Label-Free Detection of DNA via Surface-Enhanced Raman Spectroscopy Using Au@Ag Nanoparticles.

ABSTRACT: DNA is a building block of life; surface-enhanced Raman spectroscopy (SERS) has been broadly applied in the detection of biomolecules but there are challenges in obtaining high-quality DNA SERS signals under non-destructive conditions. Here, we developed a novel label-free approach for DNA detection based on SERS, in which the Au@AgNPs core-shell structure was selected as the enhancement substrate, which not only solved the problem of the weak enhancement effect of gold nanoparticles but also overcame the disadvantage of the inhomogeneous shapes of silver nanoparticles, thereby improving the sensitivity and reproducibility of the SERS signals of DNA molecules. The method obtained SERS signals for four DNA bases (A, C, G, and T) without destroying the structure, then further detected and qualified different specific structures of DNA molecules. These results promote the application of SERS technology in the field of biomolecular detection.

Label-Free Surface Enhanced Raman Spectroscopy for Cancer Detection.

Project description:Blood is a vital reservoir housing numerous disease-related metabolites and cellular components. Thus, it is also of interest for cancer diagnosis. Surface-enhanced Raman spectroscopy (SERS) is widely used for molecular detection due to its very high sensitivity and multiplexing properties. Its real potential for cancer diagnosis is not yet clear. In this study, using silver nanoparticles (AgNPs) as substrates, a number of experimental parameters and scenarios were tested to disclose the potential for this technique for cancer diagnosis. The discrimination of serum samples from cancer patients, healthy individuals and patients with chronic diseases was successfully demonstrated with over 90% diagnostic accuracies. Moreover, the SERS spectra of the blood serum samples obtained from cancer patients before and after tumor removal were compared. It was found that the spectral pattern for serum from cancer patients evolved into the spectral pattern observed with serum from healthy individuals after the removal of tumors. The data strongly suggests that the technique has a tremendous potential for cancer detection and screening bringing the possibility of early detection onto the table.

Facile Histamine Detection by Surface-Enhanced Raman Scattering using SiO2@Au@Ag Alloy Nanoparticles.

Project description:Histamine intoxication associated with seafood consumption represents a global health problem. The consumption of high concentrations of histamine can cause illnesses ranging from light symptoms, such as a prickling sensation, to death. In this study, gold-silver alloy-embedded silica (SiO2@Au@Ag) nanoparticles were created to detect histamine using surface-enhanced Raman scattering (SERS). The optimal histamine SERS signal was measured following incubation with 125 μg/mL of SiO2@Au@Ag for 2 h, with a material-to-histamine solution volume ratio of 1:5 and a phosphate-buffered saline-Tween 20 (PBS-T) solvent at pH 7. The SERS intensity of the histamine increased proportionally with the increase in histamine concentration in the range 0.1-0.8 mM, with a limit of detection of 3.698 ppm. Our findings demonstrate the applicability of SERS using nanomaterials for histamine detection. In addition, this study demonstrates that nanoalloys could have a broad application in the future.

Ag/Au Nanoparticle-Loaded Paper-Based Versatile Surface-Enhanced Raman Spectroscopy Substrates for Multiple Explosives Detection.

Project description:We present a systematic study on the fabrication, characterization of versatile, and low-cost filter paper-based surface-enhanced Raman spectroscopy (SERS) substrates loaded with salt-induced aggregated Ag/Au nanoparticles (NPs). These were demonstrated as efficient SERS substrates for the detection of multiple explosive molecules such as picric acid (5 μM), 2,4-dinitrotoluene (1 μM), and 3-nitro-1,2,4-triazol-5-one (10 μM) along with a common dye molecule (methylene blue, 5 nM). The concentrations of the dye and explosive molecules in terms of mass represent 31.98 pg, 11.45 ng, 1.82 ng, and 13.06 ng, respectively. Silver (Ag) and gold (Au) colloidal NPs were prepared by femtosecond laser (∼50 fs, 800 nm, 1 kHz) ablation of Ag/Au-target immersed in distilled water. Subsequently, the aggregated nanoparticles were achieved by mixing the pure Ag and Au NPs with different concentrations of NaCl. These aggregated NPs were characterized by UV-visible absorption and high-resolution transmission electron microscopy techniques. The SERS substrates were prepared by soaking the filter paper in aggregated NPs. The morphologies of the paper substrates were investigated using field-emission scanning electron microscopy technique. We have achieved superior enhancements with high reproducibility and sensitivity for filter paper substrates loaded with Ag/Au NPs mixed for an optimum concentration of 50 mM NaCl.

Long Spiky Au-Ag Nanostar Based Fiber Probe for Surface Enhanced Raman Spectroscopy.

Project description:The detection performances of noble metal-based surface enhanced Raman spectroscopy (SERS) devices are determined by the compositions and geometries of the metal nanostructures, as well as the substrates. In the current study, long spiky Au-Ag alloy nanostars were synthesized, and both core diameters and spike lengths were controlled by Lauryl sulfobetaine concentrations (as the nanostructure growth skeleton). The long spiky star geometries were confirmed by transmission electron micrograph images. Elements energy dispersive spectrometer mapping confirmed that Au and Ag elements were inhomogeneously distributed in the nanostructures and demonstrated a higher Ag content at surface for potential better SERS performance. Selected synthesized spiky nanostars were uniformly assembled on multi-mode silica fiber for probe fabrication by silanization. The SERS performance were characterized using crystal violet (CV) and rhodamine 6G (R6G) as analyte molecules. The lowest detection limit could reach as low as 10-8 M, with a 6.23 × 106 enhancement factor, and the relationship between analyte concentrations and Raman intensities was linear for both CV and R6G, which indicated the potential qualitative and quantitative molecule detection applications. Moreover, the fiber probes also showed good reproducibility and stability in the ambient atmosphere.

High optical enhancement in Au/Ag alloys and porous Au using Surface-Enhanced Raman spectroscopy technique.

Project description:We report high optical enhancement in Ag/Au alloys and porous gold nanostructures using Surface Enhanced Raman Spectroscopy (SERS) technique. Scanning electron microscopy investigation shows the formation of Ag/Au alloys particles during irradiation of Ag-Au bilayer deposited on FTO (SnO2:F) substrate by laser fluency equal to 0.5 J/cm2 or 1.0 J/cm2 with 12 ns laser pulse duration. The dealloying process of these Au-Ag alloy particles leads to the formation of Au nanoporous particles. The obtained nanostructures were studied with SERS and revealed a promising enhancement factor in porous Au nanostructure and tunability of localized surface plasmon resonance. The highly dense strong hot spots and large specific area in porous structure of gold nanostructures is the origin of the highly enhancement factor observed experimentally and theoretically. A very good agreement between simulation and experimental results was found confirming the potential of Au/Ag alloys and particularly porous gold nanostructure in SERS application.

Label-Free Detection of Glycan-Protein Interactions for Array Development by Surface-Enhanced Raman Spectroscopy (SERS).

Project description:A glyco-array platform has been developed, in which glycans are attached to plasmonic nanoparticles through strain-promoted azide-alkyne cycloaddition. Glycan-protein binding events can then be detected in a label-free manner employing surface-enhanced Raman spectroscopy (SERS). As proof of concept, we have analyzed the binding of Gal1, Gal3, and influenza hemagglutinins (HAs) to various glycans and demonstrated that binding partners can be identified with high confidence. The attraction of SERS for optical sensing is that it can provide unique spectral signatures for glycan-protein complexes, confirm identity through statistical validation, and minimizes false positive results common to indirect methods. Furthermore, SERS is very sensitive and has multiplexing capabilities thereby allowing the simultaneous detection of multiple analytes.

Surface Plasmon Resonance, Formation Mechanism, and Surface Enhanced Raman Spectroscopy of Ag+-Stained Gold Nanoparticles.

Project description:A series of recent works have demonstrated the spontaneous Ag+ adsorption onto gold surfaces. However, a mechanistic understanding of the Ag+ interactions with gold has been controversial. Reported herein is a systematic study of the Ag+ binding to AuNPs using several in-situ and ex-situ measurement techniques. The time-resolved UV-vis measurements of the AuNP surface plasmonic resonance revealed that the silver adsorption proceeds through two parallel pseudo-first order processes with a time constant of 16(±2) and 1,000(±35) s, respectively. About 95% of the Ag+ adsorption proceeds through the fast adsorption process. The in-situ zeta potential data indicated that this fast Ag+ adsorption is driven primarily by the long-range electrostatic forces that lead to AuNP charge neutralization, while the time-dependent pH data shows that the slow Ag+ binding process involves proton-releasing reactions that must be driven by near-range interactions. These experimental data, together with the ex-situ XPS measurement indicates that adsorbed silver remains cationic, but not as a charged-neutral silver atom proposed by the anti-galvanic reaction mechanism. The surface-enhanced Raman activities of the Ag+-stained AuNPs are slightly higher than that for AuNPs, but significantly lower than that for the silver nanoparticles (AgNPs). The SERS feature of the ligands on the Ag+-stained AuNPs can differ from that on both AuNPs and AgNPs. Besides the new insights to formation mechanism, properties, and applications of the Ag+-stained AuNPs, the experimental methodology presented in this work can also be important for studying nanoparticle interfacial interactions.

Surface-Enhanced Raman Spectroscopy on Self-Assembled Au Nanoparticles Arrays for Pesticides Residues Multiplex Detection under Complex Environment.

Project description:The high reproducibility of trace detection in complex systems is very hard but crucial to analytical technology and science. Here, we present a surface-enhanced Raman scattering (SERS) platform made by large-scale self-assembly of Au nanoparticle (NP) arrays at the cyclohexane/water interface and its use for pesticides residues trace detection. The analyte molecules spontaneously localize into the Au NPs' nanogaps during the self-assembly process, yielding excellent Raman signal enhancement by surface effects, and possibly both by the concentration of the analytes into the array and by plasmonic hot-spot formation. Transmission electron microscopy (TEM) images demonstrate a good uniformity of interparticle distances (2⁻3 nm) in the Au NP arrays. SERS experiments on crystal violet (CV) molecules demonstrated that the relative standard deviations (RSD) of the band intensities at 1173, 1376, and 1618 cm-1 were 6.3%, 6.4%, and 6.9%, respectively, indicating high reproducibility of the substrate. Furthermore, we demonstrate that two pesticides dissolved in organic and aqueous phases could be simultaneously detected, suggesting an excellent selectivity and universality of this method for multiplex detection. Our SERS platform opens vast possibilities for repeatability and sensitivity detection of targets in various complex fields.

Label-free characterization of exosome via surface enhanced Raman spectroscopy for the early detection of pancreatic cancer.

Project description:Pancreatic cancer is a highly lethal malignancy. Lack of early diagnostic markers makes timely detection of pancreatic cancer a highly challenging endeavor. Exosomes have emerged as information-rich cancer specific biomarkers. However, characterization of tumor-specific exosomes has been challenging. This study investigated the proof of principle that exosomes could be used for the detection of pancreatic cancer. Label-free analysis of exosomes purified from normal and pancreatic cancer cell lines was performed using surface enhanced Raman Spectroscopy (SERS) and principal component differential function analysis (PC-DFA), to identify tumor-specific spectral signatures. This method differentiated exosomes originating from pancreatic cancer or normal pancreatic epithelial cell lines with 90% accuracy. The cell line trained PC-DFA algorithm was next applied to SERS spectra of serum-purified exosomes. This method exhibited up to 87% and 90% predictive accuracy for HC and EPC individual samples, respectively. Overall, our study identified utility of SERS spectral signature for deciphering exosomal surface signature.
