Molecular Mechanisms of Cancer Prevention by Gooseberry (Phyllanthus emblica).

ABSTRACT: Indian gooseberry (Emblica officinalis Gaertn or Phyllanthus emblica Linn; family Phyllanthaceae) has a recognized history in Indian traditional medicine (Ayurveda). Various therapeutic properties have been attributed to gooseberry as a dietary supplement. Many parts of the plant (fruits, seed, leaves, root, bark, and flowers) possess various activities and are used to treat a range of diseases. This review focuses on the evidence for the cancer-preventive properties of gooseberry, its extracts, and its principal phytochemicals based on studies In Vitro and In Vivo. Most importantly, in multiple rodent models of cancer, treatment with P. emblica was found to prevent tumor incidence, number, and volume at various organ sites. The mechanism(s) implicated in gooseberry-mediated cancer inhibition are diverse and include antioxidants, Phase I and II enzyme modifications, anti-inflammatory action, regulation of the cell cycle, and modulation of oncogenic signaling genes. Studies in humans also indicate that P. emblica can offer various health benefits and synergize with other treatments. This review provides detailed information on the potential use of gooseberry extract as an anticarcinogenic in humans, illuminates the therapeutic applications, and discusses clinical trials.

Project description:Phyllanthus emblica (PE), a traditional multiethnic herbal medicine, is commonly applied to treat liver diseases. Our previous study demonstrated that aqueous extract of PE (AEPE) could alleviate carbon tetrachloride (CCl4)-induced liver fibrosis in vivo, but the underlying molecular mechanisms are still unclear. The present study was undertaken to clarify the multitarget mechanisms of PE in treating liver fibrosis by proteomics clues. A CCl4-induced liver fibrosis rat model was established. The anti-liver fibrosis effects of chemical fractions from AEPE were evaluated by serum biochemical indicators and pathological staining. Additionally, tandem mass tag (TMT) - based quantitative proteomics technology was used to detect the hepatic differentially expressed proteins (DEPs). The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment, gene ontology (GO) enrichment and protein-protein interaction (PPI) network were used to perform bioinformatics analysis of DEPs. Western blot analysis was used to verify the key potential targets regulated by the effective fraction of AEPE. The low-molecular-weight fraction of AEPE (LWPE) was determined to be the optimal anti-liver fibrosis active fraction, that could significantly improve ALT, AST, HA, Col IV, PCIII, LN, Hyp levels and reduce the pathological fibrotic lesion of liver tissue in model rats. A total of 195 DEPs were screened after LWPE intervention. GO analysis showed that the DEPs were related mostly to extracellular matrix organization, actin binding, and extracellular exosomes. KEGG pathway analysis showed that DEPs are mainly related to ECM-receptor interactions, focal adhesion and PI3K-Akt signaling pathway. Combined with the GO, KEGG and Western blot results, COL1A2, ITGAV, TLR2, ACE, and PDGFRB may be potential targets for PE treatment of liver fibrosis. In conclusion, LWPE exerts therapeutic effects through multiple pathways and multiple targets regulation in the treatment of liver fibrosis. This study may provide proteomics clues for the continuation of research on liver fibrosis treatment with PE.

Project description:Phyllanthus emblica L. is a well-known medicinal and edible plant species. Various medicinal compounds in the fruit make it an important medicinal and promising economic material. The plant is widely distributed in Southwestern and Southern China. However, due to massive deforestation and land reclamation as well as deterioration of its natural habitat in recent years, the wild resources of this species have been sharply reduced, and it is rare to see large-scale wild P. emblica forests so far. In order to effectively protect and rationally utilize this species, we investigated the genetic diversity, genetic structure, and population dynamics of 260 individuals from 10 populations of P. emblica sampled from the dry climate area in Yunnan and wet climate area in Guangxi using 20 polymorphic EST-SSR markers. We found high genetic diversity at the species level (He = 0.796) and within populations (He = 0.792), but low genetic differentiation among populations (F ST = 0.084). In addition, most genetic variation existed within populations (92.44%) compared with variation among the populations (7.56%). Meanwhile, the NJ tree, STRUCTURE, and hierarchical analysis suggested that the sampled individuals were clustered into two distinct genetic groups. In contrast, the genetic diversity of the dry climate group (He = 0.786, Na = 11.790, I = 1.962) was higher than that of the wet climate group (He = 0.673, Na = 9.060, I = 1.555), which might be attributed to the combined effects of altitude, precipitation, and geographic distance. Interestingly, only altitude and precipitation had significant pure effects on the genetic diversity, and the former was slightly stronger. In addition, DIYABC analysis suggested the effective population size of P. emblica might have contracted in the beginning of the Last Glacial Maximum. These genetic features provided vital information for the conservation and sustainable development of genetic resources of P. emblica, and they also provided new insights and guidelines for ecological restoration and economic development in dry-hot valleys of Yunnan and karst areas in Guangxi.

Project description:This study explores the mechanism of Phyllanthus emblica in treating epilepsy (EP) through network pharmacology and molecular docking. The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform identified the chemical composition of P emblica, Swiss ADME screened active components, and Swiss Target Prediction predicted targets. EP-related targets were identified using Gene Cards, OMIM, Drug Bank, TTD, and DisGeNET, and Venny 2.1.0 was used to find intersecting targets. Protein-protein interaction network analysis was conducted with STRING and Cytoscape. Chem 3D and Pymol were used for structural optimization and molecular docking was performed with AutoDock Tools 1.5.7 and Vina. Fifty-three active components and 126 intersecting targets were identified. Gene Ontology analysis revealed 3416 biological processes, 287 cellular components, and 457 molecular functions. Kyoto Encyclopedia of Genes and Genomes pathways showed neuroactive ligand-receptor interactions, nitrogen metabolism, and serotonergic synapses as key pathways. Molecular docking indicated strong binding energies between P emblica core components and targets, especially 2-ethylhexyl ester with MAPK3, luteolin with SRC, and kaempferol with MAPK1. This study explores the therapeutic potential of P emblica in treating EP through network pharmacology and molecular docking. A total of 53 active components were identified, with key compounds like 2-ethylhexyl ester, phyllanthin, luteolin, and kaempferol targeting critical proteins such as SRC, AKT1, APP, MAPK3, and MAPK1. These targets are involved in pathways related to synaptic transmission, oxidative stress, and inflammation, indicating potential neuroprotective and anti-inflammatory effects. Gene Ontology analysis highlighted the regulation of synaptic activity, while Kyoto Encyclopedia of Genes and Genomes pathway analysis emphasized pathways like neuroactive ligand-receptor interactions and serotonergic synapses. Molecular docking demonstrated strong binding affinities between active components and core targets, supporting the effectiveness of P emblica in modulating neuronal excitability and reducing neuroinflammation. These findings provide a theoretical basis for its clinical application in EP management.
