The Use of PGPB to Promote Plant Hydroponic Growth.

ABSTRACT: Improvements to the world's food supply chain are needed to ensure sufficient food is produced to meet increasing population demands. Growing food in soilless hydroponic systems constitutes a promising strategy, as this method utilizes significantly less water than conventional agriculture, can be situated in urban areas, and can be stacked vertically to increase yields per acre. However, further research is needed to optimize crop yields in these systems. One method to increase hydroponic plant yields involves adding plant growth-promoting bacteria (PGPB) into these systems. PGPB are organisms that can significantly increase crop yields via a wide range of mechanisms, including stress reduction, increases in nutrient uptake, plant hormone modulation, and biocontrol. The aim of this review is to provide critical information for researchers on the current state of the use of PGPB in hydroponics so that meaningful advances can be made. An overview of the history and types of hydroponic systems is provided, followed by an overview of known PGPB mechanisms. Finally, examples of PGPB research that has been conducted in hydroponic systems are described. Amalgamating the current state of knowledge should ensure that future experiments can be designed to effectively transition results from the lab to the farm/producer, and the consumer.

Project description:BackgroundThe primary objective is to address the specific needs of plants at different growth stages by delivering precise nutrient concentrations tailored to their developmental requirements. Challenges such as uneven nutrient distribution, fluctuations in pH and electrical conductivity, and inadequate nutrient delivery pose potential hindrances to achieving optimal plant health and yield in hydroponic systems. By overcoming these challenges, the hydroponic farming community aims to enhance the accuracy of nutrient dosing, streamline automation processes, and minimize resource wastage. Hydroponics, a cultivation technique without soil, facilitates the growth of organic vegetation while concurrently minimizing water use and eliminating the necessity for pesticides. In order to achieve effective cultivation of hydroponic plants, it is essential to maintain a controlled environment that encompasses essential factors such as temperature, carbon dioxide (CO2) levels, oxygen availability, and appropriate lighting conditions. Additionally, it is crucial to ensure the provision of vital nutrients to maximize output and productivity. Due to the demanding nature of a hydroponic farmer's schedule, it is necessary to minimize the amount of time dedicated to nutrient management, as well as pH and EC adjustments.MethodsIn order to determine and deliver the proper amount of vital nutrients, such as nitrogen, phosphorus, and potassium, based on the plant growth stage, we presented an automatic hydroponic nutrient estimator in this system. We noticed that the plant's nutrient consumption varies depending on its stage of growth according to plant psychology. Four peristaltic pumps with the necessary sensors are controlled by an Arduino board in the suggested system. Both filling and draining the water are done using each pump. To identify the plant stage, we apply the Plant Growth Stage Identification algorithm to encompass the seedling, vegetative, flowers, and fruit stages.ResultsThe experimental results reveal that the Growth Stage Identification algorithm obtains 97.5% accuracy for the first 5 weeks with 1,715 ppm of nutrition ingestion, identifying the vegetative state. The flowering stage is identified with 97.5% accuracy in the 6-9th week with 2,380 ppm of nutrition consumption, and the fruiting location is determined with 99.4% accuracy in the last 10-15th week with 2,730 ppm of nutrition consumption.

Project description:The implementation of bioreactor systems for the production of bacterial inoculants as biofertilizers has become very important in recent decades. However, it is essential to know the bacterial growth optimal conditions to optimize the production and efficiency of bioinoculants. The aim of this work was to identify the best nutriment and mixing conditions to improve the specific cell growth rates (µ) of two PGPB (plant growth-promoting bacteria) rhizobial strains at the bioreactor level. For this purpose, the strains Sinorhizobium mexicanum ITTG-R7T and Sinorhizobium chiapanecum ITTG-S70T were previously reactivated in a PY-Ca2+ (peptone casein, yeast extract, and calcium) culture medium. Afterward, a master cell bank (MCB) was made in order to maintain the viability and quality of the strains. The kinetic characterization of each bacterial strain was carried out in s shaken flask. Then, the effect of the carbon and nitrogen sources and mechanical agitation was evaluated through a factorial design and response surface methodology (RSM) for cell growth optimization, where µ was considered a response variable. The efficiency of biomass production was determined in a homemade bioreactor, taking into account the optimal conditions obtained during the experiment conducted at the shaken flask stage. In order to evaluate the biological quality of the product obtained in the bioreactor, the bacterial strains were inoculated in common bean (Phaseolus vulgaris var. Jamapa) plants under bioclimatic chamber conditions. The maximum cell growth rate in both PGPB strains was obtained using a Y-Ca2+ (yeast extract and calcium) medium and stirred at 200 and 300 rpm. Under these growth conditions, the Sinorhizobium strains exhibited a high nitrogen-fixing capacity, which had a significant (p < 0.05) impact on the growth of the test plants. The bioreactor system was found to be an efficient alternative for the large-scale production of PGPB rhizobial bacteria, which are intended for use as biofertilizers in agriculture.

Project description:Brassica napus, commonly known as canola, is an important oilseed crop in Canada contributing over $29.9 billion CAD to the Canadian economy annually. A major challenge facing Canadian canola is drought, which has become increasingly prevalent in recent years due to the changing climate. Research investigating novel agronomic techniques in mitigating drought is key to securing yields and sustainability in canola and other crops. One such technique is the use of bio-stimulant sprays to help offset biotic and abiotic stresses in plants through promoting stand establishment. Previous studies have shown that the application of seaweed extracts as bio-stimulant sprays to Brassicaceae has been successful in improving plant growth and development along with stress tolerance. However, this method has yet to be tested on canola. The organic nutrients that are waste products from processed seaweed help stimulate plant growth, yielding higher quality plants as a result. In association with Le Groupe Roullier, this study demonstrates that the Roullier extracts (RE) help increase plant growth characteristics and drought tolerance in canola when sprayed 3 times over a 3-week period. A high yielding but drought sensitive mutant of canola, d14 (developed through gene editing) was used for drought assays after 8 weeks of growth and where water was withheld for 6 days. Application of the REs prior to drought resulted in plants having enhanced survival rate and improved biomass retention indicating high drought tolerance. Subsequent RNA sequencing and gene ontological term analysis performed using RE treated plants in triplicates, revealed substantial levels of differential expression of growth-related genes along with stress-related genes. These REs elicited responses in plants that had previously only been achieved through gene editing and transgenic methodologies. Using bio-stimulant sprays provides a novel platform to promote beneficial agronomic traits, independent of genetic manipulation.

Project description:The dairy industry is facing challenges in balancing forage supply and crop production. Therefore, forage supply based on a farm land-saving approach should be developed to overcome the human-livestock competition on farmland. The objective of this study was to learn the potential impact of partially replacing oat hay with whole-plant hydroponic barley seedlings (HBS) produced via a land-saving hydroponic method on growth performance, digestibility, and rumen microbiota in Holstein dairy heifers. In total, 39 Holstein heifers were randomly divided into 13 blocks based on age and body weight for an 8-week experimental period. The heifers within each block were randomly allocated to one of three diets group: (1) 0% HBS and 16% oat hay (CON); (2) 4% HBS and 12% oat hay (25% HBS); and (3) 8% HBS and 8% oat hay (50% HBS). Compared to CON, feed intake, growth performance, and body N retention were similar to those in cows fed 25% HBS but lower in 50% HBS-fed animals (p < 0.05). Reduced digestibility (crude protein (CP) and organic matter (OM)) was observed in 50% HBS animals (p < 0.05). Compared to the control, the levels of Lachnospiraceae_XPB1014_group, Bacillus, and Colidextribacter were higher, but the levels of Sphaerochaeta and Ruminiclostridium were lower in 50% HBS animals (p < 0.05). Additionally, the digestibility of CP (p < 0.01, r = -0.61) and ether extract (EE) (p < 0.01, r = -0.58) was negatively correlated with Lachnospiraceae_XPB1014_group. The digestibility of OM (p = 0.01, r = -0.55), neutral detergent fiber (NDF) (p = 0.01, r = -0.56), acid detergent fiber (ADF) (p = 0.02, r = -0.52), and CP (p < 0.01, r = -0.61) was negatively correlated with Bacillus. The digestibility of NDF (p = 0.02, r = -0.52) and ADF (p = 0.03, r = -0.50) was negatively correlated with Colidextribacter. The digestibility of OM (p = 0.03, r = 0.50), NDF (p = 0.03, r = 0.50), and ADF (p = 0.03, r = 0.49) was positively correlated with Ruminiclostridium. The digestibility of OM (p = 0.04, r = 0.47), CP (p < 0.01, r = 0.58), and EE (p = 0.03, r = 0.49) was positively correlated with unclassified_f_Rikenellaceae. The digestibility of CP was positively correlated with Sphaerochaeta (p = 0.02, r = 0.53). In conclusion, the current study suggests that HBS could replace oat hay in a ratio-dependent manner. The reduced growth performance could be caused by lower feed intake and digestibility, which may be attributed to the alteration in the rumen's microbial population. Further exploration of the inhibiting factors of HBS would broaden the application of hydroponic feed in the future.
