Dengue Incidence and Aedes Vector Collections in Relation to COVID-19 Population Mobility Restrictions.

ABSTRACT: Contrary to expectation, dengue incidence decreased in many countries during the period when stringent population movement restrictions were imposed to combat COVID-19. Using a seasonal autoregressive integrated moving average model, we previously reported a 74% reduction in the predicted number of dengue cases from March 2020 to April 2021 in the whole of Sri Lanka, with reductions occurring in all 25 districts in the country. The reduction in dengue incidence was accompanied by an 87% reduction in larval collections of Aedes vectors in the northern city of Jaffna. It was proposed that movement restrictions led to reduced human contact and blood feeding by Aedes vectors, accompanied by decreased oviposition and vector densities, which were responsible for diminished dengue transmission. These findings are extended in the present study by investigating the relationship between dengue incidence, population movement restrictions, and vector larval collections between May 2021 and July 2022, when movement restrictions began to be lifted, with their complete removal in November 2021. The new findings further support our previous proposal that population movement restrictions imposed during the COVID-19 pandemic reduced dengue transmission primarily by influencing human-Aedes vector interaction dynamics.

Project description:BackgroundItaly was the first country after China to be severely affected by the COVID-19 pandemic, in early 2020. The country responded swiftly to the outbreak with a nationwide two-step lockdown, the first one light and the second one tight. By analyzing 2020 national mobile phone movements, we assessed how lockdown compliance influenced its efficacy.MethodsWe measured individual mobility during the first epidemic wave with mobile phone movements tracked through carrier networks, and related this mobility to daily new SARS-CoV-2 infections, hospital admissions, intensive care admissions and deaths attributed to COVID-19, taking into account reason for travel (work-related or not) and the means of transport.ResultsThe tight lockdown resulted in an 82% reduction in mobility for the entire country and was effective in swiftly curbing the outbreak as indicated by a shorter time-to-peak of all health outcomes, particularly for provinces with the highest mobility reductions and the most intense COVID-19 spread. Reduction of work-related mobility was accompanied by a nearly linear benefit in outbreak containment; work-unrelated movements had a similar effect only for restrictions exceeding 50%. Reduction in mobility by car and by airplane was nearly linearly associated with a decrease in most COVID-19 health outcomes, while for train travel reductions exceeding 55% had no additional beneficial effects. The absence of viral variants and vaccine availability during the study period eliminated confounding from these two sources.ConclusionsAdherence to the COVID-19 tight lockdown during the first wave in Italy was high and effective in curtailing the outbreak. Any work-related mobility reduction was effective, but only high reductions in work-unrelated mobility restrictions were effective. For train travel, there was a threshold above which no further benefit occurred. These findings could be particular to the spread of SARS-CoV-2, but might also apply to other communicable infections with comparable transmission dynamics.

Project description:BackgroundAedes aegypti is the primary vector of the four serotypes of dengue virus (DENV1-4), Chikungunya and yellow fever virus to humans. Previous population genetic studies have revealed a particular genetic structure among the vector populations in the Americas that suggests differences in the ability to transmit DENV. In Colombia, despite its high epidemiologic importance, the genetic population structure and the phylogeographic depiction of Ae. aegypti, as well as its relationship with the epidemiologic landscapes in cities with heterogeneous incidence levels, remains unknown. We conducted a spatiotemporal analysis with the aim of determining the genetic structure and phylogeography of Colombian populations of Ae. aegypti among cities with different eco-epidemiologic characteristics with regard to DENV.Methods/findingsMitochondrial cytochrome oxidase C subunit 1 (COI)--NADH dehydrogenase subunit 4 (ND4) genes were sequenced and analyzed from 341 adult mosquitoes collected during 2012 and 2013 in the Colombian cities of Bello, Riohacha and Villavicencio, which exhibit low, medium and high levels of incidence of DENV, respectively. The results demonstrated a low genetic differentiation over time and a high genetic structure between the cities due to changes in the frequency of two highly supported genetic groups. The phylogeographic analyses indicated that one group (associated with West African populations) was found in all the cities throughout the sampling while the second group (associated with East African populations) was found in all the samples from Bello and in only one sampling from Riohacha. Environmental factors such as the use of chemical insecticides showed a significant correlation with decreasing genetic diversity, indicating that environmental factors affect the population structure of Ae. aegypti across time and space in these cities.ConclusionsOur results suggest that two Ae. aegypti lineages are present in Colombia; one that is widespread and related to a West African conspecific, and a second that may have been recently introduced and is related to an East African conspecific. The first lineage can be found in cities showing a high incidence of dengue fever and the use of chemical insecticides, whereas the second is present in cities showing a low incidence of dengue fever where the use of chemical insecticides is not constant. This study helps to improve our knowledge of the population structure of Ae. aegypti involved in the diversity of dengue fever epidemiology in Colombia.

Project description:BackgroundThe COVID-19 pandemic has induced unprecedented reductions in human mobility and social contacts throughout the world. Because dengue virus (DENV) transmission is strongly driven by human mobility, behavioral changes associated with the pandemic have been hypothesized to impact dengue incidence. By discouraging human contact, COVID-19 control measures have also disrupted dengue vector control interventions, the most effective of which require entry into homes. We sought to investigate how and why dengue incidence could differ under a lockdown scenario with a proportion of the population sheltered at home.Methodology & principal findingsWe used an agent-based model with a realistic treatment of human mobility and vector control. We found that a lockdown in which 70% of the population sheltered at home and which occurred in a season when a new serotype invaded could lead to a small average increase in cumulative DENV infections of up to 10%, depending on the time of year lockdown occurred. Lockdown had a more pronounced effect on the spatial distribution of DENV infections, with higher incidence under lockdown in regions with higher mosquito abundance. Transmission was also more focused in homes following lockdown. The proportion of people infected in their own home rose from 54% under normal conditions to 66% under lockdown, and the household secondary attack rate rose from 0.109 to 0.128, a 17% increase. When we considered that lockdown measures could disrupt regular, city-wide vector control campaigns, the increase in incidence was more pronounced than with lockdown alone, especially if lockdown occurred at the optimal time for vector control.Conclusions & significanceOur results indicate that an unintended outcome of lockdown measures may be to adversely alter the epidemiology of dengue. This observation has important implications for an improved understanding of dengue epidemiology and effective application of dengue vector control. When coordinating public health responses during a syndemic, it is important to monitor multiple infections and understand that an intervention against one disease may exacerbate another.

Project description:Dengue is the most prevalent arboviral disease world-wide and its primary vector is the mosquito Aedes aegypti. The current lack of commercially-available vaccines makes control of vector populations the only effective strategy to prevent dengue transmission. Aedes aegypti geographic populations exhibit great variability in insecticide resistance and susceptibility to dengue infection. The characterization of single nucleotide polymorphisms (SNPs) as molecular markers to study quantitatively this variation is needed greatly because this species has a low abundance of microsatellite markers and limited known restriction fragments length polymorphisms (RFLPs) and single-strand conformation polymorphism (SSCP) markers.We used RNA-seq to characterize SNPs in three Ae. aegypti strains, including the Liverpool (LVP) strain, from which the current genome annotation is derived. We identified 131,764 unique genome locations with at least one alternative nucleotide to what is reported in the reference annotation. These comprised changes in both open-reading frames (ORFs) and untranslated regions (UTRs) of transcripts. An in depth-look at sequence variation in immunity genes revealed that those associated with autophagy, MD2-like receptors and Peptidoglycan Recognition Proteins had more sequence variation in their 3'UTRs than mutations associated with non-synonymous changes. This supports the conclusion that these genes had maintained their functional specificity while being adapted to different regulatory domains. In contrast, a number of peroxidases, serpins and Clip-domain serine proteases exhibited conservation of putative UTR regulatory sequences while displaying diversification of the ORFs. Transcriptome evidence also was found for ~2500 novel transcriptional units (NTUs) not annotated in the reference genome.The transcriptome-wide assessment of within and inter-strain polymorphisms in Ae. aegypti adds considerably to the number of molecular markers available for genetic studies in this mosquito. Additionally, data supporting NTU discovery emphasizes the need for continuous amendments of the reference genome annotation.

Project description:Diseases transmitted by female Aedes aegypti mosquitoes are public health issues in countries in the tropics and sub-tropics. As in other insects, A. aegypti females undergo behavioral and physiological changes upon mating that principally act to facilitate the production of progeny. The primary effectors of A. aegypti female post-mating responses are male-derived seminal proteins that are transferred to females during mating. Increased male age reduces ejaculate function in numerous taxa and alters seminal protein composition in Drosophila melanogaster, but the impacts of male age on female A. aegypti post-mating responses are unknown. Here, we used "old" (21-22 days old) and "young" (4-5 days old) A. aegypti males to assess the influence of male age on oviposition, fertility, and re-mating incidence in their mates. We also examined how age influenced paternity share in females initially mated to young or old males that subsequently re-mated with a transgenic male that transferred RFP-labeled sperm and whose progeny inherited a larval-expressed GFP marker. We found that increased male age had no effect on female fecundity or fertility but significantly impacted their ability to prevent re-mating in their mates-more than half (54.5%) of the females mated to an old male re-mated, compared to 24% of females initially mated to a young male. Polyandrous A. aegypti females displayed first male precedence regardless of the age of their initial mate. However, young males were better able to compete with rival male sperm, siring significantly more progeny (77%) compared to old males (64%). Young males had significantly more sperm in their seminal vesicles than old males at the time of mating, although males of both age groups transferred similar numbers of sperm to their mates. Our results suggest that male senescence differentially impacts the induction of some post-mating changes in A. aegypti females. As the effect of age may be further exacerbated in the field, age-related declines in male ability to induce sexual refractoriness have implications for A. aegypti population control programs that release adults into the environment.
