Computational and neural mechanisms of statistical pain learning.

ABSTRACT: Pain invariably changes over time. These fluctuations contain statistical regularities which, in theory, could be learned by the brain to generate expectations and control responses. We demonstrate that humans learn to extract these regularities and explicitly predict the likelihood of forthcoming pain intensities in a manner consistent with optimal Bayesian inference with dynamic update of beliefs. Healthy participants received probabilistic, volatile sequences of low and high-intensity electrical stimuli to the hand during brain fMRI. The inferred frequency of pain correlated with activity in sensorimotor cortical regions and dorsal striatum, whereas the uncertainty of these inferences was encoded in the right superior parietal cortex. Unexpected changes in stimulus frequencies drove the update of internal models by engaging premotor, prefrontal and posterior parietal regions. This study extends our understanding of sensory processing of pain to include the generation of Bayesian internal models of the temporal statistics of pain.

The Neural Mechanisms of Social Learning from Fleeting Experience with Pain.

Project description:Social learning is critical for humans to adapt and cope with rapidly changing surroundings. Although, neuroscience has focused on associative learning and pain empathy, the neural mechanisms of social learning through fleeting pain remains to be determined. This functional MRI study included three participant groups, to investigate how the neuro-hemodynamic response and subjective evaluation in response to the observation of hand actions were modulated by first-hand experience (FH), as well as indirect experience through social-observational (SO), and verbal-informed (VI) learning from fleeting pain. The results indicated, that these three learning groups share the common neuro-hemodynamic activations in the brain regions implicated in emotional awareness, memory, mentalizing, perspective taking, and emotional regulation. The anterior insular cortex (AIC) was commonly activated during these learning procedures. The amygdala was only activated by the FH. Dynamic causal modeling further indicated, that the SO and VI learning exhibited weaker connectivity strength from the AIC to superior frontal gyrus than did the FH. These findings demonstrate, that social learning elicits distinct neural responses from associative learning. The ontogeny of human empathy could be better understood with social learning from fleeting experience with pain.

Reinforcement learning: Computational theory and biological mechanisms.

Project description:Reinforcement learning is a computational framework for an active agent to learn behaviors on the basis of a scalar reward signal. The agent can be an animal, a human, or an artificial system such as a robot or a computer program. The reward can be food, water, money, or whatever measure of the performance of the agent. The theory of reinforcement learning, which was developed in an artificial intelligence community with intuitions from animal learning theory, is now giving a coherent account on the function of the basal ganglia. It now serves as the "common language" in which biologists, engineers, and social scientists can exchange their problems and findings. This article reviews the basic theoretical framework of reinforcement learning and discusses its recent and future contributions toward the understanding of animal behaviors and human decision making.

Tonality Tunes the Statistical Characteristics in Music: Computational Approaches on Statistical Learning.

Project description:Statistical learning is a learning mechanism based on transition probability in sequences such as music and language. Recent computational and neurophysiological studies suggest that the statistical learning contributes to production, action, and musical creativity as well as prediction and perception. The present study investigated how statistical structure interacts with tonalities in music based on various-order statistical models. To verify this in all 24 major and minor keys, the transition probabilities of the sequences containing the highest pitches in Bach's Well-Tempered Clavier, which is a collection of two series (No. 1 and No. 2) of preludes and fugues in all of the 24 major and minor keys, were calculated based on nth-order Markov models. The transition probabilities of each sequence were compared among tonalities (major and minor), two series (No. 1 and No. 2), and music types (prelude and fugue). The differences in statistical characteristics between major and minor keys were detected in lower- but not higher-order models. The results also showed that statistical knowledge in music might be modulated by tonalities and composition periods. Furthermore, the principal component analysis detected the shared components of related keys, suggesting that the tonalities modulate statistical characteristics in music. The present study may suggest that there are at least two types of statistical knowledge in music that are interdependent on and independent of tonality, respectively.

Neural and computational mechanisms of postponed decisions.

Project description:We consider the mechanisms that enable decisions to be postponed for a period after the evidence has been provided. Using an information theoretic approach, we show that information about the forthcoming action becomes available from the activity of neurons in the medial premotor cortex in a sequential decision-making task after the second stimulus is applied, providing the information for a decision about whether the first or second stimulus is higher in vibrotactile frequency. The information then decays in a 3-s delay period in which the neuronal activity declines before the behavioral response can be made. The information then increases again when the behavioral response is required. We model this neuronal activity using an attractor decision-making network in which information reflecting the decision is maintained at a low level during the delay period, and is then selectively restored by a nonspecific input when the response is required. One mechanism for the short-term memory is synaptic facilitation, which can implement a mechanism for postponed decisions that can be correct even when there is little neuronal firing during the delay period before the postponed decision. Another mechanism is graded firing rates by different neurons in the delay period, with restoration by the nonspecific input of the low-rate activity from the higher-rate neurons still firing in the delay period. These mechanisms can account for the decision making and for the memory of the decision before a response can be made, which are evident in the activity of neurons in the medial premotor cortex.

Neural mechanisms of observational learning.

Project description:Individuals can learn by interacting with the environment and experiencing a difference between predicted and obtained outcomes (prediction error). However, many species also learn by observing the actions and outcomes of others. In contrast to individual learning, observational learning cannot be based on directly experienced outcome prediction errors. Accordingly, the behavioral and neural mechanisms of learning through observation remain elusive. Here we propose that human observational learning can be explained by two previously uncharacterized forms of prediction error, observational action prediction errors (the actual minus the predicted choice of others) and observational outcome prediction errors (the actual minus predicted outcome received by others). In a functional MRI experiment, we found that brain activity in the dorsolateral prefrontal cortex and the ventromedial prefrontal cortex respectively corresponded to these two distinct observational learning signals.

When forgetting fosters learning: A neural network model for statistical learning.

Project description:Learning often requires splitting continuous signals into recurring units, such as the discrete words constituting fluent speech; these units then need to be encoded in memory. A prominent candidate mechanism involves statistical learning of co-occurrence statistics like transitional probabilities (TPs), reflecting the idea that items from the same unit (e.g., syllables within a word) predict each other better than items from different units. TP computations are surprisingly flexible and sophisticated. Humans are sensitive to forward and backward TPs, compute TPs between adjacent items and longer-distance items, and even recognize TPs in novel units. We explain these hallmarks of statistical learning with a simple model with tunable, Hebbian excitatory connections and inhibitory interactions controlling the overall activation. With weak forgetting, activations are long-lasting, yielding associations among all items; with strong forgetting, no associations ensue as activations do not outlast stimuli; with intermediate forgetting, the network reproduces the hallmarks above. Forgetting thus is a key determinant of these sophisticated learning abilities. Further, in line with earlier dissociations between statistical learning and memory encoding, our model reproduces the hallmarks of statistical learning in the absence of a memory store in which items could be placed.

Computational Protein Design with Deep Learning Neural Networks.

Project description:Computational protein design has a wide variety of applications. Despite its remarkable success, designing a protein for a given structure and function is still a challenging task. On the other hand, the number of solved protein structures is rapidly increasing while the number of unique protein folds has reached a steady number, suggesting more structural information is being accumulated on each fold. Deep learning neural network is a powerful method to learn such big data set and has shown superior performance in many machine learning fields. In this study, we applied the deep learning neural network approach to computational protein design for predicting the probability of 20 natural amino acids on each residue in a protein. A large set of protein structures was collected and a multi-layer neural network was constructed. A number of structural properties were extracted as input features and the best network achieved an accuracy of 38.3%. Using the network output as residue type restraints improves the average sequence identity in designing three natural proteins using Rosetta. Moreover, the predictions from our network show ~3% higher sequence identity than a previous method. Results from this study may benefit further development of computational protein design methods.

Biological Mechanisms for Learning: A Computational Model of Olfactory Learning in the <i>Manduca sexta</i> Moth, With Applications to Neural Nets.

Project description:The insect olfactory system, which includes the antennal lobe (AL), mushroom body (MB), and ancillary structures, is a relatively simple neural system capable of learning. Its structural features, which are widespread in biological neural systems, process olfactory stimuli through a cascade of networks where large dimension shifts occur from stage to stage and where sparsity and randomness play a critical role in coding. Learning is partly enabled by a neuromodulatory reward mechanism of octopamine stimulation of the AL, whose increased activity induces synaptic weight updates in the MB through Hebbian plasticity. Enforced sparsity in the MB focuses Hebbian growth on neurons that are the most important for the representation of the learned odor. Based upon current biophysical knowledge, we have constructed an end-to-end computational firing-rate model of the Manduca sexta moth olfactory system which includes the interaction of the AL and MB under octopamine stimulation. Our model is able to robustly learn new odors, and neural firing rates in our simulations match the statistical features of in vivo firing rate data. From a biological perspective, the model provides a valuable tool for examining the role of neuromodulators, like octopamine, in learning, and gives insight into critical interactions between sparsity, Hebbian growth, and stimulation during learning. Our simulations also inform predictions about structural details of the olfactory system that are not currently well-characterized. From a machine learning perspective, the model yields bio-inspired mechanisms that are potentially useful in constructing neural nets for rapid learning from very few samples. These mechanisms include high-noise layers, sparse layers as noise filters, and a biologically-plausible optimization method to train the network based on octopamine stimulation, sparse layers, and Hebbian growth.

Neural Correlates of Morphology Acquisition through a Statistical Learning Paradigm.

Project description:The neural basis of statistical learning as it occurs over time was explored with stimuli drawn from a natural language (Russian nouns). The input reflected the "rules" for marking categories of gendered nouns, without making participants explicitly aware of the nature of what they were to learn. Participants were scanned while listening to a series of gender-marked nouns during four sequential scans, and were tested for their learning immediately after each scan. Although participants were not told the nature of the learning task, they exhibited learning after their initial exposure to the stimuli. Independent component analysis of the brain data revealed five task-related sub-networks. Unlike prior statistical learning studies of word segmentation, this morphological learning task robustly activated the inferior frontal gyrus during the learning period. This region was represented in multiple independent components, suggesting it functions as a network hub for this type of learning. Moreover, the results suggest that subnetworks activated by statistical learning are driven by the nature of the input, rather than reflecting a general statistical learning system.

Neural processes underlying statistical learning for speech segmentation in dogs.

Project description:To learn words, humans extract statistical regularities from speech. Multiple species use statistical learning also to process speech, but the neural underpinnings of speech segmentation in non-humans remain largely unknown. Here, we investigated computational and neural markers of speech segmentation in dogs, a phylogenetically distant mammal that efficiently navigates humans' social and linguistic environment. Using electroencephalography (EEG), we compared event-related responses (ERPs) for artificial words previously presented in a continuous speech stream with different distributional statistics. Results revealed an early effect (220-470 ms) of transitional probability and a late component (590-790 ms) modulated by both word frequency and transitional probability. Using fMRI, we searched for brain regions sensitive to statistical regularities in speech. Structured speech elicited lower activity in the basal ganglia, a region involved in sequence learning, and repetition enhancement in the auditory cortex. Speech segmentation in dogs, similar to that of humans, involves complex computations, engaging both domain-general and modality-specific brain areas. VIDEO ABSTRACT.
