Synthesis of meso-pyrrole-substituted corroles by condensation of 1,9-diformyldipyrromethanes with pyrrole.

ABSTRACT: A copper triflate-mediated approach to access copper complexes of pyrrole-substituted corroles from the reaction of 1,9-diformyldipyrromethanes and an excess amount of pyrrole is presented for the first time. This procedure is a simple and efficient way for the preparation of corroles with a polymerizable substituent on meso-positions.

Project description:AbstractWe report the chemical synthesis and characterization of the stable 5,15-bis(pentafluorophenyl)-10-(trimethylsilylethynyl)corrole which serves as a precursor for the subsequent in situ sila-Sonogashira-cross-coupling reaction and metalation with copper(II) acetate. Under ambient conditions and a common catalyst system the reaction with 1-iodopyrene occurred within five hours. Due to the direct conjugation of the 18π-electronic system of the corrole macrocycle over the alkynyl group to the pyrene moiety the optical transitions in the Soret (B-) band Q-band region are significantly altered. The copper corrole exhibited complex hyperfine and superhyperfine structure in the EPR spectrum. The assignment of the EPR spectrum reveals the existence of an axial [CuII-cor∙+] species.Graphical abstract

Project description:Novel dihydro-1,3-oxazinoporphyrins and naphtho[e]bis(dihydro-1,3-oxazinoporphyrin) derivatives, in which the porphyrin macrocycle is covalently linked to the dihydro-1,3-oxazine ring system were successfully synthesized from 5-(4-aminophenyl)-10,15,20-triphenylporphyrin in good yields. The structures of the target products were established on the basis of spectral data and elemental analyses.

Atom Economical Multi-Substituted Pyrrole Synthesis from Aziridine.

Synthesis and Structure of meso-Substituted Dibenzihomoporphyrins.

Project description:Bench-stable meso-substituted di(p/m-benzi)homoporphyrins were synthesized through acid-catalyzed condensation of dipyrrole derivatives with aryl aldehydes. The insertion of a 1,1,2,2-tetraphenylethene (TPE) or but-2-ene-2,3-diyldibenzene unit in the porphyrin framework results in the formation of dibenzihomoporphyrins, merging the features of hydrocarbons and porphyrins. Single crystal X-ray analyses established the non-planar structure of these molecules, with the phenylene rings out of the mean plane, as defined by the dipyrromethene moiety and the two meso-carbon atoms. Spectroscopic and structural investigations show that the macrocycles exhibit characteristics of both TPE or but-2-ene-2,3-diyldibenzene and dipyrromethene units indicating the non-aromatic characteristics of the compounds synthesized. Additionally, the dibenzihomoporphyrins were found to generate singlet oxygen, potentially allowing their use as photosensitizers.

First synthesis of meso-substituted pyrrolo[1,2-a]quinoxalinoporphyrins.

Project description:A synthetic protocol for the construction of new meso-substituted pyrrolo[1,2-a]quinoxalinoporphyrins is described starting from 5-(4-amino-3-nitrophenyl)-10,15,20-triphenylporphyrin. The reaction of this porphyrin with 2,5-dimethoxytetrahydrofuran, followed by the reduction of the nitro group in the presence of NiCl2/NaBH4 afforded 5-(3-amino-4-(pyrrol-1-yl)phenyl)-10,15,20-triphenylporphyrin. This triphenylporphyrin underwent a Pictet-Spengler cyclization after the reaction with various aromatic aldehydes followed by in situ KMnO4 oxidation to form target porphyrin analogues in good yields. The structures of all synthesized products were established on the basis of spectral data and elemental analyses.

Synthesis of meso-substituted subphthalocyanine-subporphyrin hybrids: boron subtribenzodiazaporphyrins.

Project description:The first syntheses of hybrid structures that lie between subphthalocyanines and subporphyrins are reported. The versatile single-step synthetic method uses a preformed aminoisoindolene to provide the bridging methine unit and its substituent while trialkoxyborates simultaneously act as Lewis acid, template, and provider of the apical substituent. The selection of each component therefore allows for the controlled formation of diverse, differentially functionalized systems. The new hybrids are isolated as robust, pure materials that display intense absorption and emission in the mid-visible region. The new compounds are further characterized in solution and solid state by variable-temperature NMR spectroscopy and X-ray crystallography, respectively.

Synthesis, aggregation induced emission and through space conjugation of triphenylvinylphenyl substituted [2.2]paracyclophane-1,9-diene.

Project description:4-Bromo substituted [2.2]paracyclophane-1,9-diene was synthesized from the corresponding dithia[3.3]paracyclophane in three steps through benzyne Steven rearrangement, oxidation, and a thermal elimination reaction. 4-Triphenylvinylphenyl substituted [2.2]paracyclophane-1,9-diene was successfully prepared by the Suzuki-Miyaura cross-coupling reaction of 4-bromo substituted [2.2]paracyclophane-1,9-diene and 4,4,5,5-tetramethyl-2-(4-(1,2,2-triphenylvinyl)phenyl)-1,3,2-dioxaborolane using Pd(OAc)2 as a catalyst, S-Phos as a ligand and K3PO4 as a base. The structures of bromo substituted [2.2] paracyclophane-1,9-diene and triphenylvinylphenyl substituted [2.2]paracyclophane-1,9-diene were fully characterized by 1H NMR spectroscopy and X-ray crystallography. 4-Triphenylvinylphenyl substituted [2.2]paracyclophane-1,9-diene exhibited aggregation-induced emission characteristics when the water fraction was higher than 80% in the THF/water mixtures. 4-Triphenylvinylphenyl substituted [2.2]paracyclophane-1,9-diene displays much higher fluorescence when the water fraction is 90% compared to that of model compounds due to both through bond and through space conjugation. To the best for our knowledge, we are the first to synthesize triphenylvinylphenyl substituted [2.2]paracyclophane-1,9-diene with aggregation-induced emission characteristics.

Photophysical properties and singlet oxygen generation of meso-iodinated free-base corroles.

Project description:In order to study the effect of meso-iodination of free-base corroles on their photophysical character, we designed and synthesized a series of free-base corrole derivatives F10-OH (iodine-free), F10-OH-I (mono-iodo) and F10-OH-2I (di-iodo), with different substitution patterns at the meso-position as candidates for photodynamic therapy (PDT). We employed several optical spectroscopic techniques, including time-resolved spectroscopy from a femtosecond to microsecond and singlet oxygen luminescence to study the properties of excited singlet and triplet states, as well as the singlet oxygen quantum yields. The sub-picosecond internal conversion, ∼1 ps intramolecular vibrational energy redistribution, tens of ps vibrational cooling, are similar across the three corroles. The addition of one (F10-OH-I) and two iodine (F10-OH-2I) atoms to the remote aryl ring of triarylcorroles induces a 4.6-fold and 6.2-fold decrease in fluorescence quantum yields Φ fl and a 2.2-fold and 4.9-fold increase in the time constant of intersystem crossing k ISC. In addition, a slight increase in intersystem crossing quantum yields Φ T was also observed from F10-OH to F10-OH-2I. It means the intersystem crossing is improved by the iodination, from F10-OH to F10-OH-2I, because of the heavy atom effect. However, the sample F10-OH-I, instead of F10-OH-2I, shows the highest singlet oxygen quantum yield Φ Δ.

Synthesis and Biological Screening of New Cyano-Substituted Pyrrole Fused (Iso)Quinoline Derivatives.

Project description:Several new cyano-substituted derivatives with pyrrolo[1,2-a]quinoline and pyrrolo[2,1-a]isoquinoline scaffolds were synthesized by the [3 + 2] cycloaddition of (iso)quinolinium ylides to fumaronitrile. The cycloimmonium ylides reacted in situ as 1,3-dipoles with fumaronitrile to selectively form distinct final compounds, depending on the structure of the (iso)quinolinium salt. Eleven compounds were evaluated for their anticancer activity against a panel of 60 human cancer cell lines. The most potent compound 9a showed a broad spectrum of antiproliferative activity against cancer cell lines representing leukemia, melanoma and cancer of lung, colon, central nervous system, ovary, kidney, breast and prostate cancer. In vitro assays and molecular docking revealed tubulin interaction properties of compound 9a.

Synthesis and Optical Properties of Near-Infrared meso-Phenyl-Substituted Symmetric Heptamethine Cyanine Dyes.

Project description:Heptamethine cyanine dyes are a class of near infrared fluorescence (NIRF) probes of great interest in bioanalytical and imaging applications due to their modifiability, allowing them to be tailored for particular applications. Generally, modifications at the meso-position of these dyes are achieved through Suzuki-Miyaura C-C coupling and SRN1 nucleophilic substitution of the chlorine atom at the meso-position of the dye. Herein, a series of 15 meso phenyl-substituted heptamethine cyanines was synthesized utilizing a modified dianil linker. Their optical properties, including molar absorptivity, fluorescence, Stokes shift, and quantum yield were measured. The HSA binding affinities of two representative compounds were measured and compared to that of a series of trimethine cyanines previously synthesized by our lab. The results indicate that the binding of these compounds to HSA is not only dependent on hydrophobicity, but may also be dependent on steric interferences in the binding site and structural dynamics of the NIRF compounds.
