A Catalogue of Orthogonal Complementary Ligand Pairings for Palladium(II) Complexes.

ABSTRACT: Molecular recognition is a form of information transfer, seen in the base pairing in DNA which is derived from the identity (acceptor or donor) and number of hydrogen bond sites within each base. Here we report bis-ligand palladium(II) complexes that exhibit similar complementarity. Pd(II) has square planar four-coordinate geometry, giving control over ligand orientation and denticity. Pairings were developed using ligand denticity (3 : 1 or 2 : 2), and hydrogen bond capability (AA:DD or AD:DA) or lack thereof. Five pairings were investigated, with two sets of four being found fully orthogonal. The two 3 : 1 pairings exhibited limited ligand exchange. The extent of this exchange varied dependant on solvent from 2 : 1 (desired to undesired) to 6 : 1. A reliable and varied set of ligand pairs have therefore been developed for bis-ligand coordination sphere engineering in pursuit of sorting for complex molecular architectures and molecular-level information storage and transfer events.

Synthesis and Characterization of Zinc(II), Cadmium(II), and Palladium(II) Complexes with the Thiophene-Derived Schiff Base Ligand.

Project description:Zn(II), Pd(II), and Cd(II) complexes, [L TH MCl 2 ] (M = Zn, Pd; X = Br, Cl) and [L TH Cd(μ-X)X] n (X = Cl, Br; n = n, 2), supported by the (E)-N 1,N 1-dimethyl-N 2-(thiophen-2-ylmethylene)ethane-1,2-diamine (L TH ) ligand are synthesized and structurally characterized. Density functional theory (DFT) electronic structure calculations and variable-temperature NMR support the presence of two conformers and a dynamic interconversion process of the minor conformer to the major one in solution. It is found that the existence of two relevant complex conformers and their respective ratios in solution depend on the central metal ions and counter ions, either Cl- or Br-. Among the two relevant conformers, a single conformer is crystallized and X-ray diffraction analysis revealed a distorted tetrahedral geometry for Zn(II) complexes, and a distorted square planar and square pyramidal geometry for Pd(II) and Cd(II) complexes, respectively. It is shown that [L TH MCl 2 ]/LiO i Pr (M = Zn, Pd) and [L TH Cd(μ-Cl)Cl] n /LiO i Pr can effectively catalyze the ring-opening polymerization (ROP) reaction of rac-lactide (rac-LA) with 94% conversion within 30 s with [L TH ZnCl 2 ]/LiO i Pr at 0 °C. Overall, hetero-enriched poly(lactic acid)s (PLAs) were provided by these catalytic systems with [L TH ZnCl 2 ]/LiO i Pr producing PLA with higher heterotactic bias (P r up to 0.74 at 0 °C).

Ligand Rearrangement Leads to Tetrahydrothiophene-Functionalized <i>N,S</i>-Heterocyclic Carbene Palladium(II) Complexes.

Project description:Tetrahydrothiophene-functionalized N,S-heterocyclic carbene palladium(II) complexes are synthesized through an unexpected rearrangement that proceeds with palladium(II) trifluoroacetate and not with palladium(II) acetate, palladium(II) bromide, or palladium(II) chloride. A series of these complexes were isolated and characterized by X-ray crystallography. The mechanism of formation of these [3.2.1]-palladabicycles was explored, and the catalytic capabilities of these complexes were demonstrated in representative C-C coupling reactions.

Reversible Ligand Pairing and Sorting Processes Leading to Heteroligated Palladium(II) Complexes with Hemilabile Ligands.

Project description:Halide-induced ligand pairing and sorting processes have been observed in the context of Pd(II) complexes with hemilabile P,S and P,O ligands. Mixing of the ligands Ph2PCH2CH2SMe (7) and Ph2PCH2CH2SPh (8) with a Pd(II) precursor in CH2Cl2 results in a mixture of [(7)2ClPd]Cl, [(8)2Cl2Pd], and [(7)(8)ClPd]Cl complexes at 20 °C. This equilibrium can be driven toward the heteroligated structure [(7)(8)ClPd]Cl by (1) cooling the mixture or (2) precipitation with hexanes, leading to the exclusive formation of semiopen heteroligated complex cis-[κ 2-(7)-κ 1-(8)ClPd]Cl (9a), as confirmed by a single-crystal X-ray diffraction study and solid state CPMAS 31P{1H} NMR spectroscopy. Dissolution of 9a in CH2Cl2 leads to the original mixture of complexes, which illustrates the reversible nature of this ligand pairing and sorting process. Similar processes occur when a combination of P,S and P,O ligands is used. The semiopen heteroligated complexes can be chemically manipulated in a reversible fashion to form closed complexes, allowing for control of the relative position and flexibility between neighboring substituents in these "tweezer"-like structures. Control experiments suggest these ligand sorting and pairing processes occur via a halide-induced ligand rearrangement (HILR) reaction.

Ligand Steric Effects of α-Diimine Nickel(II) and Palladium(II) Complexes in the Suzuki-Miyaura Cross-Coupling Reaction.

Project description:Nickel catalysts represent a low cost and environmentally friendly alternative to palladium-based catalytic systems for Suzuki-Miyaura cross-coupling (SMC) reactions. However, nickel catalysts have suffered from poor air, moisture, and thermal stabilities, especially at high catalyst loading, requiring controlled reaction conditions. In this report, we examine a family of mono- and dinuclear Ni(II) and Pd(II) complexes with a diverse and versatile α-diimine ligand environment for SMC reactions. To evaluate the ligand steric effects, including the bite angle in the reaction outcomes, the structural variation of the complexes was achieved by incorporating iminopyridine- and acenaphthene-based ligands. Moreover, the impact of substrate bulkiness was investigated by reacting various aryl bromides with phenylboronic acid, 2-naphthylboronic acid, and 9-phenanthracenylboronic acid. Yields were the best with the dinuclear complex, being nearly quantitative (93-99%), followed by the mononuclear complexes, giving yields of 78-98%. Consequently, α-diimine-based ligands have the potential to deliver Ni-based systems as sustainable catalysts in SMC.

Rational Design of Orthogonal Multipolar Interactions with Fluorine in Protein-Ligand Complexes.

Project description:Multipolar interactions involving fluorine and the protein backbone have been frequently observed in protein-ligand complexes. Such fluorine-backbone interactions may substantially contribute to the high affinity of small molecule inhibitors. Here we found that introduction of trifluoromethyl groups into two different sites in the thienopyrimidine class of menin-MLL inhibitors considerably improved their inhibitory activity. In both cases, trifluoromethyl groups are engaged in short interactions with the backbone of menin. In order to understand the effect of fluorine, we synthesized a series of analogues by systematically changing the number of fluorine atoms, and we determined high-resolution crystal structures of the complexes with menin. We found that introduction of fluorine at favorable geometry for interactions with backbone carbonyls may improve the activity of menin-MLL inhibitors as much as 5- to 10-fold. In order to facilitate the design of multipolar fluorine-backbone interactions in protein-ligand complexes, we developed a computational algorithm named FMAP, which calculates fluorophilic sites in proximity to the protein backbone. We demonstrated that FMAP could be used to rationalize improvement in the activity of known protein inhibitors upon introduction of fluorine. Furthermore, FMAP may also represent a valuable tool for designing new fluorine substitutions and support ligand optimization in drug discovery projects. Analysis of the menin-MLL inhibitor complexes revealed that the backbone in secondary structures is particularly accessible to the interactions with fluorine. Considering that secondary structure elements are frequently exposed at protein interfaces, we postulate that multipolar fluorine-backbone interactions may represent a particularly attractive approach to improve inhibitors of protein-protein interactions.

Crystal Design, Antitumor Activity and Molecular Docking of Novel Palladium(II) and Gold(III) Complexes with a Thiosemicarbazone Ligand.

Project description:The current research describes the synthesis and characterization of 2-acetylpyridine N(4)-cyclohexyl-thiosemicarbazone ligand (HL) and their two metal complexes, [Au(L)Cl][AuCl2] (1) and [Pd(L)Cl]·DMF (2). The molecular structures of the compounds were determined by physicochemical and spectroscopic methods. Single crystal X-ray diffraction was employed in the structural elucidation of the new complexes. The complexes showed a square planar geometry to the metal center Au(III) and Pd(II), coordinated with a thiosemicarbazone molecule by the NNS-donor system and a chloride ion. Complex (1) also shows the [AuCl2]- counter-ion in the asymmetric unit, and complex (2) has one DMF solvent molecule. These molecules play a key role in the formation of supramolecular structures due to different interactions. Noncovalent interactions were investigated through the 3D Hirshfeld surface by the dnorm function and the 2D fingerprint plots. The biological activity of the compounds was evaluated in vitro against the human glioma U251 cells. The cytotoxicity results revealed great antitumor activity in complex (1) compared with complex (2) and the free ligand. Molecular docking simulations were used to predict interactions and properties with selected proteins and DNA of the synthesized compounds.

Project description:Metal-mediated self-assembly of isocyanides and methyl 4-aminopyrimidine-5-carboxylate leads to luminescent PdII and PtII complexes featuring C,N-cyclometalated acyclic diaminocarbene (ADC) ligands. The solid-state luminescent properties of these diaminocarbene derivatives are attributed to their triplet-state metal/metal-to-ligand charge-transfer (3MMLCT) nature, which is driven by attractive intermolecular M···M interactions further reinforced by the intramolecular π-π interactions even in the structure of the Pd compound, which is the first Pd-ADC phosphor reported.

Copper-, palladium-, and platinum-containing complexes of an asymmetric dinucleating ligand.

Project description:The coordination chemistry of an asymmetric dinucleating hexadentate ligand LH(2) comprising neutral alkyltriamine and potentially dianionic dicarboxamido-pyridyl donor sets with copper, palladium, and platinum has been explored. Monometallic, dicopper, and heterodinuclear Cu-Pd and -Pt complexes have been prepared and characterized, including by NMR, EPR, UV-vis, and IR spectroscopy and X-ray crystallography. For example, the monometallic complexes [(LH(2))MCl]X (M = Cu, X = OTf; M = Pd or Pt, X = Cl) were prepared, wherein the metal(II) ions are coordinated to the triamine portion and the pyridyldicarboxamide is unperturbed. Treatment of LH(2) with [MesCu](x) (Mes = mesityl) provided a monocopper(I) complex, again with the metal coordinated only to the trialkylamine donor set. Reaction of [(LH(2))CuCl]OTf with NaOMe resulted in an unexpected migration of the copper(II)-chloride fragment to the pyridyldicarboxamide site to yield Na[LCuCl], from which a dicopper complex LCu(2)Cl(2) and mixed-metal complexes LCu(Cl)M(Cl) (M = Pd, Pt) were prepared by addition of CuCl(2) or MCl(2), respectively. The heterodinuclear complexes were also prepared by addition of CuCl(2) to [(LH(2))MCl]Cl.

Synthesis, characterization and cytotoxicity studies of palladium(II)-proflavine complexes.

Project description:An investigation of the reaction of Pd(II) complexes with proflavine (3,6-diaminoacridine) resulted in the isolation of the compounds [Pd(terpy)(proflavine)](NO(3))(HSO(4))*3H(2)O, 1, (terpy = 2,2':6',2″-terpyridine), [Pd(en)(proflavineH))](NO(3))(SO(4)), 2, (en = ethylenediamine), and [Pd(proflavineH)Cl(2)](SO(4))(0.5)*H(2)O, 3. They have been isolated and characterized by NMR, IR, and electro-spray ionization mass spectrometry techniques and by elemental analyses. The proflavine was bonded to the Pd(II) through the endocyclic nitrogen in 1, but through the proflavine NH(2) in 2. Compound 3 appeared to be polymeric in the solid state with a 1:1 mole ratio of Pd(II):proflavine. Upon solution of 3 in DMSO, two unique species were formed. In one species the Pd(II) was bonded to two proflavines through the endocyclic nitrogen (1:2 mole ratio) and in the other species, a Pd(II) was bonded to each NH(2) group of a single proflavine (2:1 mole ratio). Molecular modeling of the equilibrium geometry by Spartan 8 produced structures which were consistent with the experimental data on the solutions of the three compounds. In vitro cytotoxicity testing against two breast cancer cell lines and one ovarian cancer cell line showed that compounds 1 and 3 had significant activity.

Binuclear palladium(I) and palladium(II) complexes of ortho-functionalized 1,3-bis(aryl)triazenido ligands.

Project description:Three new bis(aryl)triazene ligands, Ar-NNNH-Ar' [Ar = o-C(6)H(4)-CO(2)Me, Ar' = p-C(6)H(4)-CH(3) (2); Ar = Ar' = o-C(6)H(4)-CO(2)Me (3); Ar = o-C(6)H(4)-SMe, Ar' = p-C(6)H(4)-CH(3)) (4)], have been synthesized. The reaction of 1-4 with PdCl(2)(NCCH(3))(2) in the presence of a base afforded a series of binuclear diamagnetic palladium complexes. In these reactions, ligands 1-3 afforded the palladium(I) complexes [Pd(I)(o-MeO(2)C-C(6)H(4)-NNN-o-C(6)H(4)-CO(2)Me)](2) (5, monoclinic, space group P21/c, a = 8.6070(10) Angstrom, b = 14.3220(10) Angstrom, c = 12.7310(10) Angstrom, beta = 100.2950(10) degrees, Z = 2), [Pd(I)(o-MeO-C(6)H(4)-NNN-o-C(6)H(4)-OMe)](2) (6, triclinic, space group P, a = 6.6288(5) Angstrom, b = 10.2631(10) Angstrom, c = 11.0246(11) Angstrom, alpha = 85.579(6) degrees, beta = 80.885(6) degrees, gamma = 74.607(6) degrees, Z = 1), and [Pd(I)(o-MeO(2)C-C(6)H(4)-NNN-p-C(6)H(4)-CH(3))](2) (7, tetragonal, space group I41/a, a = 20.866(3) Angstrom, b = 20.866(3) Angstrom, c = 13.156(2) Angstrom, Z = 8). In contrast, the reaction of ligand 4 with PdCl(2)(NCCH(3))(2) resulted in the formation of a palladium(II) dimer, [Pd(II)(o-MeS-C(6)H(4)-NNN-p-C(6)H(4)-CH(3))Cl](2) (8, orthorhombic, space group P2(1)2(1)2, a = 10.4058(5) Angstrom, b = 16.2488(8) Angstrom, c = 9.9500(5) Angstrom, Z = 2).
