Whole-genome sequence data of cellulase-producing fungi Trichoderma asperellum PK1J2, isolated from palm empty fruit bunch in Riau, Indonesia.

ABSTRACT: Trichoderma asperellum PK1J2 is a promising cellulase-producing fungus isolated from a palm empty fruit bunch in Riau, Indonesia. Presented here is the genome assembly of T. asperellum PK1J2. The whole genome of the fungi was sequenced using Illumina NovaSeq PE150. The genome assembly was performed using SOAPdenovo, SPAdes, and Abyss software, and the assembly results of the three types of software were integrated with CISA software. T. asperellum PK1J2 has 6,835 protein-coding genes with a length of 9,233,597 bp. The final genome assembly was approximately 36 Mbp with a GC content of 48.45%. This whole genome shotgun project has been deposited at DDBJ/ENA/GenBank under accession JAGJIK000000000.

Biosynthesis of Commodity Chemicals From Oil Palm Empty Fruit Bunch Lignin.

Project description:Lignin is one of the most abundant natural resources that can be exploited for the bioproduction of value-added commodity chemicals. Oil palm empty fruit bunches (OPEFBs), byproducts of palm oil production, are abundant lignocellulosic biomass but largely used for energy and regarded as waste. Pretreatment of OPEFB lignin can yield a mixture of aromatic compounds that can potentially serve as substrates to produce commercially important chemicals. However, separation of the mixture into desired individual substrates is required, which involves expensive steps that undermine the utility of OPEFB lignin. Here, we report successful engineering of microbial hosts that can directly utilize heterogeneous mixtures derived from OPEFB lignin to produce commodity chemicals, adipic acid and levulinic acid. Furthermore, the corresponding bioconversion pathway was placed under a genetic controller to autonomously activate the conversion process as the cells are fed with a depolymerized OPEFB lignin mixture. This study demonstrates a simple, one-pot biosynthesis approach that directly utilizes derivatives of agricultural waste to produce commodity chemicals.

Enhancing Bioethanol Productivity Using Alkali-Pretreated Empty Palm Fruit Bunch Fiber Hydrolysate.

Project description:Empty palm fruit bunch fiber (EPFBF) is a renewable resource in oil palm plantations that can be used for lignocellulosic bioethanol production. To enhance ethanol productivity with high-lignin-content EPFBF, the biomass was prepared with an alkali-thermal pretreatment (sodium hydroxide, 121°C, 60 min). The delignification yield was 55.4-56.9%, in proportion to the amount of sodium hydroxide, from 0.5 to 2.0 M. The lignin and hemicellulose contents of EPFBF were reduced by the pretreatment process, whereas the proportion of cellulose was increased. During enzymatic saccharification using Celluclast 1.5L and Novozyme 188 enzyme cocktails, about 62% of glucan was converted to a fermentable sugar. In simultaneous saccharification and fermentation, comparison among three ethanologenic yeast strains showed Saccharomyces cerevisiae W303-1A to be a candidate for maximum ethanol yield. In a batch fermentation with alkali-pretreated EPFBF hydrolysate, 21 g/L ethanol was obtained within 28 h, for a production yield of 0.102 g ethanol/g dry EPFBF or 0.458 g ethanol/g glucose. Moreover, a fed-batch fermentation produced 33.8±0.5 g/L ethanol with 1.57 g/L/h productivity in 20 h. These results show that the combination of alkaline pretreatment and biomass hydrolysate is useful for enhancing bioethanol productivity using delignified EPFBF.

Effect of Cellulose Nanocrystals Extracted from Oil Palm Empty Fruit Bunch as Green Admixture for Mortar.

Project description:This paper aims to examine the effect of cellulose nanocrystals (CNCs) derived from oil palm empty fruit bunch fiber (EFB) incorporating cement mortar on its structural performances. Cellulose nanocrystals (CNCs) were extracted from ?-cellulose extracted from EFB using an acid hydrolysis process with a concentration of acid used was 64% w/v under the temperature of 45?°C for 60?minutes. The Cellulose nanocrystals (CNCs) were mixed into the cement mortar ranging from 0 to 0.8% w/w and its mechanical properties were determined. The developed CNCs mortar was characterized for their compressive and flexural properties as well as microstructure. The influence of CNCs concentration, curing method, dispersion of CNCs on mortar's mechanical performance was thoroughly examined to find out the optimum condition. Overall results revealed that an addition of 0.4% cellulose nanocrystals has shown to increase the compressive and flexural strength to 46% and 20%, respectively cured under the wrapping method. The hydration of cementitious composites also improved significantly with the addition of CNCs by the formation of highly crystalline of portlandite observed under the XRD test. This present work demonstrates the importance of palm oil empty fruit bunch waste as a sustainable resource of cellulose nanocrystals admixture to achieve structural strength of cement mortar and promotes green technologies in construction.

Characterization dataset of oil palm empty fruit bunch (OPEFB) fibers - Natural reinforcement/filler for materials development.

Project description:Natural fibers used as reinforcements or fillers for materials development greatly affect properties and performance of end-use applications. As a consequence of conditioning processes such as grinding and sieving, average fiber length varies significantly. It is thus necessary to estimate the length as statistical data distribution rather than a single mean value. This approach implies length measurement of a significant number of fibers; however, a very high number of data points requires not only long-time frames but also significative amount of work. To address these issues, this article details a facile methodology to measure the length of a large number of natural fibers of oil palm empty fruit bunch (OPEFB) together with a statistical analysis to verify the correspondence between theoretical distributions and experimental data. Moreover, further information related to spectrophotometric, physico-chemical, mechanical, thermal, and morphological characteristics of OPEFB fibers coming from oil palm cultivation in Ecuador are presented. The data will contribute to comprehensively and rigorously describe the overall effects of natural fiber lengths on material properties.

Optimisation of Simultaneous Saccharification and Fermentation (SSF) for Biobutanol Production Using Pretreated Oil Palm Empty Fruit Bunch.

Project description:This study was conducted in order to optimise simultaneous saccharification and fermentation (SSF) for biobutanol production from a pretreated oil palm empty fruit bunch (OPEFB) by Clostridium acetobutylicum ATCC 824. Temperature, initial pH, cellulase loading and substrate concentration were screened using one factor at a time (OFAT) and further statistically optimised by central composite design (CCD) using the response surface methodology (RSM) approach. Approximately 2.47 g/L of biobutanol concentration and 0.10 g/g of biobutanol yield were obtained after being screened through OFAT with 29.55% increment (1.42 fold). The optimised conditions for SSF after CCD were: temperature of 35 °C, initial pH of 5.5, cellulase loading of 15 FPU/g-substrate and substrate concentration of 5% (w/v). This optimisation study resulted in 55.95% increment (2.14 fold) of biobutanol concentration equivalent to 3.97 g/L and biobutanol yield of 0.16 g/g. The model and optimisation design obtained from this study are important for further improvement of biobutanol production, especially in consolidated bioprocessing technology.

Potential of Oil Palm Empty Fruit Bunch Resources in Nanocellulose Hydrogel Production for Versatile Applications: A Review.

Project description:Oil palm empty fruit bunch (OPEFB) is considered the cheapest natural fiber with good properties and exists abundantly in Malaysia. It has great potential as an alternative main raw material to substitute woody plants. On the other hand, the well-known polymeric hydrogel has gathered a lot of interest due to its three-dimensional (3D) cross-linked network with high porosity. However, some issues regarding its performance like poor interfacial connectivity and mechanical strength have been raised, hence nanocellulose has been introduced. In this review, the plantation of oil palm in Malaysia is discussed to show the potential of OPEFB as a nanocellulose material in hydrogel production. Nanocellulose can be categorized into three nano-structured celluloses, which differ in the processing method. The most popular nanocellulose hydrogel processing methods are included in this review. The 3D printing method is taking the lead in current hydrogel production due to its high complexity and the need for hygiene products. Some of the latest advanced applications are discussed to show the high commercialization potential of nanocellulose hydrogel products. The authors also considered the challenges and future direction of nanocellulose hydrogel. OPEFB has met the requirements of the marketplace and product value chains as nanocellulose raw materials in hydrogel applications.

Dataset of alkaline ethylene glycol pretreatment and two-staged acid hydrolysis using oil palm empty fruit bunch.

Project description:Lignocellulosic biomass can provide a consistent and sustainable source of bioenergy. Degraded empty fruit bunch (DEFB) from oil palm is a suitable candidate for biofuels and biochemicals production by using alkaline ethylene glycol pretreatment and two-staged acid hydrolysis. This paper provided several important physical and chemical properties of treated DEFB after alkaline ethylene glycol pretreatment. The dataset for analysis methods which were used in two-staged acid hydrolysis for the determination of reducing sugars, phenolic compounds, furfural and hydroxymethyfurfural (HMF) in the acid hydrolysate was also provided. The key information and dataset provided in this article can be useful for researchers to determine the product compositions from lignocellulosic biomass after the pretreatment and acid hydrolysis processes in the future.

Chemical and structural changes of pretreated empty fruit bunch (EFB) in ionic liquid-cellulase compatible system for fermentability to bioethanol.

Project description:The pretreatment of empty fruit bunch (EFB) was conducted using an integrated system of IL and cellulases (IL-E), with simultaneous fermentation in one vessel. The cellulase mixture (PKC-Cel) was derived from Trichoderma reesei by solid-state fermentation. Choline acetate [Cho]OAc was utilized for the pretreatment due to its biocompatibility and biodegradability. The treated EFB and its hydrolysate were characterized by the Fourier transform infrared spectroscopy, scanning electron microscopy, and chemical analysis. The results showed that there were significant structural changes in EFB after the treatment in IL-E system. The sugar yield after enzymatic hydrolysis by the PKC-Cel was increased from 0.058 g/g of EFB in the crude sample (untreated) to 0.283 and 0.62 ± 06 g/g in IL-E system after 24 and 48 h of treatment, respectively. The EFB hydrolysate showed the eligibility for ethanol production without any supplements where ethanol yield was 0.275 g ethanol/g EFB in the presence of the IL, while lower yield obtained without IL-pretreatment. Moreover, it was demonstrated that furfural and phenolic compounds were not at the level of suppressing the fermentation process.

Maleic Acid-Butanol Pretreatment to Enhance Cellulose Accessibility for Enzymatic Hydrolysis and Ethanol Production from Oil Palm Empty Fruit Bunch.

Project description:Pretreatment of lignocellulosic biomass is crucial yet challenging for sustainable energy production. This study focuses on enhancing enzymatic accessibility of cellulose in oil palm empty fruit bunches by optimizing pretreatment parameters to improve glucose and ethanol yields while reducing fermentation inhibitors. It evaluates the impact of maleic acid concentrations on biorefinery processes. High maleic acid concentrations (>25% w/w) may allow reuse and offer benefits over lower concentrations, such as enhanced delignification and increased sugar yield under milder conditions. Biomass undergoes pretreatment, enzymatic saccharification, and fermentation using Saccharomyces cerevisiae F118. Pretreatment with 75% maleic acid (w/w) for 60 min at 180 °C effectively removes lignin and hemicellulose, increasing cellulose accessibility but results in 74.8% crystallinity, hindering saccharification. A 50% maleic acid pretreatment yielded higher glucose (77.1%). Optimal ethanol production is achieved with 1% maleic acid pretreatment. However, the ethanol yield is negatively impacted by residual maleic acid on the solid matrix.
