused quenching, along with periodic variations in fluorescent color over time, have been demonstrated. Transient supramolecular assemblies, which act as fluorescence modulators, can also function in human hepatocellular cancer cells.

Light-Fueled Primitive Replication and Selection in Biomimetic Chemical Systems.

Project description:The concept of chemically evolvable replicators is central to abiogenesis. Chemical evolvability requires three essential components: energy-harvesting mechanisms for nonequilibrium dissipation, kinetically asymmetric replication and decomposition pathways, and structure-dependent selective templating in the autocatalytic cycles. We observed a UVA light-fueled chemical system displaying sequence-dependent replication and replicator decomposition. The system was constructed with primitive peptidic foldamer components. The photocatalytic formation-recombination cycle of thiyl radicals was coupled with the molecular recognition steps in the replication cycles. Thiyl radical-mediated chain reaction was responsible for the replicator death mechanism. The competing and kinetically asymmetric replication and decomposition processes led to light intensity-dependent selection far from equilibrium. Here, we show that this system can dynamically adapt to energy influx and seeding. The results highlight that mimicking chemical evolution is feasible with primitive building blocks and simple chemical reactions.

Light-Fueled Hydrogel Actuators with Controlled Deformation and Photocatalytic Activity.

Project description:Hydrogel actuators have shown great promise in underwater robotic applications as they can generate controllable shape transformations upon stimulation due to their ability to absorb and release water reversibly. Herein, a photoresponsive anisotropic hydrogel actuator is developed from poly(N-isopropylacrylamide) (PNIPAM) and gold-decorated carbon nitride (Au/g-C3 N4 ) nanoparticles. Carbon nitride nanoparticles endow hydrogel actuators with photocatalytic properties, while their reorientation and mobility driven by the electrical field provide anisotropic properties to the surrounding network. A variety of light-fueled soft robotic functionalities including controllable and programmable shape-change, gripping, and locomotion is elicited. A responsive flower-like photocatalytic reactor is also fabricated, for water splitting, which maximizes its energy-harvesting efficiency, that is, hydrogen generation rate of 1061.82 µmol g-1 h-1 , and the apparent quantum yield of 8.55% at 400 nm, by facing its light-receiving area adaptively towards the light. The synergy between photoactive and photocatalytic properties of this hydrogel portrays a new perspective for the design of underwater robotic and photocatalytic devices.

Multiple Light Control Mechanisms in ATP-Fueled Non-equilibrium DNA Systems.

Project description:Fuel-driven self-assemblies are gaining ground for creating autonomous systems and materials, whose temporal behavior is preprogrammed by a reaction network. However, up to now there has been a lack of simple external control mechanisms of the transient behavior, at best using remote and benign light control. Even more challenging is to use different wavelengths to modulate the reactivity of different components of the system, for example, as fuel or building blocks. Success would enable such systems to navigate along different trajectories in a wavelength-dependent fashion. Herein, we introduce the first examples of light control in ATP-fueled, dynamic covalent DNA polymerization systems organized in an enzymatic reaction network of concurrent ATP-powered ligation and restriction. We demonstrate concepts for light activation and modulation by introducing caged ATP derivatives and caged DNA building blocks, making it possible to realize light-activated fueling, self-sorting in structure and behavior, and transition across different wavelength-dependent dynamic steady states.
