Metasurface-enhanced mid-infrared spectroscopy in the liquid phase.

ABSTRACT: Vibrational spectroscopy is an important tool in chemical and biological analysis. A key issue when applying vibrational spectroscopy to dilute liquid samples is the inherently low sensitivity caused by short interaction lengths and small extinction coefficients, combined with low target molecule concentrations. Here, we introduce a novel type of surface-enhanced infrared absorption spectroscopy based on the resonance of a dielectric metasurface. We demonstrate that the method is suitable for probing vibrational bands of dilute analytes with a range of spectral linewidths. We observe that the absorption signal is enhanced by 1-2 orders of magnitude and show that this enhancement leads to a lower limit of detection compared to attenuated total reflection (ATR). Overall, the technique provides an important addition to the spectroscopist's toolkit especially for probing dilute samples.

Surface-enhanced mid-infrared absorption spectroscopy using miniaturized-disc metasurface.

Project description:Surface-enhanced infrared spectroscopy is an important technique for improving the signal-to-noise ratio of spectroscopic material identification measurements in the mid-infrared fingerprinting region. However, the lower bound of the fingerprinting region receives much less attention due to a scarcity of transparent materials, more expensive sources, and weaker plasmonic effects. In this paper, we present a miniaturized metasurface unit cell for surface-enhanced infrared spectroscopy of the 15-[Formula: see text]m vibrational band of CO[Formula: see text]. The unit cell consists of a gold disc, patterned along the edge with fine gaps/wires to create a resonant metamaterial liner. In simulation, our plasmonic metamaterial-lined disc achieves greater than [Formula: see text] the average field intensity enhancement of a comparable dipole array and a miniaturized size of [Formula: see text] using complex, 100-nm features that are patterned using 100-kV electron-beam lithography. In a simple experiment, the metamaterial-lined disc metasurface shows a high tolerance to fabrication imperfections and enhances the absorption of CO[Formula: see text] at 15 [Formula: see text]m. The resonant wavelength and reflection magnitude can be tuned over a wide range by adjusting the liner feature sizes and the metasurface array pitch to target other vibrational bands. This work is a step toward low-cost, more compact on-chip integrated gas sensors.

Mid- to long-wave infrared computational spectroscopy with a graphene metasurface modulator.

Project description:In recent years there has been much interest concerning the development of modulators in the mid- to long-wave infrared, based on emerging materials such as graphene. These have been frequently pursued for optical communications, though also for other specialized applications such as infrared scene projectors. Here we investigate a new application for graphene modulators in the mid- to long-wave infrared. We demonstrate, for the first time, computational spectroscopy in the mid- to long-wave infrared using a graphene-based metasurface modulator. Furthermore, our metasurface device operates at low gate voltage. To demonstrate computational spectroscopy, we provide our algorithm with the measured reflection spectra of the modulator at different gate voltages. We also provide it with the measured reflected light power as a function of the gate voltage. The algorithm then estimates the input spectrum. We show that the reconstructed spectrum is in good agreement with that measured directly by a Fourier transform infrared spectrometer, with a normalized mean-absolute-error (NMAE) of 0.021.

Dielectric dual-dimer metasurface for enhanced mid-infrared chiral sensing under both excitation modes.

Project description:Chirality (C) is a fundamental symmetry property of objects. Detecting and distinguishing molecular chirality in the infrared spectrum is important in life sciences, biology, and chemistry. In this paper, we demonstrate an achiral metasurface based on a gaped dual-germanium-dimer array for enhanced mid-infrared chiral sensing under both circularly polarized light (CPL) and linearly polarized light (LPL) excitations. With the metasurface, strong electric and magnetic dipole resonances with large field enhancement can be generated, resulting in an accessible superchiral hotspot in the dimer gaps under both excitation modes. The maximum electric and magnetic field enhancements exceed 220 and 100 for the bare metasurface, and exceed 70 and 60 for the metasurface coated with a 50 nm chiral biolayer under both excitations, respectively. Importantly, a high volume-averaged C enhancement C E_ave of 241 (444) and C E_ave_bio of 161 (102) under CPL (LPL) excitation can be achieved for the bare metasurface and it coated with the chiral biolayer, respectively. These results may open up new possibilities for ultrasensitive vibrational circular dichroism (VCD) and rotational optical dispersion (ORD) spectroscopy in the mid-infrared range.

Triboelectric-induced ion mobility for artificial intelligence-enhanced mid-infrared gas spectroscopy.

Project description:Isopropyl alcohol molecules, as a biomarker for anti-virus diagnosis, play a significant role in the area of environmental safety and healthcare relating volatile organic compounds. However, conventional gas molecule detection exhibits dramatic drawbacks, like the strict working conditions of ion mobility methodology and weak light-matter interaction of mid-infrared spectroscopy, yielding limited response of targeted molecules. We propose a synergistic methodology of artificial intelligence-enhanced ion mobility and mid-infrared spectroscopy, leveraging the complementary features from the sensing signal in different dimensions to reach superior accuracy for isopropyl alcohol identification. We pull in "cold" plasma discharge from triboelectric generator which improves the mid-infrared spectroscopic response of isopropyl alcohol with good regression prediction. Moreover, this synergistic methodology achieves ~99.08% accuracy for a precise gas concentration prediction, even with interferences of different carbon-based gases. The synergistic methodology of artificial intelligence-enhanced system creates mechanism of accurate gas sensing for mixture and regression prediction in healthcare.

Mid-infrared cross-comb spectroscopy.

Project description:Dual-comb spectroscopy has been proven beneficial in molecular characterization but remains challenging in the mid-infrared region due to difficulties in sources and efficient photodetection. Here we introduce cross-comb spectroscopy, in which a mid-infrared comb is upconverted via sum-frequency generation with a near-infrared comb of a shifted repetition rate and then interfered with a spectral extension of the near-infrared comb. We measure CO2 absorption around 4.25 µm with a 1-µm photodetector, exhibiting a 233-cm-1 instantaneous bandwidth, 28000 comb lines, a single-shot signal-to-noise ratio of 167 and a figure of merit of 2.4 × 106 Hz1/2. We show that cross-comb spectroscopy can have superior signal-to-noise ratio, sensitivity, dynamic range, and detection efficiency compared to other dual-comb-based methods and mitigate the limits of the excitation background and detector saturation. This approach offers an adaptable and powerful spectroscopic method outside the well-developed near-IR region and opens new avenues to high-performance frequency-comb-based sensing with wavelength flexibility.

Monitoring the effects of chemical stimuli on live cells with metasurface-enhanced infrared reflection spectroscopy.

Project description:Infrared spectroscopy has found wide applications in the analysis of biological materials. A more recent development is the use of engineered nanostructures - plasmonic metasurfaces - as substrates for metasurface-enhanced infrared reflection spectroscopy (MEIRS). Here, we demonstrate that strong field enhancement from plasmonic metasurfaces enables the use of MEIRS as a highly informative analytic technique for real-time monitoring of cells. By exposing live cells cultured on a plasmonic metasurface to chemical compounds, we show that MEIRS can be used as a label-free phenotypic assay for detecting multiple cellular responses to external stimuli: changes in cell morphology, adhesion, and lipid composition of the cellular membrane, as well as intracellular signaling. Using a focal plane array detection system, we show that MEIRS also enables spectro-chemical imaging at the single-cell level. The described metasurface-based all-optical sensor opens the way to a scalable, high-throughput spectroscopic assay for live cells.

Probing the Drug Dynamics of Chemotherapeutics Using Metasurface-Enhanced Infrared Reflection Spectroscopy of Live Cells.

Project description:Infrared spectroscopy has drawn considerable interest in biological applications, but the measurement of live cells is impeded by the attenuation of infrared light in water. Metasurface-enhanced infrared reflection spectroscopy (MEIRS) had been shown to mitigate the problem, enhance the cellular infrared signal through surface-enhanced infrared absorption, and encode the cellular vibrational signatures in the reflectance spectrum at the same time. In this study, we used MEIRS to study the dynamic response of live cancer cells to a newly developed chemotherapeutic metal complex with distinct modes of action (MoAs): tricarbonyl rhenium isonitrile polypyridyl (TRIP). MEIRS measurements demonstrated that administering TRIP resulted in long-term (several hours) reduction in protein, lipid, and overall refractive index signals, and in short-term (tens of minutes) increase in these signals, consistent with the induction of endoplasmic reticulum stress. The unique tricarbonyl IR signature of TRIP in the bioorthogonal spectral window was monitored in real time, and was used as an infrared tag to detect the precise drug delivery time that was shown to be closely correlated with the onset of the phenotypic response. These results demonstrate that MEIRS is an effective label-free real-time cellular assay capable of detecting and interpreting the early phenotypic responses of cells to IR-tagged chemotherapeutics.

Liquid-Phase Peak Force Infrared Microscopy for Chemical Nanoimaging and Spectroscopy.

Project description:Peak force infrared (PFIR) microscopy is an emerging atomic force microscopy that bypasses Abbe's diffraction limit in achieving chemical nanoimaging and spectroscopy. The PFIR microscopy mechanically detects the infrared photothermal responses in the dynamic tip-sample contact of peak force tapping mode and has been applied for a variety of samples, ranging from soft matters, photovoltaic heterojunctions, to polaritonic materials under the air conditions. In this article, we develop and demonstrate the PFIR microscopy in the liquid phase for soft matters and biological samples. With the capability of controlling fluid compositions on demand, the liquid-phase peak force infrared (LiPFIR) microscopy enables in situ tracking of the polymer surface reorganization in fluids and detecting the product of click chemical reaction in the aqueous phase. Both broadband spectroscopy and infrared imaging with ∼10 nm spatial resolution are benchmarked in the fluid phase, together with complementary mechanical information. We also demonstrate the LiPFIR microscopy on revealing the chemical composition of a budding site of yeast cell wall particles in water as an application on biological structures. The label-free, nondestructive chemical nanoimaging and spectroscopic capabilities of the LiPFIR microscopy will facilitate the investigations of soft matters and their transformations at the solid/liquid interface.

Mid-infrared photothermal heterodyne spectroscopy in a liquid crystal using a quantum cascade laser.

Project description:We report a technique to measure the mid-infrared photothermal response induced by a tunable quantum cascade laser in the neat liquid crystal 4-octyl-4'-cyanobiphenyl (8CB), without any intercalated dye. Heterodyne detection using a Ti:sapphire laser of the response in the solid, smectic, nematic and isotropic liquid crystal phases allows direct detection of a weak mid-infrared normal mode absorption using an inexpensive photodetector. At high pump power in the nematic phase, we observe an interesting peak splitting in the photothermal response. Tunable lasers that can access still stronger modes will facilitate photothermal heterodyne mid-infrared vibrational spectroscopy.

Surface-enhanced molecular spectroscopy (SEMS) based on perfect-absorber metamaterials in the mid-infrared.

Project description:Surface-enhanced infrared absorption spectroscopy has attracted increased attention for direct access to molecular vibrational fingerprints in the mid-infrared. Perfect-absorber metamaterials (PAMs) with multi-band spectral responses and significant enhancement of the local near-field intensity were developed to improve the intrinsic absorption cross sections of absorption spectrum to identify the vibrational spectra of biomolecules. To verify its performance, the proposed infrared PAM array was used to identify the molecular stretches of a Parylene C film. The resonant responses of the infrared PAMs were accurately tuned to the vibrational modes of the C = C target bonds. The vibrational stretches of the C = C moiety were observed and the auto-fluorescence mechanisms of the Parylene C film were monitored. The unique properties of the PAMs indicate that this approach is a promising strategy for surface-enhanced molecular absorption spectroscopy (SEMS) in the mid-infrared region and for the tracking of characteristic molecular vibrational modes.
