Fabrication of Encapsulated Gemini Surfactants.

ABSTRACT: (1) Background: Encapsulation of surfactants is an innovative approach that allows not only protection of the active substance, but also its controlled and gradual release. This is primarily used to protect metallic surfaces against corrosion or to create biologically active surfaces. Gemini surfactants are known for their excellent anticorrosion, antimicrobial and surface properties; (2) Methods: In this study, we present an efficient methods of preparation of encapsulated gemini surfactants in form of alginate and gelatin capsules; (3) Results: The analysis of infrared spectra and images of the scanning electron microscope confirm the effectiveness of encapsulation; (4) Conclusions: Gemini surfactants in encapsulated form are promising candidates for corrosion inhibitors and antimicrobials with the possibility of protecting the active substance against environmental factors and the possibility of controlled outflow.

Project description:Diseases that are linked to defective genes or mutations can in principle be cured by gene therapy, in which damaged or absent genes are either repaired or replaced by new DNA in the nucleus of the cell. Related to this, disorders associated with elevated protein expression levels can be treated by RNA interference via the delivery of siRNA to the cytoplasm of cells. Polynucleotides can be brought into cells by viruses, but this is not without risk for the patient. Alternatively, DNA and RNA can be delivered by transfection, i.e. by non-viral vector systems such as cationic surfactants, which are also referred to as cationic lipids. In this review, recent progress on cationic lipids as transfection vectors will be discussed, with special emphasis on geminis, surfactants with 2 head groups and 2 tails connected by a spacer.

Antimicrobial Activity of Gemini Surfactants with Azapolymethylene Spacer.

Project description:A series of 21 azapolymethylene gemini surfactants were obtained. The synthesis of the title surfactants in one- or two-step reaction proceeds with good yields. The structure and the purity of the synthesized compounds were determined by 1H and 13C NMR, ESI-MS spectra, and elemental analysis. Moreover, 2D COSY, HMBC, and HSQC spectra were performed. The minimal inhibitory concentrations (MIC) of the synthesized compounds were determined against fungi: Candida albicans, Aspergillus niger, Penicillium chrysogenum and bacteria: Escherichia coli,Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus subtilis. Also, the critical micelle concentrations (CMC) were determined. The relationship between antimicrobial and surface activity and surfactant structure has been determined.

Structure-Stability Relationship in Aqueous Colloids of Latex Particles and Gemini Surfactants.

Project description:The influence of gemini surfactants (GSs) on the charging and aggregation features of anionic sulfate modified latex (SL) particles was investigated by light scattering techniques in aqueous dispersions. The GSs of short alkyl chains (2-4-2 and 4-4-4) resembled simple inert salts and aggregated the particles by charge screening. The adsorption of GSs of longer alkyl chains (8-4-8, 12-4-12, and 12-6-12) on SL led to charge neutralization and overcharging of the particles, giving rise to destabilization and restabilization of the dispersions, respectively. The comparison of the interfacial behavior of dimeric and the corresponding monomeric surfactants revealed that the former shows a more profound influence on the colloidal stability due to the presence of double positively charged head groups and hydrophobic tails, which is favorable to enhancing both electrostatic and hydrophobic particle-GS and GS-GS interactions at the interface. The different extent of the particle-GS interactions was responsible for the variation of the GS destabilization power, following the 2-4-2 < 4-4-4 < 8-4-8 < 12-4-12 order, while the length of the GS spacer did not affect the adsorption and aggregation processes. The valence of the background salts strongly influenced the stability of the SL-GS dispersions through altering the electrostatic interactions, which was more pronounced for multivalent counterions. These findings indicate that both electrostatic and hydrophobic effects play crucial roles in the adsorption of GSs on oppositely charged particles and in the corresponding aggregation mechanism. The major interparticle forces can be adjusted by changing the structure and concentration of the GSs and inorganic electrolytes present in the systems.

Evaluation of the antiviral potential of gemini surfactants against influenza virus H1N1.

Project description:Influenza A virus (IAV) affects human health worldwide as a high-risk disease. It can neither be easily controlled by current vaccines and nor be treated by conventional drugs. Gemini surfactants (GS) have shown several properties including antiviral activity. In this study, the antiviral capacity of some GS compounds with different levels of hydrophobicity was examined. The 50% cytotoxic (CC50) and non-cytotoxic (NCTC) concentrations of the compounds were determined by MTT method. The NCTCs, the same as effective concentrations (EC50s), were tested for the antiviral capacity against IAV in different combination treatments for 1 h incubation on MDCK cells. The HA and MTT assays were used to evaluate the virus titer and cell viabilities, respectively. The hemolytic activity of the compounds was also assessed using an HA inhibition assay. To evaluate the apoptotic effect of GS compounds, Annexin V-PI kit was used. The HA titers decreased between 1-6.5 logs, 1-4.5 logs, and 1-5.5 logs in simultaneous, pre- and post-penetration combination treatments, respectively. The cell viability values in all combination treatments were favorable. The HI assay indicated the hemolytic potential of GSs and their physical interaction with viral HA. The apoptosis test results highlighted anti-apoptotic capacity of the GS compounds alone and in the presence of influenza virus especially for the hydrophobic ones. Gemini surfactants were generally more efficacious in simultaneous treatment. Their antiviral potential may be attributed to their physical interaction with viral membrane or HA glycoprotein that disrupts viral particle or blocks viral entry to the cell and inhibits its propagation.

Antimicrobial Activity of Gemini Surfactants with Ether Group in the Spacer Part.

Project description:Due to their large possibility of the structure modification, alkylammonium gemini surfactants are a rapidly growing class of compounds. They exhibit significant surface, aggregation and antimicrobial properties. Due to the fact that, in order to achieve the desired utility effect, the minimal concentration of compounds are used, they are in line with the principle of greenolution (green evolution) in chemistry. In this study, we present innovative synthesis of the homologous series of gemini surfactants modified at the spacer by the ether group, i.e., 3-oxa-1,5-pentane-bis(N-alkyl-N,N-dimethylammonium bromides). The critical micelle concentrations were determined. The minimal inhibitory concentrations of the synthesized compounds were determined against bacteria Escherichia coli ATCC 10536 and Staphylococcus aureus ATCC 6538; yeast Candida albicans ATCC 10231; and molds Aspergillus niger ATCC 16401 and Penicillium chrysogenum ATCC 60739. We also investigated the relationship between antimicrobial activity and alkyl chain length or the nature of the spacer. The obtained results indicate that the synthesized compounds are effective microbicides with a broad spectrum of biocidal activity.

Monolayer formed by l-Asp-based gemini surfactants self-assembled in 1D nanostructures.

Project description:Herein, studies on the surface activities of newly synthesized l-Asp-based gemini surfactants, both nonionic and anionic, are presented. Conductometry, tensiometry, and the Langmuir-Blodgett (LB) film technique were applied for this purpose. π-A isotherms were obtained with a Langmuir trough and Wilhelmy balance. The structures of the monolayers assembled at the air/water interface and those deposited as LB films were studied via Brewster angle microscopy (BAM) and atomic force microscopy (AFM). The 2D films formed by the anion-active compounds show a well-known pattern of a monolayer film, whereas the nonionogenic amphiphiles have been found to be 1D structures with nano-widths and micro-lengths that align with each other during the process of compression; this is the first study where the organization of 1D fibrils in 2D films during compression is reported. The scanning electron microscopy (SEM) study reveals that 1D nanostructure formation is an intrinsic tendency of these molecules as not only nonionogenic surfactants, but also the anion active representatives have been constructed in the solid state by fibrillary structures.

Action of Monomeric/Gemini Surfactants on Free Cells and Biofilm of Asaia lannensis.

Project description:We investigated the biological activity of surfactants based on quaternary ammonium compounds: gemini surfactant hexamethylene-1,6-bis-(N,N-dimethyl-N-dodecylammonium bromide) (C6), synthesized by the reaction of N,N-dimethyl-N-dodecylamine with 1,6-dibromohexane, and its monomeric analogue dodecyltrimethylammonium bromide (DTAB). The experiments were performed with bacteria Asaia lannensis, a common spoilage in the beverage industry. The minimal inhibitory concentration (MIC) values were determined using the tube standard two-fold dilution method. The growth and adhesive properties of bacterial cells were studied in different culture media, and the cell viability was evaluated using plate count method. Both of the surfactants were effective against the bacterial strain, but the MIC of gemini compound was significantly lower. Both C6 and DTAB exhibited anti-adhesive abilities. Treatment with surfactants at or below MIC value decreased the number of bacterial cells that were able to form biofilm, however, the gemini surfactant was more effective. The used surfactants were also found to be able to eradicate mature biofilms. After 4 h of treatment with C6 surfactant at concentration 10 MIC, the number of bacterial cells was reduced by 91.8%. The results of this study suggest that the antibacterial activity of the gemini compound could make it an effective microbiocide against the spoilage bacteria Asaia sp. in both planktonic and biofilm stages.

Synthesis, surface activity, and corrosion inhibition capabilities of new non-ionic gemini surfactants.

Project description:Several environmentally acceptable non-ionic gemini surfactants are synthesized in this work using natural sources, including polyethenoxy di-dodecanoate (GSC12), polyethenoxy di-hexadecanoate (GSC16), and polyethenoxy di-octadecenoate (GSC18). The produced surfactants are confirmed by spectrum studies using FT-IR, 1HNMR, and 13CNMR. It explored and examined how the length of the hydrocarbon chain affected essential properties like foaming and emulsifying abilities. Surface tension examinations are used to assess the surface activity of the examined gemini surfactants. The lower value of critical micelle concentrations (0.381 × 10-4M) is detected for GSC18. Their spontaneous character is shown by the negative values of the free energy of adsorption (ΔGads) and micellization (ΔGmic) which arranged in the order GSC18 > GSC16 > GSC12. Based on theoretical, weight loss, and electrochemical investigations, these novel surfactants were investigated for their possible use in inhibiting carbon steel from corroding in 1 M HCl. Measuring results show that GSC18 inhibits corrosion in carbon steel by 95.4%. The isotherm of adsorption evaluated for the investigated inhibitors and their behavior obeys Langmuir isotherm.

A Systematic Approach: Molecular Dynamics Study and Parametrisation of Gemini Type Cationic Surfactants.

Project description:The spreading of antibiotic-resistant bacteria strains is one of the most serious problem in medicine to struggle nowadays. This triggered the development of alternative antimicrobial agents in recent years. One of such group is Gemini surfactants which are massively synthesised in various structural configurations to obtain the most effective antibacterial properties. Unfortunately, the comparison of antimicrobial effectiveness among different types of Gemini agents is unfeasible since various protocols for the determination of Minimum Inhibitory Concentration are used. In this work, we proposed alternative, computational, approach for such comparison. We designed a comprehensive database of 250 Gemini surfactants. Description of structure parameters, for instance spacer type and length, are included in the database. We parametrised modelled molecules to obtain force fields for the entire Gemini database. This was used to conduct in silico studies using the molecular dynamics to investigate the incorporation of these agents into model E. coli inner membrane system. We evaluated the effect of Gemini surfactants on structural, stress and mechanical parameters of the membrane after the agent incorporation. This enabled us to select four most likely membrane properties that could correspond to Gemini's antimicrobial effect. Based on our results we selected several types of Gemini spacers which could demonstrate a particularly strong effect on the bacterial membranes.
