Evidence-Based Anti-Diabetic Properties of Plant from the Occitan Valleys of the Piedmont Alps.

ABSTRACT: Data on urban and rural diabetes prevalence ratios show a significantly lower presence of diabetes in rural areas. Several bioactive compounds of plant origin are known to exert anti-diabetic properties. Interestingly, most of them naturally occur in different plants present in mountainous areas and are linked to traditions of herbal use. This review will aim to evaluate the last 10 years of evidence-based data on the potential anti-diabetic properties of 9 plants used in the Piedmont Alps (North-Western Italy) and identified through an ethnobotanical approach, based on the Occitan language minority of the Cuneo province (Sambucus nigra L., Achillea millefolium L., Cornus mas L., Vaccinium myrtillus L., Fragaria vesca L., Rosa canina L., Rubus idaeus L., Rubus fruticosus/ulmifolius L., Urtica dioica L.), where there is a long history of herbal remedies. The mechanism underlying the anti-hyperglycemic effects and the clinical evidence available are discussed. Overall, this review points to the possible use of these plants as preventive or add-on therapy in treating diabetes. However, studies of a single variety grown in the geographical area, with strict standardization and titration of all the active ingredients, are warranted before applying the WHO strategy 2014-2023.

Project description:BackgroundPhyto-remedies for diabetic control are popular among patients with Type II Diabetes mellitus (DM), in addition to other diabetic control measures. A number of plant species are known to possess diabetic control properties. Costus pictus D. Don is popularly known as "Insulin Plant" in Southern India whose leaves have been reported to increase insulin pools in blood plasma. Next Generation Sequencing is employed as a powerful tool for identifying molecular signatures in the transcriptome related to physiological functions of plant tissues. We sequenced the leaf transcriptome of C. pictus using Illumina reversible dye terminator sequencing technology and used combination of bioinformatics tools for identifying transcripts related to anti-diabetic properties of C. pictus.ResultsA total of 55,006 transcripts were identified, of which 69.15% transcripts could be annotated. We identified transcripts related to pathways of bixin biosynthesis and geraniol and geranial biosynthesis as major transcripts from the class of isoprenoid secondary metabolites and validated the presence of putative norbixin methyltransferase, a precursor of Bixin. The transcripts encoding these terpenoids are known to be Peroxisome Proliferator-Activated Receptor (PPAR) agonists and anti-glycation agents. Sequential extraction and High Performance Liquid Chromatography (HPLC) confirmed the presence of bixin in C. pictus methanolic extracts. Another significant transcript identified in relation to anti-diabetic, anti-obesity and immuno-modulation is of Abscisic Acid biosynthetic pathway. We also report many other transcripts for the biosynthesis of antitumor, anti-oxidant and antimicrobial metabolites of C. pictus leaves.ConclusionSolid molecular signatures (transcripts related to bixin, abscisic acid, and geranial and geraniol biosynthesis) for the anti-diabetic properties of C. pictus leaves and vital clues related to the other phytochemical functions like antitumor, anti-oxidant, immuno-modulatory, anti-microbial and anti-malarial properties through the secondary metabolite pathway annotations are reported. The data provided will be of immense help to researchers working in the treatment of DM using herbal therapies.

Project description:BackgroundConserving both biodiversity and ecosystem services is a major goal of the Convention on Biological Diversity. Hotspots for biodiversity in the Andes significantly overlap with areas with dense human populations that sustain their economy through agricultural production. Therefore, developing management forms that reconcile food provisioning services-such as agriculture-with biodiversity conservation must be addressed to avoid social conflicts and to improve conservation in areas where biodiversity co-occurs with other ecosystem services. Here, we present a high-resolution conservation plan for vascular plants and agriculture in the Ecuadorian Dry Inter-Andean Valleys (DIAV) hotspot. Trade-offs in conserving important areas for both biodiversity and agriculture were explored.MethodsWe used a dataset containing 5,685 presence records for 95 plant species occurring in DIAVs, of which 14 species were endemic. We developed habitat suitability maps for the 95 species using Maxent. Prioritization analyses were carried out using a conservation planning framework. We developed three conservation scenarios that selected important areas for: biodiversity only, agriculture only, and for both biodiversity and agriculture combined.ResultsOur conservation planning analyses, capture 33.5% of biodiversity and 11% of agriculture under a scenario solely focused on the conservation of biodiversity. On the other hand, the top 17% fraction of the agriculture only scenario captures 10% of biodiversity and 28% of agriculture. When biodiversity and agriculture were considered in combination, their representation varied according to the importance given to agriculture. The most balanced solution that gives a nearly equal representation of both biodiversity and agriculture, was obtained when agriculture was given a slightly higher importance over biodiversity during the selection process.DiscussionThis is the first evaluation of trade-offs between important areas for biodiversity and agriculture in Ecuadorian DIAV. Our results showed that areas with high agricultural productivity and high biodiversity partly overlapped. Our study suggests that a land-sharing strategy would be appropriate for conserving plant diversity and agriculture in the DIAV. Overall, our study reinforces the idea that friendly practices in agriculture can contribute to biodiversity conservation.
