Nutritional Value of Eggplant Cultivars and Association with Sequence Variation in Genes Coding for Major Phenolics.

ABSTRACT: Eggplant is a widely consumed vegetable, with significant nutritional value and high antioxidant content, mainly due to its phenolic constituents. Our goal was to determine the levels of carbohydrates, proteins, total phenolics, anthocyanins, flavonoids, chlorogenic acid, and the antioxidant capacity in thirteen eggplant cultivars cultivated in Greece and to identify sequence polymorphisms in key regulating genes of the phenylpropanoid pathway (C4H, HCT, HQT, C3H, F3H, ANS, MYB1), which might relate to the phytochemical content of those cultivars. The carbohydrates' content differs among and within cultivars, while the rest of the phytochemicals differ only among cultivars. The cultivars 'EMI' and 'Lagkada' scored higher than the rest in phenolics, anthocyanins, ascorbic acid, caffeoylquinic acid, and antioxidant capacity. Moreover, significant correlations were observed between various ingredients and the antioxidant capacity (FRAP and DPPH). Sequence analysis revealed several SNPs in C4H, HQT, F3H, ANS, and MYB1 among the cultivars studied. According to chi-square and logistic regression analyses, the missense mutation C4H4-108 correlates significantly with flavonoids, anthocyanins, and proteins; the synonymous mutation HQT-105 correlates with anthocyanins and ascorbic acid; the missense mutation HQT-438 correlates with flavonoids and chlorogenic acid, while the missense mutation ANS1-65 correlates with anthocyanins and sugars. These polymorphisms can be potentially utilized as molecular markers in eggplant breeding, while our data also contribute to the study of eggplant's secondary metabolism and antioxidant properties.

Project description:The imperative need to produce safe foodstuffs using environmentally sustainable practices has highlighted the incorporation of legumes in human and animal diets as an emerging nutritional staple. Since legumes comprise a family of plants known to display an extensive agricultural diversity with significant bioactivities, we report herein the exploitation outcome of the nutritional and bio-functional content of hay, derived from the post-harvest aerial parts of eight novel-improved Greek cultivars belonging to the following six taxa: Larthyrus sativus L., Medicago sativa L., Cicer arietinum L., Pisum sativum L., Vicia faba L., and Phaseolus vulgaris L. In specific, the phytochemical content of their hay extracts was screened against the presence of 37 fatty acids and 67 polyphenols using, respectively, GC-FID and LC-MS/MS instruments, while the determination of their total phenolic and tannin contents was achieved with spectrophotometric methods. In this respect, the presence of 26 different fatty acids was detected of which the acids linoleic, linolenic and palmitic were traced in all extracts in concentrations exceeding the 10 mg/g, while the oleic acid was determined as the major component of C. arietinum (77.58 mg/g), V. faba (36.11 mg/g), and L. sativus (12.89 mg/g) extracts. In addition, 30 different phenolics were traced as extracts' components with the most abundant the molecule of isoquercetin, which was present in six extracts and isoliquiritigenin, calycosin, and chlorogenic acid which were detected in five extracts. Finally, the antioxidant properties of the extracts were evaluated by performing both DPPH• and FRAP assays, which highlighted the V. faba extract as the most potent in both tests, followed by the extracts of P. sativum and P. vulgaris. Results herein are indicative of the significant advances achieved, for the improvement of investigated plant cultivars with respect to their utilization as nutritional crops, since these novel cultivars hays have been found to contain significant amounts of essential nutrients in the form of fatty acids along with bioactive ingredients in the form of polyphenols, while simultaneously they were determined as deprived of tannins that constitute essential anti-nutritional factors.

Project description:Basella alba is a frequently consumed leafy vegetable. However, research on its nutritional components is limited. This study aimed to explore the variation in the nutritional components and antioxidant capacity of different cultivars and organs of Basella alba. Here, we primarily chose classical spectrophotometry and high-performance liquid chromatography (HPLC) to characterize the variation in nutritional components and antioxidant capacity among different organs (inflorescences, green fruits, black fruits, leaves, and stems) of eight typical cultivars of Basella alba. The determination indices (and methods) included the total soluble sugar (anthrone colorimetry), total soluble protein (the Bradford method), total chlorophyll (the ethanol-extracting method), total carotenoids (the ethanol-extracting method), total ascorbic acid (the HPLC method), total proanthocyanidins (the p-dimethylaminocinnamaldehyde method), total flavonoids (AlCl3 colorimetry), total phenolics (the Folin method), and antioxidant capacity (the FRAP and ABTS methods). The results indicated that M5 and M6 exhibited advantages in their nutrient contents and antioxidant capacities. Additionally, the inflorescences demonstrated the highest total ascorbic acid and total phenolic contents, while the green and black fruits exhibited relatively high levels of total proanthocyanidins and antioxidant capacity. In a comparison between the green and black fruits, the green fruits showed higher levels of total chlorophyll (0.77-1.85 mg g-1 DW), total proanthocyanidins (0.62-2.34 mg g-1 DW), total phenolics (15.28-27.35 mg g-1 DW), and ABTS (43.39-59.16%), while the black fruits exhibited higher levels of total soluble protein (65.45-89.48 mg g-1 DW) and total soluble sugar (56.40-207.62 mg g-1 DW) in most cultivars. Chlorophyll, carotenoids, and flavonoids were predominantly found in the leaves of most cultivars, whereas the total soluble sugar contents were highest in the stems of most cultivars. Overall, our findings underscore the significant influence of the cultivars on the nutritional composition of Basella alba. Moreover, we observed notable variations in the nutrient contents among the different organs of the eight cultivars, and proanthocyanidins may contribute significantly to the antioxidant activity of the fruits. On the whole, this study provides a theoretical basis for the genetic breeding of Basella alba and dietary nutrition and serves as a reference for the comprehensive utilization of this vegetable.
