Risk assessment for the native anurans from an alien invasive species, American bullfrogs (Lithobates catesbeianus), in South Korea.

ABSTRACT: The invasive species are of global concern, and the Invasive American Bullfrog (IAB; Lithobates catesbeianus) is one of the worst invasive amphibian species worldwide. Like other countries, South Korea is also facing challenges from IAB. Although many studies indicated impacts of IAB on native anurans in Korea, the actual risk at the specific level is yet to evaluate. Considering the putative invasiveness of IAB, it is hypothesized that any species with the possibility of physical contact or habitat sharing with them, will have a potential risk. Thus, we estimated and observed their home range, preferred habitats, morphology, behavior, and ecology. Then, comparing with existing knowledge, we assessed risks to the native anurans. We found a home range of 3474.2 ± 5872.5 m² and identified three types of habitats for IAB. The analyses showed at least 84% of native anurans (frogs and toads) were at moderate to extreme risks, which included all frogs but only 33% of toads. Finally, we recommended immediate actions to conserve the native anurans based on our results. As this study is the first initiative to assess the specific risk level from the invasiveness of L. catesbeianus, it will help the managers to set conservation priorities and strategies.

Project description:Poor utilization efficiency of plant protein diets always leads to intestinal barrier dysfunction and growth inhibition in animals. Probiotics have shown promise in improving growth performance and gut health of the host. However, obtaining the host-beneficial probiotic from thousands of bacterial phylotypes is challenging. Here, four intestinal autochthonous bacteria were isolated from fast-growing bullfrog after a 60-day feeding on a soybean meal (SM)-based diet. Another feeding trial was conducted to evaluate the effects of supplementing these strains in an SM-based diet on growth, nutrient digestibility, immunity, and gut health of bullfrog. A high-SM basal diet was used as a non-supplemented control group (NC), and four other diets were prepared by supplementing the basal diet with 1 × 107 CFU/g of Bacillus siamensis, Bacillus tequilensis (BT), Bacillus velezensis, and Lactococcus lactis (LL). Results showed that weight gain, feed efficiency, nitrogen retention, and apparent digestibility coefficients of dry matter and protein were significantly higher in the LL group compared with the NC group (p < 0.05). Furthermore, compared with the NC group, both BT and LL groups showed markedly higher jejunal protease and amylase activities, serum complement 4 and immunoglobulin M levels, jejunal muscularis thickness (p < 0.05), and up-regulated expression of il-10 and zo-1 genes (p < 0.05). High-throughput sequencing revealed higher abundances of Bacillus and Cetobacterium in BT and LL groups, respectively, accompanied with decreased abundances of Enterobacter and Escherichia-Shigella. Besides, KEGG pathways related to metabolisms were significantly enhanced by the LL diet relative to the NC diet (p < 0.05). Overall, the beneficial effects of two frog-derived probiotics were determined: supplementation of L. lactis in SM-based diet promoted growth and nutrient digestibility; both B. tequilensis and L. lactis supplementation improved immune response and intestinal barrier function of bullfrogs.

Project description:In this study, we analysed the regional distribution status, species composition differences, seasonal occurrence and habitat types of invasive alien insects that are distirbuted in natural ecosystems nationwide, targeting 3,802 locations in eight regions between 2019 and 2021. A total of 95,017 invasive alien insects belonging to nine orders, 48 families and 77 species were identified. Amongst the taxonomic groups, Hemiptera (35.1%) was dominant, followed by Coleoptera (18.2%) and Lepidoptera (14.3%). Gyeongsangnam-do had the highest percentage of invasive alien insects, with 55 species, while Gyeonggi-do had the highest number of invasive alien insects at 890 sites. We used Non-metric multidimensional scaling (NMDS) to analyse differences in invasive alien insect species composition by region, occurrence by season and habitat type. To compare the differences in invasive alien insect species composition by region, we divided them into four groups (Group 1: Jeollanam-do, Jeollabuk-do; Group 2: Chungcheongnam-do, Chungcheongbuk-do; Group 3: Gyeongsangnam-do, Gyeongsangbuk-do; and Group 4: Gyeonggi-do, Gangwon-do). We found an extensive overlap in invasive alien insects composition by region. Additionally, species composition exhibited seasonal differences, with the highest number of invasive alien insects occurring between July and September. Species occurring in spring (April to May) included Dryocosmuskuriphilus, Hyperapostica and Brachyperazoilus, whereas those occurring in autumn (September to October) included Vespavelutinanigrithorax, Thecodiplosisjaponensis and Hermetiaillucens. Habitat type analysis showed that invasive alien insects lived along roadsides (31.9%), farmlands (27.9%) and parks (19.4%), exhibiting high densities in anthropogenic and disturbed sites, such as parks, residences and farmlands. Ecological statistical analysis showed that the dominance index was 0.6 in Chungcheongbuk-do, the diversity index was 2.7 in Gyeongsangbuk-do, the abundance index was 5.4 in Gyeongsnagnam-do and the equality index was 0.6-0.7 in all regions. Therefore, we aimed to identify the habitat status of introduced and settled invasive insects to provide a basis for selecting primary management target species.

Project description:The invasive American bullfrog (Lithobates catesbeianus) imperils freshwater biodiversity worldwide. Effective management hinges on early detection of incipient invasions and subsequent rapid response, as established populations are extremely difficult to eradicate. Although environmental DNA (eDNA) detection methods provide a highly sensitive alternative to conventional surveillance techniques, extensive testing is imperative to generate reliable output. Here, we tested and compared the performance of two primer/probe assays to detect and quantify the abundance of bullfrogs in Western Europe in silico and in situ using digital droplet PCR (ddPCR). Although both assays proved to be equally target-specific and sensitive, one outperformed the other in ddPCR detection resolution (i.e., distinguishing groups of target-positive and target-negative droplets), and hence was selected for further analyses. Mesocosm experiments revealed that tadpole abundance and biomass explained 99% of the variation in eDNA concentration. Because per individual eDNA emission rates did not differ significantly among tadpoles and juveniles, and adults mostly reside out of the water, eDNA concentration can be used as an approximation of local bullfrog abundance in natural populations. Seasonal eDNA patterns in three colonized ponds showed parallel fluctuations in bullfrog eDNA concentration. An increase in eDNA concentration was detected in spring, followed by a strong peak coinciding with the breeding season (August, September or October), and continuously low eDNA concentrations during winter. With this study, we report the validation process required for appropriately implementing eDNA barcoding analyses in lentic systems. We demonstrate that this technique can serve as a solid and reliable tool to detect the early stages of bullfrog invasions and to quantify temporal changes in abundance that will be useful in coordinating large-scale bullfrog eradication programs and evaluating their efficiency.
