Project description:We integrate spectroscopic optical coherence tomography (SOCT) with stimulated Raman scattering (SRS) to enable simultaneously multiplexed spatial and spectral imaging with sensitivity to many endogenous biochemical species that play an important role in biology and medicine. The combined approach, termed SRS-SOCT, overcomes the limitations of each individual method. Ultimately, SRS-SOCT has the potential to achieve fast, volumetric, and highly sensitive label-free molecular imaging. We demonstrate the approach by imaging excised human adipose tissue and detecting the lipids' Raman signatures in the high-wavenumber region.

Project description:AbstractA stringent limitation in many optoelectronic devices, such as solar cells and light emitting diodes, is the intrinsic need for a transparent electrode. Uniting relevant aspects, indium tin oxide (ITO) is often the material of choice, however, alternatives are sought and being in particular found in conductive polymers. In this work, we present a novel doping strategy to arrive at highly conducting polymeric material based on poly-3,4-ethylenedioxythiophene (PEDOT). Based on commercial high conductivity PEDOT:PSS (Clevios PH 1000), and a post processing with aqueous triflic acid delivers a material that is both transparent and of low resistivity (5.23 × 10-4 Ω cm). Furthermore, this material retains its conductive character over a large temperature range, indicating metallic behaviour. This is further supported by positive magnetoconductance effects at low temperatures (1.8-10 K) and extended mean free paths of the conduction electrons are observed-evidencing for a metallic state in this polymer.Graphical abstract

Project description:This work describes a detailed quantitative interaction study between the novel plastidial chaperone receptor OEP61 and isoforms of the chaperone types Hsp70 and Hsp90 using the optical method of total internal reflection ellipsometry (TIRE). The receptor OEP61 was electrostatically immobilized on a gold surface via an intermediate layer of polycations. The TIRE measurements allowed the evaluation of thickness changes in the adsorbed molecular layers as a result of chaperone binding to receptor proteins. Hsp70 chaperone isoforms but not Hsp90 were shown to be capable of binding OEP61. Dynamic TIRE measurements were carried out to evaluate the affinity constants of the above reactions and resulted in clear discrimination between specific and nonspecific binding of chaperones as well as differences in binding properties between the highly similar Hsp70 isoforms.

Project description:Hydrogels of conducting polymers, particularly poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS), provide a promising electrical interface with biological tissues for sensing and stimulation, owing to their favorable electrical and mechanical properties. While existing methods mostly blend PEDOT:PSS with other compositions such as non-conductive polymers, the blending can compromise resultant hydrogels' mechanical and/or electrical properties. Here, we show that designing interconnected networks of PEDOT:PSS nanofibrils via a simple method can yield high-performance pure PEDOT:PSS hydrogels. The method involves mixing volatile additive dimethyl sulfoxide (DMSO) into aqueous PEDOT:PSS solutions followed by controlled dry-annealing and rehydration. The resultant hydrogels exhibit a set of properties highly desirable for bioelectronic applications, including high electrical conductivity (~20 S cm-1 in PBS, ~40 S cm-1 in deionized water), high stretchability (> 35% strain), low Young's modulus (~2 MPa), superior mechanical, electrical and electrochemical stability, and tunable isotropic/anisotropic swelling in wet physiological environments.

Project description:Spectroscopic ellipsometry is a means of investigating optical and dielectric material responses. Conventional spectroscopic ellipsometry is subject to trade-offs between spectral accuracy, resolution, and measurement time. Polarization modulation has afforded poor performance because of its sensitivity to mechanical vibrational noise, thermal instability, and polarization-wavelength dependency. We combine spectroscopic ellipsometry with dual-comb spectroscopy, namely, dual-comb spectroscopic ellipsometry. Dual-comb spectroscopic ellipsometry (DCSE). DCSE directly and simultaneously obtains the ellipsometric parameters of the amplitude ratio and phase difference between s-polarized and p-polarized light signals with ultra-high spectral resolution and no polarization modulation, beyond the conventional limit. Ellipsometric evaluation without polarization modulation also enhances the stability and robustness of the system. In this study, we construct a polarization-modulation-free DCSE system with a spectral resolution of up to 1.2 × 10-5 nm throughout the spectral range of 1514-1595 nm and achieved an accuracy of 38.4 nm and a precision of 3.3 nm in the measurement of thin-film samples.Spectroscopic ellipsometry is an established technique to characterize the optical properties of a material. Here, Minamikawa et al. combine the method with dual-comb spectroscopy, which allows them to obtain ellipsometric parameters including the phase difference between s-polarized and p-polarized light.

Project description:The conducting polymer of poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate) (PEDOT:PSS) is one of the most important and intensively investigated organic conducting materials. The PEDOT:PSS water dispersions with various concentrations of poly (4-styrenesulfonic acid) solution (PSSAS) were synthesized by 3,4-ethylenedioxythiophene (EDOT) in the presence of water. The fabrication of the solvent-treated PEDOT:PSS films through spin coating and solvent treatment processes was achieved with a solvent of pure water mixed with acetone (or MeOH, EtOH) in a ratio of 50:50. Moreover, both the organic solvent and water have synergetic effects while the PSS and PEDOT-attached PSS segments will form a coil-like and a linear (or extended-coil) structure, respectively. That may induce a stacking of the linear and planar PEDOT-attached PSS segments, which favors the formation of a crystalline phase. Finally, the maximum electrical conductivity of the PEDOT:PSS thin films with solvent treatment was investigated by means of X-ray diffraction (XRD) patterns and scanning electron microscope (SEM) images. Furthermore, we aimed to explain the synergetic effects of phase separation of the PEDOT:PSS thin films by both the organic solvent and water.

Project description:We have for the first time found that completely insulating Halloysite nanotubes (HNTs) significantly enhance electrical conductivity of PEDOT/PSS films by simply mixing. Based on this accident finding we have created highly porous and conductive PEDOT/PSS films hybridized with the HNTs. Through further optimization of the mixing condition we have obtained flexible and conductive hybrid films with high specific surface area. Based on experimental evidences we proposed a plausible mechanism of the phenomenon where the PEDOT/PSS colloidal particle with particle size of several tens nanometers well pack at the nano-channels into well-ordered structures of PEDOT/PSS particles, which show conductivity as higher as several order of magnitude than that of PEDOT/PSS particles in outside of the HNTs.

Project description:A highly sensitive multilayer interlaced silver (Ag) nanosheet (MISN) film was prepared on a PPy@PEDOT:PSS film via an electrodeposition method for surface-enhanced Raman scattering (SERS) applications. After the PPy@PEDOT:PSS film was pretreated with ascorbic acid solution, many sparse Ag nanoparticles (NPs) could be directly reduced on the surface of the PPy@PEDOT:PSS film in AgNO3 solution. Then, the MISN film was directionally grown along the surface of sparse Ag NPs by using an electrochemical galvanostatic method to form a Ag/PPy@PEDOT:PSS film for a SERS substrate. The results indicated that with the increase in electrodeposition time, the density of Ag nanosheets was also increased for boosting the SERS effect. Accordingly, owing to the directional growth of Ag NPs, the increase in the length-width ratio of single Ag nanosheets would further promote the SERS signal of the substrate. Moreover, the maximum enhancement factor of the SERS substrate could reach to 12,478, and the minimum limit of detection of melamine solution was down to 5.42 ng/mL. The SERS sensitivity of the Ag nanosheet film reached 100.65. This method of preparing the SERS substrate provides a novel and robust strategy for the low-cost and high-sensitivity detection in biomedicine, drugs, and food.

Project description:Molecular vibrational spectroscopy is widely used in various sensing and imaging applications, providing intrinsic information at the molecular level. Nonlinear optical interactions using ultrashort laser pulses facilitate the selective coherent excitation of molecular vibrational modes by focusing energy into specific molecular bonds, boosting the signal level for multiple orders of magnitude. The dephasing of such coherence, which is susceptible to the local molecular environment, however, is often neglected. The unique capability of vibrational dephasing dynamics to serve as a unique probe for complex molecular interactions and the effect of local nano- and microenvironments are beyond the reach of conventional, intensity-based spectroscopy. Here, we developed a novel multiorder coherent Raman spectroscopy platform with a special focus on the temporal evolution of molecular vibrational dephasing, termed as time-resolved coherent Raman scattering (T-CRS) spectroscopy. By utilizing a high dynamic range detection, molecular vibrational dynamics and the environmental effects are demonstrated with multidimensional spectroscopic sensing, which promises a new range of applications in biology, materials, and chemical sciences.

Project description:Introduction/aimsElectromyography (EMG) remains a key component of the diagnostic work-up for suspected neuromuscular disease, but it does not provide insight into the molecular composition of muscle which can provide diagnostic information. Raman spectroscopy is an emerging neuromuscular biomarker capable of generating highly specific, molecular fingerprints of tissue. Here, we present "optical EMG," a combination of EMG and Raman spectroscopy, achieved using a single needle.MethodsAn optical EMG needle was created to collect electrophysiological and Raman spectroscopic data during a single insertion. We tested functionality with in vivo recordings in the SOD1G93A mouse model of amyotrophic lateral sclerosis (ALS), using both transgenic (n = 10) and non-transgenic (NTg, n = 7) mice. Under anesthesia, compound muscle action potentials (CMAPs), spontaneous EMG activity and Raman spectra were recorded from both gastrocnemius muscles with the optical EMG needle. Standard concentric EMG needle recordings were also undertaken. Electrophysiological data were analyzed with standard univariate statistics, Raman data with both univariate and multivariate analyses.ResultsA significant difference in CMAP amplitude was observed between SOD1G93A and NTg mice with optical EMG and standard concentric needles (p = .015 and p = .011, respectively). Spontaneous EMG activity (positive sharp waves) was detected in transgenic SOD1G93A mice only. Raman spectra demonstrated peaks associated with key muscle components. Significant differences in molecular composition between SOD1G93A and NTg muscle were identified through the Raman spectra.DiscussionOptical EMG can provide standard electrophysiological data and molecular Raman data during a single needle insertion and represents a potential biomarker for neuromuscular disease.
