Association between rumen microbiota and marbling grade in Hu sheep.

ABSTRACT: The marbling fat regulates the flavor of mutton and measures the fat density in the loin eye and is the most important parameter of carcass grading. The objective of this study was to explore the relationship of rumen microbiota and mutton marbling grade. One hundred and eighty-seven feedlot-finished Hu male lambs (Age: 180 day; Final BW: 46.32 ± 6.03 kg) were slaughtered, and ruminal contents and marbling grade were collected. Ruminal microbial DNA extraction and 16S rRNA gene sequencing was performed to investigate microbial composition and to predict microbial metabolic pathways. The animal cohort was then grouped based on marbling grades [low marbling (LM), marbling grade ≤ 1; Medium marbling (MM), 1 < marbling grade ≤ 3; High Marbling (HM), 3 < marbling grade ≤ 5] and intramuscular fat-associated microorganisms were pinpointed using LEfSe and random forest classification model. Intramuscular fat content had significantly differences among the three groups (P < 0.05), and was significantly correlated with VFAs profiling. HM sheep showed a higher abundance of one bacterial taxon (Kandleria), and two taxa were overrepresented in the MM sheep (Pseudobutyrivibrio and Monoglobus), respectively. In addition, the main intramuscular fat deposition pathway was found to involve peroxisome proliferator-activated receptor (PPAR) fatty acid synthesis. By studying the effect of the ruminal microbiome on the marbling of sheep, the present study provides insights into the production of high-quality mutton.

Project description:BackgroundCellulosic biomass has great potential as a renewable biofuel resource. Robust, high-performance enzymes are needed to effectively utilize this valuable resource. In this study, metatranscriptomics was used to explore the carbohydrate-active enzymes (CAZymes), especially glycoside hydrolases (GHs), present in the rumen microbiome of Hu sheep. Select CAZymes were experimentally verified and characterized after cloning and expression in E. coli.ResultsThe metatranscriptomes of six Hu sheep rumen microbiomes yielded 42.3 Gbp of quality-checked sequence data that represented in total 2,380,783 unigenes after de novo assembling using Trinity and clustered with CD-HIT-EST. Annotation using the CAZy database revealed that 2.65% of the unigenes encoded GHs, which were assigned to 111 different CAZymes families. Firmicutes (18.7%) and Bacteroidetes (13.8%) were the major phyla to which the unigenes were taxonomically assigned. In total, 14,489 unigenes were annotated to 15 cellulase-containing GH families, with GH3, GH5 and GH9 being the predominant. From these putative cellulase-encoding unigenes, 4225 open reading frames (ORFs) were predicted to contain 2151 potential cellulase catalytic modules. Additionally, 147 ORFs were found to encode proteins that contain carbohydrate-binding modules (CBMs). Heterogeneous expression of 30 candidate cDNAs from the GH5 family in E. coli BL21 showed that 17 of the tested proteins had endoglucanase activity, while 7 exhibited exoglucanase activity. Interestingly, two of the GH5 proteins (Cel5A-h28 and Cel5A-h11) showed high specific activity against carboxymethylcellulose (CMC) and p-nitrophenyl-β-d-cellobioside (pNPC) (222.2 and 142.8 U/mg), respectively. The optimal pH value for activity of Cel5A-h11 and Cel5A-h28 was 6.0 for both enzymes, and optimal temperatures were 40 and 50 °C, respectively. Both enzymes retained over 70 and 60%, respectively, of their original activities after incubation at 40 °C for 60 min. However, their activities were rapidly diminished upon exposure to higher temperatures. Cel5A-h11 and Cel5A-h28 retained more than 80 and 60% of their maximal enzymatic activities after incubation for 16 h in buffered solutions in the pH range from 4.0 to 9.0.ConclusionThe metatranscriptomic results revealed that the rumen microbiome of Hu sheep encoded a repertoire of new enzymes capable of cellulose degradation and metatranscriptomics was an effective method to discover novel cellulases for biotechnological applications.

Project description:BackgroundThis research aimed to investigate differences in rumen fermentation characteristics between Karakul sheep and Hu sheep reared under identical conditions. The test subjects included newborn Hu and Karakul sheep, which were monitored across three stages: stage I (Weaning period: 15 ~ 30 days), stage II (Supplementary feeding period: 31 ~ 90 days), and stage III (Complete feeding period: 91 ~ 150 days). During the supplementary feeding period, cottonseed hulls were the main roughage source. To analyze the dynamics of rumen fermentation, 16S rRNA sequencing and metabolomics methods were employed, alongside measurements of rumen fermentation parameters and cellulase activity. This comprehensive approach aimed to investigate the potential impact of breed on rumen fermentation indicators, microbial community structure, and metabolites in Hu and Karakul sheep.ResultsThe 16S rRNA sequencing analysis revealed no significant differences in the relative abundance or dominant bacterial communities in the rumen across all stages. In stage II, rumen bacteria in both Hu and Karakul sheep were relatively stable. However, the Simpson index of Hu sheep in stage II was substantially greater than that of Karakul sheep, demonstrating similarities in the rumen microbial structure between stages II and III. Dynamic variations in fermentation parameters and cellulase activity in the rumen revealed that the indicators in both sheep breeds stabilized at 150 days. Metabolomic results revealed that the metabolic pathways in stage I were mainly concentrated in purine metabolism and lipid metabolism, while stage II was dominated by amino acid metabolism. Stage III involved mainly in pyrimidine and purine metabolism. An exploration of the relationships among rumen microbial biomarkers, key differentially abundant metabolites and rumen characteristics indicated that Karakul sheep exhibited superior lipid metabolism compared to Hu sheep.ConclusionThese findings reveal that there were no interbreed differences in the rumen characteristics of Hu and Karakul sheep when fed the same cottonseed hull diet, despite differences in their metabolic pathways. The findings also indicate that the first 20 days represent the initial stage of rumen bacteria in Hu sheep, followed by a transition phase between 20 and 90 days, and a relatively stable stage from 90 to 150 days. These results provide a scientific basis for further understanding the rumen function of sheep and for optimizing their feeding strategies. Video Abstract.
