Decision formation in parietal cortex transcends a fixed frame of reference.

ABSTRACT: Neurons in the lateral intraparietal cortex represent the formation of a decision when it is linked to a specific action, such as an eye movement to a choice target. However, these neurons should be unable to represent a decision that transpires across actions that would disrupt this linkage. We investigated this limitation by simultaneously recording many neurons from two rhesus monkeys. Although intervening actions disrupt the representation by single neurons, the ensemble achieves continuity of the decision process by passing information from currently active neurons to neurons that will become active after the action. In this way, the representation of an evolving decision can be generalized across actions and transcends the frame of reference that specifies the neural response fields. The finding extends previous observations of receptive field remapping, thought to support the stability of perception across eye movements, to the continuity of a thought process, such as a decision.

Decision and navigation in mouse parietal cortex.

Project description:Posterior parietal cortex (PPC) has been implicated in navigation, in the control of movement, and in visually-guided decisions. To relate these views, we measured activity in PPC while mice performed a virtual navigation task driven by visual decisions. PPC neurons were selective for specific combinations of the animal's spatial position and heading angle. This selectivity closely predicted both the activity of individual PPC neurons, and the arrangement of their collective firing patterns in choice-selective sequences. These sequences reflected PPC encoding of the animal's navigation trajectory. Using decision as a predictor instead of heading yielded worse fits, and using it in addition to heading only slightly improved the fits. Alternative models based on visual or motor variables were inferior. We conclude that when mice use vision to choose their trajectories, a large fraction of parietal cortex activity can be predicted from simple attributes such as spatial position and heading.

Diverse spatial reference frames of vestibular signals in parietal cortex.

Project description:Reference frames are important for understanding how sensory cues from different modalities are coordinated to guide behavior, and the parietal cortex is critical to these functions. We compare reference frames of vestibular self-motion signals in the ventral intraparietal area (VIP), parietoinsular vestibular cortex (PIVC), and dorsal medial superior temporal area (MSTd). Vestibular heading tuning in VIP is invariant to changes in both eye and head positions, indicating a body (or world)-centered reference frame. Vestibular signals in PIVC have reference frames that are intermediate between head and body centered. In contrast, MSTd neurons show reference frames between head and eye centered but not body centered. Eye and head position gain fields were strongest in MSTd and weakest in PIVC. Our findings reveal distinct spatial reference frames for representing vestibular signals and pose new challenges for understanding the respective roles of these areas in potentially diverse vestibular functions.

Synaptic wiring motifs in posterior parietal cortex support decision-making.

Project description:The posterior parietal cortex exhibits choice-selective activity during perceptual decision-making tasks1-10. However, it is not known how this selective activity arises from the underlying synaptic connectivity. Here we combined virtual-reality behaviour, two-photon calcium imaging, high-throughput electron microscopy and circuit modelling to analyse how synaptic connectivity between neurons in the posterior parietal cortex relates to their selective activity. We found that excitatory pyramidal neurons preferentially target inhibitory interneurons with the same selectivity. In turn, inhibitory interneurons preferentially target pyramidal neurons with opposite selectivity, forming an opponent inhibition motif. This motif was present even between neurons with activity peaks in different task epochs. We developed neural-circuit models of the computations performed by these motifs, and found that opponent inhibition between neural populations with opposite selectivity amplifies selective inputs, thereby improving the encoding of trial-type information. The models also predict that opponent inhibition between neurons with activity peaks in different task epochs contributes to creating choice-specific sequential activity. These results provide evidence for how synaptic connectivity in cortical circuits supports a learned decision-making task.

Encoding and decoding in parietal cortex during sensorimotor decision-making.

Project description:It has been suggested that the lateral intraparietal area (LIP) of macaques plays a fundamental role in sensorimotor decision-making. We examined the neural code in LIP at the level of individual spike trains using a statistical approach based on generalized linear models. We found that LIP responses reflected a combination of temporally overlapping task- and decision-related signals. Our model accounts for the detailed statistics of LIP spike trains and accurately predicts spike trains from task events on single trials. Moreover, we derived an optimal decoder for heterogeneous, multiplexed LIP responses that could be implemented in biologically plausible circuits. In contrast with interpretations of LIP as providing an instantaneous code for decision variables, we found that optimal decoding requires integrating LIP spikes over two distinct timescales. These analyses provide a detailed understanding of neural representations in LIP and a framework for studying the coding of multiplexed signals in higher brain areas.

Rapid and independent memory formation in the parietal cortex.

Project description:Previous evidence indicates that the brain stores memory in two complementary systems, allowing both rapid plasticity and stable representations at different sites. For memory to be established in a long-lasting neocortical store, many learning repetitions are considered necessary after initial encoding into hippocampal circuits. To elucidate the dynamics of hippocampal and neocortical contributions to the early phases of memory formation, we closely followed changes in human functional brain activity while volunteers navigated through two different, initially unknown virtual environments. In one condition, they were able to encode new information continuously about the spatial layout of the maze. In the control condition, no information could be learned because the layout changed constantly. Our results show that the posterior parietal cortex (PPC) encodes memories for spatial locations rapidly, beginning already with the first visit to a location and steadily increasing activity with each additional encounter. Hippocampal activity and connectivity between the PPC and hippocampus, on the other hand, are strongest during initial encoding, and both decline with additional encounters. Importantly, stronger PPC activity related to higher memory-based performance. Compared with the nonlearnable control condition, PPC activity in the learned environment remained elevated after a 24-h interval, indicating a stable change. Our findings reflect the rapid creation of a memory representation in the PPC, which belongs to a recently proposed parietal memory network. The emerging parietal representation is specific for individual episodes of experience, predicts behavior, and remains stable over offline periods, and must therefore hold a mnemonic function.

Free choice activates a decision circuit between frontal and parietal cortex.

Project description:We often face alternatives that we are free to choose between. Planning movements to select an alternative involves several areas in frontal and parietal cortex that are anatomically connected into long-range circuits. These areas must coordinate their activity to select a common movement goal, but how neural circuits make decisions remains poorly understood. Here we simultaneously record from the dorsal premotor area (PMd) in frontal cortex and the parietal reach region (PRR) in parietal cortex to investigate neural circuit mechanisms for decision making. We find that correlations in spike and local field potential (LFP) activity between these areas are greater when monkeys are freely making choices than when they are following instructions. We propose that a decision circuit featuring a sub-population of cells in frontal and parietal cortex may exchange information to coordinate activity between these areas. Cells participating in this decision circuit may influence movement choices by providing a common bias to the selection of movement goals.

Maximizing Without Borders: Evidence That Maximizing Transcends Decision Domains.

Project description:Do maximizers maximize across decision domains? An assumption underlying the literature on maximizing is that the tendency to strive to make the best choice spans domains. The current research provides a direct test of this assumption by examining the association between trait maximizing and domain-specific maximizing, consisting of maximizing measures in a wide range of decisions (consumer goods, services and experiences, and life decisions). Study 1 tested this association at two different time points in order to minimize common method bias. Study 2 was a high-powered pre-registered cross-sectional replication. Results of both studies showed that trait maximizing was associated with higher maximizing tendencies across all three decision domains. However, in line with prior research suggesting that people generally maximize less in experiential than in material domains, trait maximizing was associated with maximizing in services and experiences significantly less than with maximizing in consumer goods or in life decisions. These results provide empirical support for a central tenet of maximizing theory and suggest useful directions for future research.

Transformation of acoustic information to sensory decision variables in the parietal cortex.

Project description:The process by which sensory evidence contributes to perceptual choices requires an understanding of its transformation into decision variables. Here, we address this issue by evaluating the neural representation of acoustic information in the auditory cortex-recipient parietal cortex, while gerbils either performed a two-alternative forced-choice auditory discrimination task or while they passively listened to identical acoustic stimuli. During task engagement, stimulus identity decoding performance from simultaneously recorded parietal neurons significantly correlated with psychometric sensitivity. In contrast, decoding performance during passive listening was significantly reduced. Principal component and geometric analyses revealed the emergence of low-dimensional encoding of linearly separable manifolds with respect to stimulus identity and decision, but only during task engagement. These findings confirm that the parietal cortex mediates a transition of acoustic representations into decision-related variables. Finally, using a clustering analysis, we identified three functionally distinct subpopulations of neurons that each encoded task-relevant information during separate temporal segments of a trial. Taken together, our findings demonstrate how parietal cortex neurons integrate and transform encoded auditory information to guide sound-driven perceptual decisions.

Neural Mechanisms of Post-error Adjustments of Decision Policy in Parietal Cortex.

Project description:Humans often slow down after mistakes (post-error slowing [PES]), but the neural mechanism and adaptive role of PES remain controversial. We studied changes in the neural mechanisms of decision making after errors in humans and monkeys that performed a motion direction discrimination task. We found that PES is mediated by two factors: a reduction in sensitivity to sensory information and an increase in the decision bound. Both effects are implemented through dynamic changes in the decision-making process. Neuronal responses in the monkey lateral intraparietal area revealed that bound changes are implemented by decreasing an evidence-independent urgency signal. They also revealed a reduction in the rate of evidence accumulation, reflecting reduced sensitivity. These changes in the bound and sensitivity provide a quantitative account of choices and response times. We suggest that PES reflects an adaptive increase of decision bound in anticipation of maladaptive reductions in sensitivity to incoming evidence.

Choice-specific sequences in parietal cortex during a virtual-navigation decision task.

Project description:The posterior parietal cortex (PPC) has an important role in many cognitive behaviours; however, the neural circuit dynamics underlying PPC function are not well understood. Here we optically imaged the spatial and temporal activity patterns of neuronal populations in mice performing a PPC-dependent task that combined a perceptual decision and memory-guided navigation in a virtual environment. Individual neurons had transient activation staggered relative to one another in time, forming a sequence of neuronal activation spanning the entire length of a task trial. Distinct sequences of neurons were triggered on trials with opposite behavioural choices and defined divergent, choice-specific trajectories through a state space of neuronal population activity. Cells participating in the different sequences and at distinct time points in the task were anatomically intermixed over microcircuit length scales (<100 micrometres). During working memory decision tasks, the PPC may therefore perform computations through sequence-based circuit dynamics, rather than long-lived stable states, implemented using anatomically intermingled microcircuits.
