Endophytes from Halotolerant Plants Aimed to Overcome Salinity and Draught.

ABSTRACT: The aim of our research was to study the endosphere of four halophytic plants: Salicornia europaea L., Salsola australis (R.Br.), Bassia sedoides (Pall.) and Kochia prostrata (L.) Schrad. from arid and saline areas of the Stavropol Territory, Russia. In total, 28 endophyte strains were isolated from the roots and stems of these halophytic plants. Most of the isolates (23 out of 28) were identified as Bacillus sp. while others belonged to the genera Oceanobacillus, Paenibacillus, Pantoea, Alcaligenes and Myroides. Three strains of Bacillus sp. (Se5R, Se1-1R, and Se1-3S), isolated from the S. europaea were capable of growth at 55 °C and in 10% of NaCl. Strains Se1-4S, Kp20-2S, and Bs11-2S Bacillus sp. (isolated from the S. australis, K. prostrata and B. sedoides, respectively) demonstrated strong plant growth promoting activity: 85-265% over control lettuce plants and a high degree of growth suppression (59.1-81.2%) of pathogenic fungi Fusarium oxysporum, Bipolaris sorokiniana and Rhizoctonia solani. Selected strains can be promising candidates for the development of bioinoculants to facilitate salt soil phytoremediation and be beneficial for mitigating the salt stress to the plants growing in salt-affected habitats.

Project description:Plant diseases caused by phytopathogens are responsible for significant crop losses worldwide. Resistance induction and biological control have been exploited in agriculture due to their enormous potential. In this study, we investigated the antimicrobial potential of endophytic fungi of leaves and petioles of medicinal plants Vochysia divergens and Stryphnodendron adstringens located in two regions of high diversity in Brazil, Pantanal, and Cerrado, respectively. We recovered 1,304 fungal isolates and based on the characteristics of the culture, were assigned to 159 phenotypes. One isolate was selected as representative of each phenotype and studied for antimicrobial activity against phytopathogens. Isolates with better biological activities were identified based on DNA sequences and phylogenetic analyzes. Among the 159 representative isolates, extracts from 12 endophytes that inhibited the mycelial growth (IG) of Colletotrichum abscissum (≥40%) were selected to expand the antimicrobial analysis. The minimum inhibitory concentrations (MIC) of the extracts were determined against citrus pathogens, C. abscissum, Phyllosticta citricarpa and Xanthomonas citri subsp. citri and the maize pathogen Fusarium graminearum. The highest activity against C. abscissum were from extracts of Pseudofusicoccum stromaticum CMRP4328 (IG: 83% and MIC: 40 μg/mL) and Diaporthe vochysiae CMRP4322 (IG: 75% and MIC: 1 μg/mL), both extracts also inhibited the development of post-bloom fruit drop symptoms in citrus flowers. The extracts were promising in inhibiting the mycelial growth of P. citricarpa and reducing the production of pycnidia in citrus leaves. Among the isolates that showed activity, the genus Diaporthe was the most common, including the new species D. cerradensis described in this study. In addition, high performance liquid chromatography, UV detection, and mass spectrometry and thin layer chromatography analyzes of extracts produced by endophytes that showed high activity, indicated D. vochysiae CMRP4322 and P. stromaticum CMRP4328 as promising strains that produce new bioactive natural products. We report here the capacity of endophytic fungi of medicinal plants to produce secondary metabolites with biological activities against phytopathogenic fungi and bacteria. The description of the new species D. cerradensis, reinforces the ability of medicinal plants found in Brazil to host a diverse group of fungi with biotechnological potential.

Project description:Halophytic plants can tolerate a high level of salinity through several morphological and physiological adaptations along with the presence of salt tolerant rhizo-microbiome. These microbes release phytohormones which aid in alleviating salinity stress and improve nutrient availability. The isolation and identification of such halophilic PGPRs can be useful in developing bio-inoculants for improving the salt tolerance and productivity of non-halophytic plants under saline conditions. In this study, salt-tolerant bacteria with multiple plant growth promoting characteristics were isolated from the rhizosphere of a predominant halophyte, Sesuvium portulacastrum grown in the coastal and paper mill effluent irrigated soils. Among the isolates, nine halotolerant rhizobacterial strains that were able to grow profusely at a salinity level of 5% NaCl were screened. These isolates were found to have multiple plant growth promoting (PGP) traits, especially 1-aminocyclopropane-1-carboxylic acid deaminase activity (0.32–1.18 μM of α-ketobutyrate released mg−1 of protein h−1) and indole acetic acid (9.4–22.8 μg mL−1). The halotolerant PGPR inoculation had the potential to improve salt tolerance in Vigna mungo L. which was reflected in significantly (p < 0.05) higher germination percentage (89%) compared to un-inoculated seeds (65%) under 2% NaCl. Similarly, shoot length (8.9–14.6 cm) and vigor index (792–1785) were also higher in inoculated seeds. The strains compatible with each other were used for the preparation of two bioformulations and these microbial consortia were tested for their efficacy in salt stress alleviation of Vigna mungo L. under pot study. The inoculation improved the photosynthetic rate (12%), chlorophyll content (22%), shoot length (5.7%) and grain yield (33%) in Vigna mungo L. The enzymatic activity of catalase and superoxide dismutase were found to be lower (7.0 and 1.5%, respectively) in inoculated plants. These results revealed that halotolerant PGPR isolated from S. portulacastrum can be a cost-effective and ecologically sustainable method to improve crop productivity under high saline conditions.
