Project description:Vehicles are a major source of anthropogenic emissions of carbon monoxide (CO), nitrogen oxides (NOx), and black carbon (BC). CO and NOx are known to be harmful to human health and contribute to ozone formation, while BC absorbs solar radiation that contributes to global warming and also has negative impacts on human health and visibility. Travel restrictions implemented during the COVID-19 pandemic provide researchers the opportunity to study the impact of large, on-road traffic reductions on local air quality. Traffic counts collected along Interstate-95, a major eight-lane highway in Maryland (US), reveal a 60% decrease in passenger car totals and an 8.6% (combination-unit) and 21% (single-unit) decrease in truck traffic counts in April 2020 relative to prior Aprils. The decrease in total on-road vehicles led to the near-elimination in stop-and-go traffic and a 14% increase in the mean vehicle speed during April 2020. Ambient near-road (NR) BC, CO, NOx, and carbon dioxide (CO2) measurements were used to determine vehicular emission ratios (ΔBC/ΔCO, ΔBC/ΔCO2, ΔNOx/ΔCO, ΔNOx/ΔCO2, and ΔCO/ΔCO2), with each ratio defined as the slope value of a linear regression performed on the concentrations of two pollutants within an hour. A decrease of up to a factor of two in ΔBC/ΔCO, ΔBC/ΔCO2, ΔNOx/ΔCO2, and in the fraction of on-road diesel vehicles from weekdays to weekends shows diesel vehicles to be the dominant source of BC and NOx emissions at this NR site. We estimate up to a 70% reduction in BC emissions in April 2020 compared to earlier years, and attribute much of this to lower diesel BC emissions resulting from improvements in traffic flow and fewer instances of acceleration and braking. Future efforts to reduce vehicular BC emissions should focus on improving traffic flow or turbocharger lag within diesel engines. Inferred BC emissions from the NR site also depend on ambient temperature, with an increase of 54% in ΔBC/ΔCO from -5 to 20 °C during the cold season, similar to previous studies that reported increasing BC emissions with rising temperature. The default setting of MOVES3, the current version of the mobile emission model used by the US EPA, does not adjust hot-running BC emissions for ambient temperature. Future work will focus on improving the accuracy of mobile emissions in air quality modeling by incorporating the effects of temperature and traffic flow in the system used to generate mobile emissions input for commonly used air quality models.
Project description:Accurately characterizing human exposures to traffic-related air pollutants (TRAPs) is critical to public health protection. However, quantifying exposure to this single source is challenging, given its extremely heterogeneous chemical composition. Efforts using single-species tracers of TRAP are, thus, lacking in their ability to accurately reflect exposures to this complex mixture. There have been recent discussions centered on adopting a multipollutant perspective for sources with many emitted pollutants to maximize the benefits of control expenditures as well as to minimize population and ecosystem exposure. As part of a larger study aimed to assess a complete emission-to-exposure pathway of primary traffic pollution and understand exposure of individuals in the near-road environment, an intensive field campaign measured TRAPs and related data (e.g., meteorology, traffic counts, and regional air pollutant levels) in Atlanta along one of the busiest highway corridors in the US. Given the dynamic nature of the near-road environment, a multipollutant exposure metric, the Integrated Mobile Source Indicator (IMSI), which was generated based on emissions-based ratios, was calculated and compared to traditional single-species methods for assessing exposure to mobile source emissions. The current analysis examined how both traditional and non-traditional metrics vary spatially and temporally in the near-road environment, how they compare with each other, and whether they have the potential to offer more accurate means of assigning exposures to primary traffic emissions. The results indicate that compared to the traditional single pollutant specie, the multipollutant IMSI metric provided a more spatially stable method for assessing exposure, though variations occurred based on location with varying results among the six sites within a kilometer of the highway.
Project description:Air pollution health studies often use outdoor concentrations as exposure surrogates. Failure to account for variability of residential infiltration of outdoor pollutants can induce exposure errors and lead to bias and incorrect confidence intervals in health effect estimates. The residential air exchange rate (AER), which is the rate of exchange of indoor air with outdoor air, is an important determinant for house-to-house (spatial) and temporal variations of air pollution infiltration. Our goal was to evaluate and apply mechanistic models to predict AERs for 213 homes in the Near-Road Exposures and Effects of Urban Air Pollutants Study (NEXUS), a cohort study of traffic-related air pollution exposures and respiratory effects in asthmatic children living near major roads in Detroit, Michigan. We used a previously developed model (LBL), which predicts AER from meteorology and questionnaire data on building characteristics related to air leakage, and an extended version of this model (LBLX) that includes natural ventilation from open windows. As a critical and novel aspect of our AER modeling approach, we performed a cross validation, which included both parameter estimation (i.e., model calibration) and model evaluation, based on daily AER measurements from a subset of 24 study homes on five consecutive days during two seasons. The measured AER varied between 0.09 and 3.48 h(-1) with a median of 0.64 h(-1). For the individual model-predicted and measured AER, the median absolute difference was 29% (0.19 h‑1) for both the LBL and LBLX models. The LBL and LBLX models predicted 59% and 61% of the variance in the AER, respectively. Daily AER predictions for all 213 homes during the three year study (2010-2012) showed considerable house-to-house variations from building leakage differences, and temporal variations from outdoor temperature and wind speed fluctuations. Using this novel approach, NEXUS will be one of the first epidemiology studies to apply calibrated and home-specific AER models, and to include the spatial and temporal variations of AER for over 200 individual homes across multiple years into an exposure assessment in support of improving risk estimates.
Project description:Exposure to vehicular emissions has been linked to numerous adverse health effects. In response to the arising concerns, near-road monitoring is conducted to better characterize the impact of mobile source emissions on air quality and exposure in the near-road environment. An intensive measurement campaign measured traffic-related air pollutants (TRAPs) and related data (e.g., meteorology, traffic, regional air pollutant levels) in Atlanta, along one of the busiest highway corridors in the US. Given the complexity of the near-road environment, the study aimed to compare two near-road monitors, located in close proximity to each other, to assess how observed similarities and differences between measurements at these two sites inform the siting of other near-road monitoring stations. TRAP measurements, including carbon monoxide (CO) and nitrogen dioxide (NO2), are analyzed at two roadside monitors in Atlanta, GA located within 325m of each other. Both meteorological and traffic conditions were monitored to assess the temporal impact of these factors on traffic-related pollutant concentrations. The meteorological factors drove the diurnal variability of primary pollutant concentration more than traffic count. In spite of their proximity, while the CO and NO2 concentrations were correlated with similar diurnal variations, pollutant concentrations at the two closely sited monitors differed, likely due to the differences in the siting characteristics reducing the dispersion of the primary emissions out of the near-road environment. Overall, the near-road TRAP concentrations at all sites were not as elevated as seen in prior studies, supporting that decreased vehicle emissions have led to significant reductions in TRAP levels, even along major interstates. Further, the differences in the observed levels show that use of single near-road observations will not capture pollutant levels representative of the local near-road environment and that additional approaches (e.g., air quality models) are needed to characterize exposures.
Project description:Epidemiological and toxicological studies continue to demonstrate correlative and causal relationships between exposure to traffic-related air pollution and various metrics of adverse pulmonary, cardiovascular, and neurological health effects. The key challenge for in vivo studies is replicating real-world, near-roadway exposure dynamics in laboratory animal models that mimic true human exposures. The advantage of animal models is the accelerated time scales to show statistically significant physiological and/or behavioral response. This work describes a novel exposure facility adjacent to a major freeway tunnel system that provides a platform for real-time chronic exposure studies. The primary conclusion is that particulate matter (PM) concentrations at this facility are routinely well below the National Ambient Air Quality Standards (NAAQS), but studies completed to date still demonstrate significant neurological and cardiovascular effects. Internal combustion engines produce large numbers of ultrafine particles that contribute negligible mass to the atmosphere relative to NAAQS regulated PM2.5 but have high surface area and mobility in the body. It is posited here that current federal and state air quality standards are thus insufficient to fully protect human health, most notably the developing and aging brain, due to regulatory gaps for ultrafine particles.
Project description:Roadside vegetation has been shown to impact downwind, near-road air quality, with some studies identifying reductions in air pollution concentrations and others indicating increases in pollutant levels when vegetation is present. These widely contradictory results have resulted in confusion regarding the capability of vegetative barriers to mitigate near-road air pollution, which numerous studies have associated with significant adverse human health effects. Roadside vegetation studies have investigated the impact of many different types and conditions of vegetation barriers and urban forests, including preserved, existing vegetation stands usually consisting of mixtures of trees and shrubs or plantings of individual trees. A study was conducted along a highway with differing vegetation characteristics to identify if and how the changing characteristics affected downwind air quality. The results indicated that roadside vegetation needed to be of sufficient height, thickness, and coverage to achieve downwind air pollutant reductions. A vegetation stand which was highly porous and contained large gaps within the stand structure had increased downwind pollutant concentrations. These field study results were consistent with other studies that the roadside vegetation could lead to reductions in average, downwind pollutant concentrations by as much as 50% when this vegetation was thick with no gaps or openings. However, the presence of highly porous vegetation with gaps resulted in similar or sometimes higher concentrations than measured in a clearing with no vegetation. The combination of air quality and meteorological measurements indicated that the vegetation affects downwind pollutant concentrations through attenuation of meteorological and vehicle-induced turbulence as air passes through the vegetation, enhanced mixing as portions of the traffic pollution plume are blocked and forced over the vegetation, and through particulate deposition onto leaf and branch surfaces. Computational fluid dynamic modeling highlighted that density of the vegetation barrier affects pollutant levels, with a leaf area density of 3.0 m2 m-3 or higher needed to ensure downwind pollutant reductions for airborne particulate matter. These results show that roadside bushes and trees can be preserved or planted along highways and other localized pollution sources to mitigate air quality and human health impacts near the source if the planting adheres to important characteristics of height, thickness, and density with full coverage from the ground to the top of the canopy. The results also highlight the importance of planting denser vegetation and maintaining the integrity and structure of these vegetation barriers to achieve pollution reductions and not contribute to unintended increases in downwind air pollutant concentrations.
Project description:BackgroundEvidence linking long-term exposure to particulate air pollution to blood pressure (BP) in high-income countries may not be transportable to low- and middle-income countries. We examined cross-sectional associations between ambient fine particulate matter (PM2.5) and black carbon (BC) with BP (systolic [SBP] and diastolic [DBP]) and prevalent hypertension in adults from 28 peri-urban villages near Hyderabad, India.MethodsWe studied 5531 participants from the Andhra Pradesh Children and Parents Study (18-84 years, 54% men). We measured BP (2010-2012) in the right arm and defined hypertension as SBP ≥130 mmHg and/or DBP ≥80 mmHg. We used land-use regression models to estimate annual average PM2.5 and BC at participant's residence. We applied linear and logistic nested mixed-effect models stratified by sex and adjusted by cooking fuel type to estimate associations between within-village PM2.5 or BC and health.ResultsMean (SD) PM2.5 was 33 µg/m (2.7) and BC was 2.5 µg/m (0.23). In women, a 1 µg/m increase in PM2.5 was associated with 1.4 mmHg higher SBP (95% confidence interval [CI]: 0.12, 2.7), 0.87 mmHg higher DBP (95% CI: -0.18, 1.9), and 4% higher odds of hypertension (95% CI: 0%, 9%). In men, associations with SBP (0.52 mmHg; 95% CI: -0.82, 1.8), DBP (0.41 mmHg; 95% CI: -0.69, 1.5), and hypertension (2% higher odds; 95% CI: -2%, 6%) were weaker. No associations were observed with BC.ConclusionWe observed a positive association between ambient PM2.5 and BP and hypertension in women. Longitudinal studies in this region are needed to corroborate our findings.
Project description:In recent years, ozone (O3) concentrations in the southeastern coastal areas of China have shown a gradual upward trend. As precursors and intermediates in the formation of O3, carbonyl compounds play key roles in the atmospheric photochemical oxidation cycle. To explore the main pollution characteristics of carbonyl compounds in a typical coastal city in southeast China, ambient samples were collected in Fuzhou (the provincial capital of Fujian province, located on the southeast coast of China) and analyzed using high-performance liquid chromatography with ultraviolet detection. The study was continuously carried out at an urban site (Jinjishan) and a suburban site (Gushan) in Fuzhou from May 8 to 20, 2018. The total concentration of 16 carbonyl compounds at the urban site was 15.45 ± 11.18 ppbv, and the total concentration at the suburban site was 17.57 ± 12.77 ppbv. Formaldehyde (HCHO), acetaldehyde, and acetone were the main species detected in the samples, and acetone had the highest concentration among the species detected. The suburban site had a higher formaldehyde/acetaldehyde ratio and lower acetaldehyde/propionaldehyde ratio than the urban site, implying that biogenic sources potentially contributed to the carbonyl compound concentrations at the suburban site. The results of an observation-based model showed that anthropogenic hydrocarbons promoted HCHO production on May 17 at the urban site. Compared to biogenic emissions, anthropogenic activity is a more important source of carbonyl compounds.
Project description:Chimeric antigen receptors are genetically encoded artificial fusion molecules that can re-program the specificity of peripheral blood polyclonal T-cells against a selected cell surface target. Unparallelled clinical efficacy has recently been demonstrated using this approach to treat patients with refractory B-cell malignancy. However, the approach is technically challenging and can elicit severe toxicity in patients. Moreover, solid tumours have largely proven refractory to this approach. In this review, we describe the important structural features of CARs and how this may influence function. Emerging clinical experience is summarized in both solid tumours and haematological malignancies. Finally, we consider the particular challenges imposed by solid tumours to the successful development of CAR T-cell immunotherapy, together with a number of innovative strategies that have been developed in an effort to reverse the balance in favour of therapeutic benefit.
Project description:As part of the United States Environmental Protection Agency's 2010 Nitrogen Dioxide (NO2) National Ambient Air Quality Standards (NAAQS) review, a national network of near-road sites was established to characterize pollutant behavior, interaction, and dispersion in the ambient near-road environment. Using spatial interpolation to estimate the near-road concentration increments of NO2 and particulate matter with an aerodynamic diameter of 2.5 μm and less (PM2.5) relative to nearby non-near-road monitors, we found that the 2013-2018 national average increment is 6.9 ppb and 1.0 μg m-3 for NO2 and PM2.5, respectively. Analyses of the hourly near-road NO2, nitric oxide (NO), and PM2.5 increments showed distinct diurnal cycles; the NO2 increment peaks at ∼9 ppb during the early afternoon (2-4 pm local time) while the NO and PM2.5 increments peak during the morning rush hour (5-8 am local time) at 25 ppb and 1.8 μg m-3 for NO and PM2.5, respectively. Although long-term trends are not yet available for this network of sites, a similar analysis of the NO2 and PM2.5 increment at a quasi-near-road site outside of the official network in Elizabeth, NJ showed gradual decreases in the increment over time since the mid-2000s.
