Theranostic Applications of an Ultra-Sensitive T₁ and T₂ Magnetic Resonance Contrast Agent Based on Cobalt Ferrite Spinel Nanoparticles.

ABSTRACT: Nano-dimensional materials have become a focus of multiple clinical applications due to their unique physicochemical properties. Magnetic nanoparticles represent an important class of nanomaterials that are widely studied for use as magnetic resonance (MR) contrast and drug delivery agents, especially as they can be detected and manipulated remotely. Using magnetic cobalt ferrite spinel (MCFS) nanoparticles, this study was aimed at developing a multifunctional drug delivery platform with MRI capability for use in cancer treatment. We found that MCFS nanoparticles demonstrated outstanding properties for contrast MRI (r₁ = 22.1 s^-1mM^-1 and r₂ = 499 s^-1mM^-1) that enabled high-resolution T₁- and T₂-weighted MRI-based signal detection. Furthermore, MCFS nanoparticles were used for the development of a multifunctional targeted drug delivery platform for cancer treatment that is concurrently empowered with the MR contrast properties. Their therapeutic effect in systemic chemotherapy and unique MRI double-contrast properties were confirmed in vivo using a breast cancer mouse tumor model. Our study thus provides an empirical basis for the development of a novel multimodal composite drug delivery system for anticancer therapy combined with noninvasive MRI capability.

Project description:Spinel ferrite nanocubes (NCs), consisting of pure iron oxide or mixed ferrites, are largely acknowledged for their outstanding performance in magnetic hyperthermia treatment (MHT) or magnetic resonance imaging (MRI) applications while their magnetic particle imaging (MPI) properties, particularly for this peculiar shape different from the conventional spherical nanoparticles (NPs), are relatively less investigated. In this work, we report on a non-hydrolytic synthesis approach to prepare mixed transition metal ferrite NCs. A series of NCs of mixed zinc-cobalt-ferrite were prepared and their magnetic theranostic properties were compared to those of cobalt ferrite or zinc ferrite NCs of similar sizes. For each of the nanomaterials, the synthesis parameters were adjusted to obtain NCs in the size range from 8 up to 15 nm. The chemical and structural nature of the different NCs was correlated to their magnetic properties. In particular, to evaluate magnetic losses, we compared the data obtained from calorimetric measurements to the data measured by dynamic magnetic hysteresis obtained under alternating magnetic field (AMF) excitation. Cobalt-ferrite and zinc-cobalt ferrite NCs showed high specific adsorption rate (SAR) values in aqueous solutions but their heating ability was drastically suppressed once in viscous media even for NCs as small as 12 nm. On the other hand, non-stoichiometric zinc-ferrite NCs showed significant but lower SAR values than the other ferrites, but these zinc-ferrite NCs preserved almost unaltered their heating trend in viscous environments. Also, the presence of zinc in the crystal lattice of zinc-cobalt ferrite NCs showed increased contrast enhancement for MRI with the highest T2 relaxation time and in the MPI signal with the best point spread function and signal-to-noise ratio in comparison to the analogue cobalt-ferrite NC. Among the different compositions investigated, non-stoichiometric zinc-ferrite NCs can be considered the most promising material as a multifunctional theranostic platform for MHT, MPI and MRI regardless of the media viscosity in which they will be applied, while ensuring the best biocompatibility with respect to the cobalt ferrite NCs.

Project description:Copper ferrite nanoparticles for use as MRI contrast agents were synthesized using two different methods. A novel microwave-assisted (MW) solvothermal method was developed and compared to a conventional 12-hour synthesis (Reflux) as an eco-friendlier approach. This innovative synthesis method successfully produced nanoparticles with enhanced properties compared to traditional ferrite materials. The nanoparticles' morphological and magnetic properties were evaluated and tested in in-vivo MRI studies. The results revealed both similarities and differences between the amine-functionalized copper ferrite nanoparticles. FTIR, XRD, HRTEM, and VSM analyses indicated improved properties in the CuFe2O4-NH2 MW particles, while AFM confirmed successful polymer encapsulation of the nanoparticles. For the CuFe2O4-NH2 MW sample, 76.8 wt% copper ferrite and 23.2 wt% magnetite were detected, with crystallite sizes of 8 ± 2 nm and 13 ± 2 nm, respectively. In the CuFe2O4-NH2 Refl. sample, in addition to these two magnetic phases, larger copper particles (31.6 wt%) were also formed. DLS analysis demonstrated that the CuFe2O4-NH2 MW sample exhibited excellent colloidal stability, maintaining its size distribution in aqueous media for 3 hours without aggregation, unlike the CuFe2O4-NH2 Refl. sample, which showed slight aggregation. The CuFe2O4-NH2 MW sample displayed superparamagnetic behavior (Ms: 15 emu/g, Mr: 0 emu/g, Hc: 0 Oe), while the CuFe2O4-NH2 Refl. sample exhibited ferromagnetic characteristics (Ms: 40 emu/g, Mr: 1.35 emu/g, Hc: 30 Oe). Both samples produced comparable results during in vitro MRI measurements, showing similar T2* relaxation and signal characteristics. Further in vivo studies demonstrated that both samples induced significant hypointense changes. The study provides valuable insights into the synthesis, properties, and potential applications of these materials, emphasizing the importance of eco-friendly methods and the optimization of ferrite-based MRI contrast agents.
