Direct force measurement of microscopic droplets pulled along soft surfaces.

ABSTRACT: When a droplet is placed on a soft surface, surface tension deforms the substrate, creating a capillary ridge. We study how the motion of the ridge dissipates energy in microscopic droplets. Using a micropipette based method, we are able to simultaneously image and measure forces on a microscopic droplet moving at a constant speed along a soft film supported on a rigid substrate. Changing the thickness of the thin film tunes the effective stiffness of the substrate. Thus we can control the ridge size without altering the surface chemistry. We find that the dissipation depends strongly on the film thickness, decreasing monotonically as effective stiffness increases. This monotonic trend is beyond the realm of small deformation theory, but can be explained with a simple scaling analysis.

Elasticity and damping ratio measurement of droplets on super-hydrophobic surfaces.

Project description:The measurement of the droplets' elasticity is vitally important in microfluidic and ink-jet printing. It refers to the ability of the droplet to restore its original shape and strong robustness. This study investigated a novel method to measure elasticity. The plate coated with super-hydrophobic layers pressed on a droplet and the elastic force was recorded by an electronic balance. Meanwhile, a mathematical model was constructed to calculate the changes of the droplet area under the force. The measurement showed that external work mainly converts into surface energy and the damping ratio increases from 0.068 to 0.261 with the increase of mass fraction from 0 to 80 wt%. It also indicates that the novel method can accurately and efficiently measure the elasticity of droplets.

Direct and accurate measurement of size dependent wetting behaviors for sessile water droplets.

Project description:The size-dependent wettability of sessile water droplets is an important matter in wetting science. Although extensive studies have explored this problem, it has been difficult to obtain empirical data for microscale sessile droplets at a wide range of diameters because of the flaws resulting from evaporation and insufficient imaging resolution. Herein, we present the size-dependent quantitative change of wettability by directly visualizing the three phase interfaces of droplets using a cryogenic-focused ion beam milling and SEM-imaging technique. With the fundamental understanding of the formation pathway, evaporation, freezing, and contact angle hysteresis for sessile droplets, microdroplets with diameters spanning more than three orders of magnitude on various metal substrates were examined. Wetting nature can gradually change from hydrophobic at the hundreds-of-microns scale to super-hydrophobic at the sub-μm scale, and a nonlinear relationship between the cosine of the contact angle and contact line curvature in microscale water droplets was demonstrated. We also showed that the wettability could be further tuned in a size-dependent manner by introducing regular heterogeneities to the substrate.

Suppression of wetting transition on evaporative fakir droplets by using slippery superhydrophobic surfaces with low depinning force.

Project description:This study experimentally investigated the evaporation and wetting transition behavior of fakir droplets on five different microstructured surfaces. Diamond-like carbon was introduced as the substrate, and the influence of varying the width, height, and pitch of the micropillars was assessed. The experimental results showed that the interfacial properties of the surfaces change the evaporation behavior and the starting point of the wetting transition. An important result of this study is the demonstration of a slippery superhydrophobic surface with low depinning force that suppresses the transition from the Cassie-Baxter state to the Wenzel state for microdroplets less than 0.37 mm in diameter, without employing large pillar height or multiscale roughness. By selecting an appropriate pillar pitch and employing tapered micropillars with small pillar widths, the solid-liquid contact at the three-phase contact line was reduced and low depinning forces were obtained. The underlying mechanism by which slippery superhydrophobic surfaces suppress wetting transitions is also discussed. The accuracy of the theoretical models for predicting the critical transition parameters was assessed, and a numerical model was developed in the surface evolver to compute the penetration of the droplet bottom meniscus within the micropillars.

Hanging droplets from liquid surfaces.

Project description:Natural and man-made robotic systems use the interfacial tension between two fluids to support dense objects on liquid surfaces. Here, we show that coacervate-encased droplets of an aqueous polymer solution can be hung from the surface of a less dense aqueous polymer solution using surface tension. The forces acting on and the shapes of the hanging droplets can be controlled. Sacs with homogeneous and heterogeneous surfaces are hung from the surface and, by capillary forces, form well-ordered arrays. Locomotion and rotation can be achieved by embedding magnetic microparticles within the assemblies. Direct contact of the droplet with air enables in situ manipulation and compartmentalized cascading chemical reactions with selective transport. Applications including functional microreactors, motors, and biomimetic robots are evident.

Direct measurement of the lamellipodial protrusive force in a migrating cell.

Project description:There has been a great deal of interest in the mechanism of lamellipodial protrusion (Pollard, T., and G. Borisy. 2003. Cell. 112:453-465). However, one of this mechanism's endpoints, the force of protrusion, has never been directly measured. We place an atomic force microscopy cantilever in the path of a migrating keratocyte. The deflection of the cantilever, which occurs over a period of approximately 10 s, provides a direct measure of the force exerted by the lamellipodial leading edge. Stall forces are consistent with approximately 100 polymerizing actin filaments per micrometer of the leading edge, each working as an elastic Brownian ratchet and generating a force of several piconewtons. However, the force-velocity curves obtained from this measurement, in which velocity drops sharply under very small loads, is not sensitive to low loading forces, and finally stalls rapidly at large loads, are not consistent with current theoretical models for the actin polymerization force. Rather, the curves indicate that the protrusive force generation is a complex multiphase process involving actin and adhesion dynamics.

Structural ensemble and microscopic elasticity of freely diffusing DNA by direct measurement of fluctuations.

Project description:Precisely measuring the ensemble of conformers that a macromolecule populates in solution is highly challenging. Thus, it has been difficult to confirm or falsify the predictions of nanometer-scale dynamical modeling. Here, we apply an X-ray interferometry technique to probe the solution structure and fluctuations of B-form DNA on a length scale comparable to a protein-binding site. We determine an extensive set of intrahelix distance distributions between pairs of probes placed at distinct points on the surface of the DNA duplex. The distributions of measured distances reveal the nature and extent of the thermally driven mechanical deformations of the helix. We describe these deformations in terms of elastic constants, as is common for DNA and other polymers. The average solution structure and microscopic elasticity measured by X-ray interferometry are in striking agreement with values derived from DNA-protein crystal structures and measured by force spectroscopy, with one exception. The observed microscopic torsional rigidity of DNA is much lower than is measured by single-molecule twisting experiments, suggesting that torsional rigidity increases when DNA is stretched. Looking forward, molecular-level interferometry can provide a general tool for characterizing solution-phase structural ensembles.

Direct force measurement and loading on developing tissues in intact avian embryos.

Project description:Developmental morphogenesis is driven by tissue stresses acting on tissue rheology. Direct measurements of forces in small tissues (100 µm-1 mm) in situ, such as in early embryos, require high spatial precision and minimal invasiveness. Here, we introduce a control-based approach, tissue force microscopy (TiFM), that integrates a mechanical cantilever probe and live imaging with closed-loop feedback control of mechanical loading in early chicken embryos. By testing previously qualitatively characterized force-producing tissues in the elongating body axis, we show that TiFM quantitatively captures stress dynamics with high sensitivity. TiFM also provides the means to apply stable, minimally invasive and physiologically relevant loads to drive tissue deformation and to follow the resulting morphogenetic progression associated with large-scale cell movements. Together, TiFM allows us to control tissue force measurement and manipulation in small developing embryos, and promises to contribute to the quantitative understanding of complex multi-tissue mechanics during development.

Direct Measurement of Intermolecular Mechanical Force for Nonspecific Interactions between Small Molecules.

Project description:Mechanical force can evaluate intramolecular interactions in macromolecules. Because of the rapid motion of small molecules, it is extremely challenging to measure mechanical forces of nonspecific intermolecular interactions. Here, we used optical tweezers to directly examine the intermolecular mechanical force (IMMF) of nonspecific interactions between two cholesterols. We found that IMMFs of dimeric cholesterol complexes were dependent on the orientation of the interaction. The surprisingly high IMMF in cholesterol dimers (∼30 pN) is comparable to the mechanical stability of DNA secondary structures. Using Hess-like cycles, we quantified that changes in free energy of solubilizing cholesterol (ΔGsolubility) by β-cyclodextrin (βCD) and methylated βCD (Me-βCD) were as low as -16 and -27 kcal/mol, respectively. Compared to the ΔGsolubility of cholesterols in water (5.1 kcal/mol), these values indicated that cyclodextrins can easily solubilize cholesterols. Our results demonstrated that the IMMF can serve as a generic and multipurpose variable to dissect nonspecific intermolecular interactions among small molecules into orientational components.

Direct measurement of cortical force generation and polarization in a living parasite.

Project description:Apicomplexa is a large phylum of intracellular parasites that are notable for the diseases they cause, including toxoplasmosis, malaria, and cryptosporidiosis. A conserved motile system is critical to their life cycles and drives directional gliding motility between cells, as well as invasion of and egress from host cells. However, our understanding of this system is limited by a lack of measurements of the forces driving parasite motion. We used a laser trap to measure the function of the motility apparatus of living Toxoplasma gondii by adhering a microsphere to the surface of an immobilized parasite. Motion of the microsphere reflected underlying forces exerted by the motile apparatus. We found that force generated at the parasite surface begins with no preferential directionality but becomes directed toward the rear of the cell after a period of time. The transition from nondirectional to directional force generation occurs on spatial intervals consistent with the lateral periodicity of structures associated with the membrane pellicle and is influenced by the kinetics of actin filament polymerization and cytoplasmic calcium. A lysine methyltransferase regulates both the magnitude and polarization of the force. Our work provides a novel means to dissect the motile mechanisms of these pathogens.

Persisting water droplets on water surfaces.

Project description:Droplets of various liquids may float on the respective surfaces for extended periods of time prior to coalescence. We explored the features of delayed coalescence in highly purified water. Droplets several millimeters in diameter were released from a nozzle onto a water surface. Results showed that droplets had float times up to hundreds of milliseconds. When the droplets did coalesce, they did so in stepwise fashion, with periods of quiescence interspersed between periods of coalescence. Up to six steps were noted before the droplet finally vanished. Droplets were released in a series, which allowed the detection of unexpected abrupt float-time changes throughout the duration of the series. Factors such as electrostatic charge, droplet size, and sideways motion had considerable effect on droplet lifetime, as did reduction of pressure, which also diminished the number of steps needed for coalescence. On the basis of present observations and recent reports, a possible mechanism for noncoalescence is considered.
