New Species of Talaromyces (Trichocomaceae, Eurotiales) from Southwestern China.

ABSTRACT: Species of Talaromyces are cosmopolitan and ubiquitous, and some are of industrial and medicinal importance. Species of Talaromyces have been successively reported in China. During our examinations of samples collected from southwestern China, two new species belonging to Talaromyces sect. Talaromyces were further discovered based on phylogenetic analyses and morphological comparisons. Talaromyces ginkgonis sp. nov., isolated from a partially colonized fruit of Ginkgo biloba, differs from closely-related fungi in the combination of conidia ellipsoidal, smooth and 3.5-4 × 2-3 μm, no growth on CYA at 37 °C and sequence divergences; T. shilinensis sp. nov. is distinguished from its related allies in the combination of smooth conidia, colonies 10-11 mm diameter on CYA at 25 °C and sequence differences. Detailed descriptions and illustrations of the new taxa are given.

Project description:During a previous study on microfungi associated with clematis roots, Penicillium-like fungi were isolated and identified based on morphology. In this study, we subjected those strains to a detailed examination which led to the proposal of two taxonomic novelties, named Rasamsonia chlamydospora and Talaromyces clematidis. The first taxon is characterized by rough-walled mycelium, acerose to flask shaped phialides, cylindrical conidia and by production of chlamydospore-like structures. The four-loci-based phylogeny analysis delineated the taxon as a taxonomic novelty in Rasamsonia. Talaromyces clematidis is characterized by restricted growth on Czapek yeast extract agar, dichloran 18% glycerol agar and yeast extract sucrose agar, and production of yellow ascomata on oatmeal agar. Phylogenetic analyses placed this taxon as a taxonomic novelty in Talaromyces sect. Bacillispori. Both taxa are introduced here with detailed descriptions, photoplates and information on their phylogenetic relationship with related species.

Characterization and phylogenetic analysis of the Talaromyces liani (kamyschko) Yilmaz, Frisvad & Samson, 2014 (Eurotiales: trichocomaceae) mitochondrial genome.

Project description:The filamentous fungus Talaromyces liani (Kamyschko) Yilmaz, Frisvad & Samson, 2014, has attracted considerable interest in biotechnology due to its diverse industrial applications and physiological characteristics. However, the mitochondrial genome of T. liani remains uncharacterized. Here, we present the complete mitochondrial genome of T. liani, comprising 38,000 bp with a GC content of 24.61%. This genome includes 15 core protein-coding genes, 4 independent ORFs, 6 intronic ORFs, 26 tRNAs, and 2 rRNA genes. Phylogenetic analysis using Bayesian inference (BI) revealed the evolutionary relationships among 15 fungi from Eurotiales, strongly supporting distinct clades and indicating that T. liani most closely related to T. pinophilus.

A New Species and Five New Records of Talaromyces (Eurotiales, Aspergillaceae) Belonging to Section Talaromyces in Korea.

Project description:Talaromyces is a genus within the phylum Ascomycota (class Eurotiomycetes, order Eurotiales, family Trichocomaceae). Many species in this genus are known to produce diverse secondary metabolites with great potential for agricultural, medical, and pharmaceutical applications. During a survey on fungal diversity in the genus Talaromyces in Korea, six strains were isolated from soil, indoor air, and freshwater environments. Based on morphological, physiological, and multi-locus (ITS, BenA, CaM, and RPB2) phylogenetic analyses, we identified five previously unrecorded species in Korea (T. brevis, T. fusiformis, T. muroii, T. ruber, and T. soli) and a new species (T. echinulatus sp. nov.) belonging to section Talaromyces. Herein, detailed descriptions, illustrations, and phylogenetic tree are provided.

Three new species of Talaromyces sect. Talaromyces discovered from soil in China.

Project description:Three new Talaromyces species isolated from soil are reported here, namely T. dimorphus (ex-type strain AS3.15692 T), T. lentulus (ex-type strain AS3.15689 T) and T. mae (ex-type strain AS3.15690 T). T. dimorphus is characterized by biverticillate and monoverticillate penicilli, ampulliform phialides, slimy texture with sparse mycelial funicles and absent conidiogenesis on MEA. T. lentulus is featured by vivid yellow mycelium on Cz and MEA, absent conidiogenesis on CYA, and globose smooth-walled conidia. T. mae presents sparse conidia on CYA and YES, funiculous and floccose texture on MEA, and ovoid smooth-walled conidia. Both morphological and molecular characters show that T. dimorphus is unique and has no close relatives. Although T. lentulus and T. mae resembles T. adpressus and T. pinophilus very much, phylogenetic analyses of CaM, BenA, ITS and Rpb2 sequences all support their status as novel species.

New Talaromyces species from indoor environments in China.

Project description:Talaromyces contains both asexual and sexually reproducing species. This genus is divided in seven sections and currently has 105 accepted species. In this study we investigated the Talaromyces isolates that were obtained during a study of indoor air collected in Beijing, China. These indoor Talaromyces strains are resolved in four sections, seven of them are identified as T. islandicus, T. aurantiacus, T. siamensis and T. albobiverticillius according to BenA sequences, while 14 isolates have divergent sequences and are described here as nine new species. The new species are placed in four sections, namely sections Helici, Islandici, Talaromyces and Trachyspermi. They are described based on sequence data (ITS, BenA, CaM and RPB2) in combination with phenotypic and extrolite characters. Morphological descriptions and notes for distinguishing similar species are provided for each new species. The recently described T. rubrifaciens is synonymised with T. albobiverticillius based on presented phylogenetic results.

Dysosmavillosa (Berberidaceae), a new species from Guizhou, Southwestern China.

Project description:A new species, Dysosmavillosa Z.W.Wang & H.C.Xi, is described and illustrated based on collections from the Yueliang Mountains in Congjiang County of Guizhou, Southwestern China. It is morphologically similar to D.difformis (Hemsl. & E.H.Wilson) T.H.Wang ex T.S.Ying, but can be easily distinguished from D.difformis by its inflorescences bearing a greater number of flowers (5-13 vs. 2-5), white-villous stems, petioles, and abaxial leaf blade, and stigma dark purple-red. In addition, we also compare this new species to the three species of Podophyllum (i.e., P.glaucescens J.M.H.Shaw, P.hemsleyi J.M.H.Shaw & Stearn, and P.trilobulum J.M.H.Shaw) which are insufficiently known and listed as putative members of Dysosma in Flora of China. Dysosmavillosa can also be easily distinguished from P.glaucescens (7-flowered; to 40 cm tall) and P.hemsleyi (4-flowered; to 40 cm tall) by its inflorescences bearing a greater number of flowers, relatively smaller stature (9-23 cm tall), stems, petioles and abaxial leaf blade densely white-villose. Although the stems and petioles of P.trilobulum also possess fine short hairs, it can be easily distinguished from D.villosa by its trilobulate leaves, inflorescence with fewer flowers (2-5), and the position of inflorescence (inserted at or above midpoint on petiole of upper leaf).

Two New Sexual Talaromyces Species Discovered in Estuary Soil in China.

Project description:In the survey of mycobiota of mudflats in China, two new sexually reproducing Talaromyces sect. Talaromyces species were discovered and studied using a polyphasic approach. These species are named here Talaromyces haitouensis (ex-type AS3.160101T) and Talaromyces zhenhaiensis (ex-type AS3.16102T). Morphologically, T. haitouensis is distinguished by moderate growth, green-yellow gymnothecia, orange-brown mycelium, and echinulate ellipsoidal ascospores. T. zhenhaiensis is characterized by fast growth, absence of sporulation, cream yellow to naphthalene yellow gymnothecia and mycelium, and smooth-walled ellipsoidal ascospores with one equatorial ridge. The two novelties are further confirmed by phylogenetic analyses based on either individual sequences of BenA, CaM, Rpb2, and ITS1-5.8S-ITS2 or the concatenated BenA-CaM-Rpb2 sequences.

Four New Species of Hemileccinum (Xerocomoideae, Boletaceae) from Southwestern China.

Project description:The genus Hemileccinum belongs to the subfamily Xerocomoideae of the family Boletaceae. In this study, phylogenetic inferences of Hemileccinum based on sequences of a single-locus (ITS) and a multi-locus (nrLSU, tef1-α, rpb1, rpb2) were conducted. Four new species, namely H. abidum, H. brevisporum, H. ferrugineipes and H. parvum were delimited and proposed based on morphological and molecular evidence. Descriptions and line-drawings of them were presented, as well as their comparisons to allied taxa. Our study shed new light on the recognition of the genus. The pileipellis of the species in this genus should mostly be regarded as (sub)epithelium to hyphoepithelium, because the pileipellis of most studied species here is composed of short inflated cells in the inner layer (subpellis) and filamentous hyphae in outer layer (suprapellis). The basidiospores of the studied species, including the type species, H. impolitum, have a warty surface.

Veronicastrum wulingense (Plantaginaceae), a new species from Southwestern Hubei, China.

Project description:BackgroundThe genus Veronicastrum Heist. ex Fabr. are mainly distributed in East Asia, and only Veronicastrum virginicum (L.) Farw. is disjunctively distributed in eastern North America. The south area of China (extending to Taiwan Island) is the richest in Veronicastrum species. It is of medicinal importance in China as traditional herbs used to treat ascites diseases that caused by schistosomiasis. During field investigation of plant resources in Pingbaying National Forest Park, Southwestern Hubei, China, an unknown flowering population of Veronicastrum was discovered from thick humus layers adjacent to rocks under broad-leaved forests by walkways. They were collected and morphological characters assesed for further taxonomic treatment. Molecular analysis was also conducted to ascertain its phylogenetic position in the genus Veronicastrum.ResultsThis species is similar to Veronicastrum liukiuense (Ohwi) T.Yamaz. from the Ryukyu Islands, but can be distinctly differed by its axillary inflorescences (versus terminal on short leafy branches), pedicels up to 2.5 mm (versus sessile), corollas purple to purple-red (versus white tinged with pale purple) and florescence June to July (versus September to October). Also, phylogenetic studies showed the species was an independent clade in the genus Veronicastrum based on the maximum likelihood (ML) analyses using two different matrix sequences of concatenated molecular markers. The plastid genome of this new species is also reported in this study for the first time.ConclusionThe morphological and molecular evidences support the recognition of Veronicastrum wulingense as a new species.
