Project description:A water-soluble perylene diimide, aspartic acid-functionalized perylene diimide (APDI), has shown significant sequential "turn-off" and "turn-on" responses toward Cu2+ and inorganic pyrophosphate (PPi), respectively. APDI was found to show selectivity toward Cu2+ and inorganic PPi over adenosine monophosphate, adenosine diphosphate, and adenosine triphosphate. The detection has been studied by absorption and emission spectroscopy techniques. Incorporation of Cu2+ into the solution of APDI results in a distinct quenching of the fluorescence intensity, while there was no spectral change in the presence of other metal ions. The formed APDI-Cu2+ ensemble can turn on its fluorescence signal when PPi is present. The detection of PPi could be traced by looking at the change in color of the solution under the naked eye. No interference was observed from other anions, making the APDI-Cu2+aggregate a highly selective biosensor for PPi.

Project description:Biothiols such as cysteine (Cys) and homocysteine (Hcy) are essential biomolecules participating in molecular and physiological processes in an organism. However, their selective detection remains challenging. In this study, ethyl 2-(3-formyl-4-hydroxyphenyl)-4-methylthiazole-5-carboxylate (NL) was synthesized as a ratiometric fluorescent probe for the rapid and selective detection of Cys and Hcy over glutathione (GSH) and other amino acids. The fluorescence intensity of the probe in the presence of Cys/Hcy increased about 3-fold at a concentration of 20 equiv. of the probe, compared with that in the absence of these chemicals in aqueous media. The limits of detection of the fluorescent assay were 0.911 μM and 0.828 μM of Cys and Hcy, respectively. ¹H-NMR and MS analyses indicated that an excited-state intramolecular proton transfer is the mechanism of fluorescence sensing. This ratiometric probe is structurally simple and highly selective. The results suggest that it has useful applications in analytical chemistry and diagnostics.

Project description:A novel fluorescent quinolizinium-based turn-off probe has been developed for selective detection of cysteine. The probe showed high selectivity and sensitivity towards cysteine over other amino acids including the similarly structured homocysteine and glutathione with a detection limit of 0.18 μM (S/N = 3). It was successfully applied to cysteine detection in living cells with low cytotoxicity and quantitative analysis of spiked mouse serum samples with moderate to good recovery (96-109%).

Project description:An anthracene based fluorescent probe, integrated with thiophene moiety, exhibited selective and sensitive detection of chromium (III) ions over other metal ions. Its synthesis was achieved by simple mixing of two commercially available compounds, 2-aminoanthracene, and 2-thiophenecarboxaldehyde, in onestep without the needed complex purification process. The probe molecule ( ANT-Th ) offered exceptional features such as "turn-on" fluorescence response, low detection limit (0.4 μM), and fast response time (<1 min) via C=N bond hydrolysis. Also, a simple test paper system was developed for the rapid detection of chromium (III) ions with the naked eye.

Project description:A benzothiazole-based near-infrared (NIR) ratiometric fluorescent probe (HBT-Cys) was developed for discriminating cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH). The probe was designed by masking phenol group in the conjugated benzothiazole derivative with methacrylate group that could be selectively removed by Cys, and therefore an intramolecular charge transfer (ICT) fluorescence was switched on in the NIR region. In the absence of Cys, the probe exhibited a strong blue fluorescence emission at 431 nm, whereas a NIR fluorescence emission at 710 nm was significantly enhanced accompanied by a decrease of emission at 431 nm in the presence of Cys, allowing a ratiometric fluorescence detection of Cys. The fluorescence intensity ratio (I710nm/I431nm) showed a good linear relationship with Cys concentration of 1-40 μM with the detection limit of 0.5 μM. The sensing mechanism was explored based on MS experimental analysis and DFT theoretical calculation. Moreover, the fluorescent probe was successfully used for fluorescence bioimaging of Cys in living cells.

Project description:Two of the most critical factors for the survival of glioblastoma (GBM) patients are precision diagnosis and the tracking of treatment progress. At the moment, various sophisticated and specific diagnostic procedures are being used, but there are relatively few simple diagnosis methods. This work introduces a sensing probe based on a turn-on type fluorescence response that can measure the cysteine (Cys) level, which is recognized as a new biomarker of GBM, in human-derived cells and within on-site human clinical biopsy samples. The Cys-initiated chemical reactions of the probe cause a significant fluorescence response with high selectivity, high sensitivity, a fast response time, and a two-photon excitable excitation pathway, which allows the imaging of GBM in both mouse models and human tissue samples. The probe can distinguish the GBM cells and disease sites in clinical samples from individual patients. Besides, the probe has no short or long-term toxicity and immune response. The present findings hold promise for application of the probe to a relatively simple and straightforward following of GBM at clinical sites.

Project description:There is increasing interest in the development of noble metal separation/recovery processes, especially for applications to "urban mining". Common separation/recovery processes for noble metals use a solvent (liquid-liquid) extraction technique in hydrometallurgy. However, these processes are time-consuming and not environmentally friendly, because they use organic solvents for sequential metal ion extractions. Electrowinning is an alternative approach for selective metal precipitation that involves controlling the redox potentials of electrodes but requires specialized equipment and generates hydrogen as a byproduct at the cathode surface under dilute conditions. In the present study, we investigated selective gold recovery from a homogenous aqueous solution containing a mixture of dilute HAuCl4 and H2PtCl6 (5.0 × 10-5 M each) and aromatic amino acid-containing peptides (2.0 × 10-4 M each). Gold selectivity was determined by analyzing the compositions of the solids and supernatants obtained from the reaction mixtures. A much higher gold selectivity (gold/platinum (Au/Pt) atomic ratio = 7.5) was obtained using an anthracene-containing peptide compared to peptides containing one or two naphthalene ring(s). Our proposed approach is applicable to the sequential separation of several noble metal ions, such as Au, palladium (Pd), Pt, iridium (Ir) and rhodium (Rh), and simply requires developing aromatics suitable for each noble metal of interest.

Project description:We reported here a turn-on fluorescent probe (1) for the detection of cysteine (Cys) by incorporating the recognition unit of 2,4-dinitrobenzenesulfonyl ester (DNBS) to a coumarin derivative. The structure of the obtained probe was confirmed by NMR and HRMS techniques. The probe shows a remarkable fluorescence off-on response (∼52-fold) by the reaction with Cys in 100% aqueous buffer. The sensing mechanism was verified by the HPLC test. Probe 1 also displays high selectivity towards Cys. The detection limit was calculated to be 23 nM. Moreover, cellular experiments demonstrated that the probe is highly biocompatible and can be used for monitoring intracellular Cys.

Project description:Cysteine (Cys), homocysteine (Hcy) and glutathione (GSH) are essential biothiols for cellular growth, metabolism, and maintenance of a biological system. Thus, the detection of biothiols is highly important for early diagnosis and evaluation of disease progression. In this article, a series of sulfonyl aza-BODIPYs was synthesized, characterized, and examined by 1H-NMR, 13C-NMR, crystallization, photophysical properties and DFT calculation. Among these structures, a fluorescent probe, BDP-1, exhibited selective detection of Cys among various biothiols via nucleophilic aromatic substitution and typical size of Cys molecules. BDP-1 showed color change and near-infrared (NIR) fluorescence enhancement after reaction with Cys to generate BDP-OH, confirmed by HRMS. The red shift of absorption wavelength showed a similar tendency resulting in time-dependent density functional theory (TD-DFT). Furthermore, the calculated detection limit of BDP-1 toward Cys was 5.23 μM. This probe facilitates the colorimetric and fluorescent detection of Cys over other biothiols.

Project description:A coumarin-based dual responsive fluorescent probe with a simple structure was developed for the detection of Cys and HSO3 -. Under simulated physiological conditions, Cou-F displayed an on-off fluorescence response to Cys at 521 nm and an off-on fluorescence response to HSO3 - at 500 nm. Furthermore, Cou-F had the advantages of high sensitivity, strong specificity and rapid response. The detection limits of Cou-F toward Cys and HSO3 - were 0.54 μM and 0.65 μM, respectively. Cou-F enabled high selective responses to Cys and HSO3 - over other biologically related species. The response times of Cou-F toward Cys and HSO3 - were 80 s and 100 s. The fluorescence imaging of Cys and HSO3 - was achieved in living RAW246.7 cells.
