Exploring the Potential Enhancing Effects of Trans-Zeatin and Silymarin on the Productivity and Antioxidant Defense Capacity of Cadmium-Stressed Wheat.

ABSTRACT: Pot trials were performed to explore the impacts of seed priming (SPr) plus leaf treatment (LTr) with trans-zeatin-type cytokinin (tZck; 0.05 mM) and silymarin (Sim; 0.5 mM) on growth, yield, physio-biochemical responses, and antioxidant defense systems in Cd-stressed wheat. tZck + Sim applied as SPr + LTr was more effective than individual treatments, and the impacts were more pronounced under stress conditions. Cd stress (0.6 mM) severely declined growth and yield traits, and photosynthesis efficiency (pigment contents, instantaneous carboxylation efficiency, and photochemical activity) compared to the control. These negative impacts coincided with increased levels of Cd²⁺, O₂^•- (superoxide), H₂O₂ (hydrogen peroxide), MDA (malondialdehyde), and EL (electrolyte leakage). Non-enzymatic and enzymatic antioxidant activities, and tZck and Sim contents were also increased. However, tZck + Sim increased photosynthesis efficiency, and further boosted antioxidant activities, and contents of tZck and Sim, while minimizing Cd²⁺ levels in roots, leaves, and grains. The levels of O₂^•-, H₂O₂, MDA, and EL were also minimized, reflecting positively on growth and productivity. tZck + Sim applied as SPr + LTr was highly effective in promoting antioxidants and photosynthesis machineries, minimizing oxidative stress biomarkers and Cd²⁺ levels, boosting tolerance to Cd stress, and improving wheat productivity under Cd stress.

Project description:The improvement of crop productivity under abiotic stress is one of the biggest challenges faced by the agricultural scientific community. Despite extensive research, the research-to-commercial transfer rate of abiotic stress-resistant crops remains very low. This is mainly due to the complexity of genotype × environment interactions and in particular, the ability to quantify the dynamic plant physiological response profile to a dynamic environment. Most existing phenotyping facilities collect information using robotics and automated image acquisition and analysis. However, their ability to directly measure the physiological properties of the whole plant is limited. We demonstrate a high-throughput functional phenotyping system (HFPS) that enables comparing plants' dynamic responses to different ambient conditions in dynamic environments due to its direct and simultaneous measurement of yield-related physiological traits of plants under several treatments. The system is designed as one-to-one (1:1) plant-[sensors+controller] units, i.e., each individual plant has its own personalized sensor, controller and irrigation valves that enable (i) monitoring water-relation kinetics of each plant-environment response throughout the plant's life cycle with high spatiotemporal resolution, (ii) a truly randomized experimental design due to multiple independent treatment scenarios for every plant, and (iii) reduction of artificial ambient perturbations due to the immobility of the plants or other objects. In addition, we propose two new resilience-quantifying-related traits that can also be phenotyped using the HFPS: transpiration recovery rate and night water reabsorption. We use the HFPS to screen the effects of two commercial biostimulants (a seaweed extract -ICL-SW, and a metabolite formula - ICL-NewFo1) on Capsicum annuum under different irrigation regimes. Biostimulants are considered an alternative approach to improving crop productivity. However, their complex mode of action necessitates cost-effective pre-field phenotyping. The combination of two types of treatment (biostimulants and drought) enabled us to evaluate the precision and resolution of the system in investigating the effect of biostimulants on drought tolerance. We analyze and discuss plant behavior at different stages, and assess the penalty and trade-off between productivity and resilience. In this test case, we suggest a protocol for the screening of biostimulants' physiological mechanisms of action.

Project description:Plant productivity is adversely affected by exposure to abiotic stresses including salinity. Leaf tissue undergoes accelerated senescence with exposure to salt stress conditions, culminating in lowered efficiency of photosynthetic machinery. The N6-adenine derivative phytohormone Cytokinin (CK) is well known to delay natural leaf senescence by maintaining functioning photosynthesis processes. How various forms of CK function to control this activity and alter physiological and molecular responses under salt-induced leaf senescence is still not known. Here the CKs, trans-Zeatin (tZ) and its N-glucosylated (tZNGs) forms; tZ7G and tZ9G, were tested for their ability to delay leaf senescence in Arabidopsis thaliana. A modified dark-induced leaf senescence CK bioassay was used to study the physiology of photosynthesis performance as well as transcriptome and proteome level changes in detached leaves, across different phases of salt-induced leaf senescence from 2h-72h. Treatments with different CK forms (tZ, tZ7G, and tZ9G) showed that tZNGs can improve PSII efficiency (Fv/Fm) and increase chlorophyll levels compared to salt only treatments, indicating that these CK forms function as active CK forms in delaying senescence. At the transcript level, tZ, tZ7G, and tZ9G revealed differentially expressed genes (DEGs) across both developmental and salt-treated senescence, with tZNGs predominantly altering DEG in the terminal timepoint of salt-treated leaf senescence. Additionally, we found that tZNGs uniquely regulate crucial biological processes (BPs) including CK signaling compared to tZ. Proteome analysis showed that tZNGs and tZ showed similarities in number of differentially abundant proteins (DAPs) trends across senescence timepoints although each of the three forms showing uniquely abundant proteins. GO term analysis for tZNG regulated genes and proteins revel the enrichment of senescence- and chloroplast-related BPs. This study highlights the role of tZNGs as active cytokinin forms towards delaying salt-treated leaf senescence in Arabidopsis at the physiology, transcriptome, and proteome level.

Project description:BackgroundHeat stress is a major restrictive factor that causes yield loss in rice. We previously reported the priming effect of abscisic acid (ABA) on rice for enhanced thermotolerance at the germination, seedling and heading stages. In the present study, we aimed to understand the priming effect and mechanism of ABA on grain filling capacity in rice under heat stress.ResultsRice plants were pretreated with distilled water, 50 μM ABA and 10 μM fluridone by leaf spraying at 8 d or 15 d after initial heading (AIH) stage and then were subjected to heat stress conditions of 38 °C day/30 °C night for 7 days, respectively. Exogenous ABA pretreatment significantly super-activated the ABA signaling pathway and improved the SOD, POD, CAT and APX enzyme activity levels, as well as upregulated the ROS-scavenging genes; and decreased the heat stress-induced ROS content (O2- and H2O2) by 15.0-25.5% in rice grain under heat stress. ABA pretreatment also increased starch synthetase activities in rice grain under heat stress. Furthermore, ABA pretreatment significantly improved yield component indices and grain yield by 14.4-16.5% under heat stress. ABA pretreatment improved the milling quality and the quality of appearance and decreased the incidence of chalky kernels and chalkiness in rice grain and improved the rice grain cooking quality by improving starch content and gel consistence and decreasing the amylose percentage under heat stress. The application of paraquat caused overaccumulation of ROS, decreased starch synthetase activities and ultimately decreased starch content and grain yield. Exogenous antioxidants decreased ROS overaccumulation and increased starch content and grain yield under heat stress.ConclusionTaken together, these results suggest that exogenous ABA has a potential priming effect for enhancing rice grain filling capacity under heat stress at grain filling stage mainly by inhibiting ROS overaccumulation and improving starch synthetase activities in rice grain.
