The Representation of Time Windows in Primate Auditory Cortex.

ABSTRACT: Whether human and nonhuman primates process the temporal dimension of sound similarly remains an open question. We examined the brain basis for the processing of acoustic time windows in rhesus macaques using stimuli simulating the spectrotemporal complexity of vocalizations. We conducted functional magnetic resonance imaging in awake macaques to identify the functional anatomy of response patterns to different time windows. We then contrasted it against the responses to identical stimuli used previously in humans. Despite a similar overall pattern, ranging from the processing of shorter time windows in core areas to longer time windows in lateral belt and parabelt areas, monkeys exhibited lower sensitivity to longer time windows than humans. This difference in neuronal sensitivity might be explained by a specialization of the human brain for processing longer time windows in speech.

The neuronal representation of pitch in primate auditory cortex.

Project description:Pitch perception is critical for identifying and segregating auditory objects, especially in the context of music and speech. The perception of pitch is not unique to humans and has been experimentally demonstrated in several animal species. Pitch is the subjective attribute of a sound's fundamental frequency (f(0)) that is determined by both the temporal regularity and average repetition rate of its acoustic waveform. Spectrally dissimilar sounds can have the same pitch if they share a common f(0). Even when the acoustic energy at f(0) is removed ('missing fundamental') the same pitch is still perceived. Despite its importance for hearing, how pitch is represented in the cerebral cortex is unknown. Here we show the existence of neurons in the auditory cortex of marmoset monkeys that respond to both pure tones and missing fundamental harmonic complex sounds with the same f(0), providing a neural correlate for pitch constancy. These pitch-selective neurons are located in a restricted low-frequency cortical region near the anterolateral border of the primary auditory cortex, and is consistent with the location of a pitch-selective area identified in recent imaging studies in humans.

The topography of frequency and time representation in primate auditory cortices.

Project description:Natural sounds can be characterised by their spectral content and temporal modulation, but how the brain is organized to analyse these two critical sound dimensions remains uncertain. Using functional magnetic resonance imaging, we demonstrate a topographical representation of amplitude modulation rate in the auditory cortex of awake macaques. The representation of this temporal dimension is organized in approximately concentric bands of equal rates across the superior temporal plane in both hemispheres, progressing from high rates in the posterior core to low rates in the anterior core and lateral belt cortex. In A1 the resulting gradient of modulation rate runs approximately perpendicular to the axis of the tonotopic gradient, suggesting an orthogonal organisation of spectral and temporal sound dimensions. In auditory belt areas this relationship is more complex. The data suggest a continuous representation of modulation rate across several physiological areas, in contradistinction to a separate representation of frequency within each area.

Task-specific invariant representation in auditory cortex.

Project description:Categorical sensory representations are critical for many behaviors, including speech perception. In the auditory system, categorical information is thought to arise hierarchically, becoming increasingly prominent in higher-order cortical regions. The neural mechanisms that support this robust and flexible computation remain poorly understood. Here, we studied sound representations in the ferret primary and non-primary auditory cortex while animals engaged in a challenging sound discrimination task. Population-level decoding of simultaneously recorded single neurons revealed that task engagement caused categorical sound representations to emerge in non-primary auditory cortex. In primary auditory cortex, task engagement caused a general enhancement of sound decoding that was not specific to task-relevant categories. These findings are consistent with mixed selectivity models of neural disentanglement, in which early sensory regions build an overcomplete representation of the world and allow neurons in downstream brain regions to flexibly and selectively read out behaviorally relevant, categorical information.

Causal contribution of primate auditory cortex to auditory perceptual decision-making.

Project description:Auditory perceptual decisions are thought to be mediated by the ventral auditory pathway. However, the specific and causal contributions of different brain regions in this pathway, including the middle-lateral (ML) and anterolateral (AL) belt regions of the auditory cortex, to auditory decisions have not been fully identified. To identify these contributions, we recorded from and microstimulated ML and AL sites while monkeys decided whether an auditory stimulus contained more low-frequency or high-frequency tone bursts. Both ML and AL neural activity was modulated by the frequency content of the stimulus. But, only the responses of the most stimulus-sensitive AL neurons were systematically modulated by the monkeys' choices. Consistent with this observation, microstimulation of AL, but not ML, systematically biased the monkeys' behavior toward the choice associated with the preferred frequency of the stimulated site. Together, these findings suggest that AL directly and causally contributes sensory evidence to form this auditory decision.

Sparse representation of sounds in the unanesthetized auditory cortex.

Project description:How do neuronal populations in the auditory cortex represent acoustic stimuli? Although sound-evoked neural responses in the anesthetized auditory cortex are mainly transient, recent experiments in the unanesthetized preparation have emphasized subpopulations with other response properties. To quantify the relative contributions of these different subpopulations in the awake preparation, we have estimated the representation of sounds across the neuronal population using a representative ensemble of stimuli. We used cell-attached recording with a glass electrode, a method for which single-unit isolation does not depend on neuronal activity, to quantify the fraction of neurons engaged by acoustic stimuli (tones, frequency modulated sweeps, white-noise bursts, and natural stimuli) in the primary auditory cortex of awake head-fixed rats. We find that the population response is sparse, with stimuli typically eliciting high firing rates (>20 spikes/second) in less than 5% of neurons at any instant. Some neurons had very low spontaneous firing rates (<0.01 spikes/second). At the other extreme, some neurons had driven rates in excess of 50 spikes/second. Interestingly, the overall population response was well described by a lognormal distribution, rather than the exponential distribution that is often reported. Our results represent, to our knowledge, the first quantitative evidence for sparse representations of sounds in the unanesthetized auditory cortex. Our results are compatible with a model in which most neurons are silent much of the time, and in which representations are composed of small dynamic subsets of highly active neurons.

Neuronal figure-ground responses in primate primary auditory cortex.

Project description:Figure-ground segregation, the brain's ability to group related features into stable perceptual entities, is crucial for auditory perception in noisy environments. The neuronal mechanisms for this process are poorly understood in the auditory system. Here, we report figure-ground modulation of multi-unit activity (MUA) in the primary and non-primary auditory cortex of rhesus macaques. Across both regions, MUA increases upon presentation of auditory figures, which consist of coherent chord sequences. We show increased activity even in the absence of any perceptual decision, suggesting that neural mechanisms for perceptual grouping are, to some extent, independent of behavioral demands. Furthermore, we demonstrate differences in figure encoding between more anterior and more posterior regions; perceptual saliency is represented in anterior cortical fields only. Our results suggest an encoding of auditory figures from the earliest cortical stages by a rate code.

Representation of multiple, independent categories in the primate prefrontal cortex.

Project description:Neural correlates of visual categories have been previously identified in the prefrontal cortex (PFC). However, whether individual neurons can represent multiple categories is unknown. Varying degrees of generalization versus specialization of neurons in the PFC have been theorized. We recorded from lateral PFC neural activity while monkeys switched between two different and independent categorical distinctions (Cats versus Dogs, Sports Cars versus Sedans). We found that many PFC neurons reflected both categorical distinctions. In fact, these multitasking neurons had the strongest category effects. This stands in contrast to our lab's recent report that monkeys switching between competing categorical distinctions (applied to the same stimulus set) showed independent representations. We suggest that cognitive demands determine whether PFC neurons function as category "multitaskers."

The synaptic representation of sound source location in auditory cortex.

Project description:A key function of the auditory system is to provide reliable information about the location of sound sources. Here, we describe how sound location is represented by synaptic input arriving onto pyramidal cells within auditory cortex by combining free-field acoustic stimulation in the frontal azimuthal plane with in vivo whole-cell recordings. We found that subthreshold activity was panoramic in that EPSPs could be evoked from all locations in all cells. Regardless of the sound location that evoked the largest EPSP, we observed a slowing in the EPSP slope along the contralateral-ipsilateral plane that was reflected in a temporal sequence of peak EPSP times. Contralateral sounds evoked EPSPs with earlier peak times and consequently generated action potential firing with shorter latencies than ipsilateral sounds. Thus, whereas spiking probability reflected the region of space evoking the largest EPSP, across the population, synaptic inputs enforced a gradient of spike latency and precision along the horizontal axis. Therefore, within auditory cortex and regardless of preferred location, the time window of synaptic integration reflects sound source location and ensures that spatial acoustic information is represented by relative timings of pyramidal cell output.

Vowel and formant representation in the human auditory speech cortex.

Project description:Vowels, a fundamental component of human speech across all languages, are cued acoustically by formants, resonance frequencies of the vocal tract shape during speaking. An outstanding question in neurolinguistics is how formants are processed neurally during speech perception. To address this, we collected high-density intracranial recordings from the human speech cortex on the superior temporal gyrus (STG) while participants listened to continuous speech. We found that two-dimensional receptive fields based on the first two formants provided the best characterization of vowel sound representation. Neural activity at single sites was highly selective for zones in this formant space. Furthermore, formant tuning is adjusted dynamically for speaker-specific spectral context. However, the entire population of formant-encoding sites was required to accurately decode single vowels. Overall, our results reveal that complex acoustic tuning in the two-dimensional formant space underlies local vowel representations in STG. As a population code, this gives rise to phonological vowel perception.

Enhanced representation of spectral contrasts in the primary auditory cortex.

Project description:The role of early auditory processing may be to extract some elementary features from an acoustic mixture in order to organize the auditory scene. To accomplish this task, the central auditory system may rely on the fact that sensory objects are often composed of spectral edges, i.e., regions where the stimulus energy changes abruptly over frequency. The processing of acoustic stimuli may benefit from a mechanism enhancing the internal representation of spectral edges. While the visual system is thought to rely heavily on this mechanism (enhancing spatial edges), it is still unclear whether a related process plays a significant role in audition. We investigated the cortical representation of spectral edges, using acoustic stimuli composed of multi-tone pips whose time-averaged spectral envelope contained suppressed or enhanced regions. Importantly, the stimuli were designed such that neural responses properties could be assessed as a function of stimulus frequency during stimulus presentation. Our results suggest that the representation of acoustic spectral edges is enhanced in the auditory cortex, and that this enhancement is sensitive to the characteristics of the spectral contrast profile, such as depth, sharpness and width. Spectral edges are maximally enhanced for sharp contrast and large depth. Cortical activity was also suppressed at frequencies within the suppressed region. To note, the suppression of firing was larger at frequencies nearby the lower edge of the suppressed region than at the upper edge. Overall, the present study gives critical insights into the processing of spectral contrasts in the auditory system.
