Phomoxanthone A, Compound of Endophytic Fungi Paecilomyces sp. and Its Potential Antimicrobial and Antiparasitic.

ABSTRACT: The present work reports the isolation and biological evaluation of three dimeric xanthones from Paecilomyces sp. EJC01.1 isolated as endophytic from Schnella splendens, a typical plant of the Amazon. The compounds phomoxanthone A (1), phomoxanthone B (2) and dicerandrol B (3) were isolated by chromatographic procedures and identified by spectroscopic methods of 1D and 2D NMR and MS. The extracts and compound 1 showed antimicrobial activities against Bacillus subtilis, Escherichia coli, Staphylococcus aureus, Salmonella typhimurium and Pseudomonas aeruginosa. The compound phomoxanthone A (1) showed greater inhibitory activity against B. subtilis (MIC of 7.81 µg mL^-1); in addition, it also pronounced inhibitory effect against promastigote forms of Leishmania amazonensis (IC₅₀ of 16.38 ± 1.079 µg mL^-1) and epimastigote forms Trypanosoma cruzi (IC₅₀ of 28.61 ± 1.071 µg mL^-1). To provide more information about the antibacterial activity of compound 1, an unprecedented molecular docking study was performed using S-ribosyl-homocysteine lyase (LuxS) (PDB ID 2FQO), which showed a possible interaction of phomoxanthone A with two of the residues (His58 and Cys126) that are fundamental for the catalysis mechanism in B. subtilis, which may be associated with the higher activity, when compared to other bacteria, observed in experimental studies. Additionally, quantum studies (DFT) were performed, for which a low gap value (5.982 eV) was observed, which corroborates the reactivity of phomoxanthone A. Thus, phomoxanthone A can be a good agent against pathogenic bacteria.

Project description:Endophytic fungi are microorganisms that, exhibiting within the plant tissues without causing any apparent harm to the host, establish a symbiotic relationship with plants. Host plants provide endophytic fungi with essential nutrients and a protected environment. In exchange, the fungi can enhance the plant's ability to acquire nutrients. They can also play a crucial role in increasing the host plant's tolerance to various abiotic and biotic stresses. Endophytic fungi can produce a wide range of bioactive compounds, some similar to those found in the host plant. In Iraq's Duhok province of the Kurdistan region, the plant species Vitis vinifera has been explored as a habitat for diverse endophytic microorganisms across various ecological environments. During the period from 2021 to 2022, a total of 600 samples were collected from four distinct locations: Bagera, Besfke, Barebhar, and Atrush. From these samples, twelve endophytic fungal species were isolated, including Aspergillus flavipes, Botryosphaeria dothidea, Fusarium oxysporum, Fusarium ruscicol, Fusarium venenatum, Chaetomium globosum, Clonostachys rosea, Mucor racemosus, Penicillium glabrum, Aspergillus terreus, Aspergillus nidulans, and Aspergillus niger, Alternaria alternata, Paecilomyces maximus, Curvularia buchloes. These fungi were introduced for their potential as biocontrol agents against grapevine trunk diseases and grape rotting fungi, which pose significant risks to grapevine health and productivity. Penicilium radiatolobatum, Botrysphaeria dothidea, Fusarium ruscicola, Fusarium venenatum, and Paecilomyces maximus represented the first record as endophytes on grapevine in Iraq. Based on ITS and SSU sequencing, molecular identification confirmed these fungi's presence with sequence identities ranging from 99% to 100%. Phylogenetic analysis revealed that these endophytes could be categorized into five main clusters (A, B, C, D, and E), showing high intra-group similarity. Utilizing the Dual Culture method, the endophyte Paecilomyces maximus demonstrated a 70.83% inhibition rate against Ilyonectria destructans. In the Food Poisoning method, A. flavipes and P. maximus emerged as the most effective inhibitors of Ilyonectria destructans, whereas A. terreus, M. racemosus, and P. maximus achieved complete inhibition (100%) of Botrytis cinerea. Additionally, M. racemosus was identified as the most effective biocontrol agent against Neoscytalidium dimidiatum. In conclusion, the study emphasizes the potential of endophytic fungi from Vitis vinifera as effective biocontrol agents against grapevine diseases, highlighting their role in sustainable vineyard management. These findings lead to further exploration and implementation of these fungi-inserted pest management strategies.

Project description:Nowadays, endophytic fungi represent a rich source of biological active compounds. In the current study, twelve endophytic fungal species were isolated from Avicennia marina leaves. From the isolates, Aspergillus niger, Penicillium rubens and Alternaria alternata recorded the highest isolation frequency (80%), relative density (12.5%) and antimicrobial activity. The antimicrobial and anticancer activities of P. rubens were more effective than those of A. niger and A. alternata; therefore, its identification was confirmed via the ITS rRNA gene. Filtrate extracts of P. rubens, A. alternata and A. niger were analyzed using GC-MS and showed different detected constituents, such as acetic acid ethyl ester, N-(4,6-Dimethyl-2-pyrimidinyl)-4-(4-nitrobenzylideneamino) benzenesulfonamide, 1,2-benzenedicarboxylic acid, hexadecanoic acid and octadecanoic acid. Filtrate extract of P. rubens exhibited the presence of more compounds than A. alternata and A. niger. Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Aspergillus fumigatus were more inhibited by P. rubens extract than A. alternata or A. niger, with inhibition zones of 27.2 mm, 22.21 mm, 26.26 mm, 27.33 mm, 28.25 mm and 8.5 mm, respectively. We observed negligible cytotoxicity of P. rubens extract against normal cells of human lung fibroblasts (WI-38 cell line), unlike A. alternata and A. niger extracts. Proliferation of prostate cancer (PC-3) was inhibited using P. rubens extract, exhibiting mortality levels of 75.91% and 76.2% at 200 µg/mL and 400 µg/mL of the extract. Molecular docking studies against the crystal structures of C. albicans (6TZ6) and the cryo-EM structure of B. subtilis (7CKQ) showed significant interactions with benzenedicarboxylic acid and N-(4,6-dimethyl-2-pyrimidinyl)-4-(4-nitrobenzylideneamino) benzenesulfonamide as a constituent of P. rubens extract. N-(4,6-dimethyl-2-pyrimidinyl)-4-(4-nitrobenzylideneamino) benzenesulfonamide had the highest scores of −6.04905 kcal/mol and −6.590 kcal/mol towards (6tz6) and (7CKQ), respectively.
