Dataset of phenotyping recombinant inbred lines population of wheat under heat stress conditions.

ABSTRACT: Heat stress is a genetically complex and physiologically diverse phenomenon. To overcome the effect of heat stress identification of genomic locations associated with heat stress tolerance is essential. This article provides the dataset of phenotyping used in the research article entitled "Mapping QTLs for grain yield components in wheat under heat stress". The presented data included the phenotyping of the 249 RIL population of F₈ and F₉ generations under timely and late sown conditions during the 2013-14 and 2014-15 crop seasons, respectively. The RIL population was derived from the cross between HUW510 and HD2808 wheat genotypes. A total of eight agronomic traits were subjected to phenotype and the heat susceptibility index (HSI) of these traits was estimated to identify the effect of heat stress on the parents and RIL population. The presented dataset could be utilized to understand the genetic basis for heat stress tolerance in wheat.

Project description:Salinity majorly hinders horizontal and vertical expansion in worldwide wheat production. Productivity can be enhanced using salt-tolerant wheat genotypes. However, the assessment of salt tolerance potential in bread wheat doubled haploid lines (DHL) through agro-physiological traits and stress-related gene expression analysis could potentially minimize the cost of breeding programs and be a powerful way for the selection of the most salt-tolerant genotype. We used an extensive set of agro-physiologic parameters and salt-stress-related gene expressions. Multivariate analysis was used to detect phenotypic and genetic variations of wheat genotypes more closely under salinity stress, and we analyzed how these strategies effectively balance each other. Four doubled haploid lines (DHLs) and the check cultivar (Sakha93) were evaluated in two salinity levels (without and 150 mM NaCl) until harvest. The five genotypes showed reduced growth under 150 mM NaCl; however, the check cultivar (Sakha93) died at the beginning of the flowering stage. Salt stress induced reduction traits, except the canopy temperature and initial electrical conductivity, which was found in each of the five genotypes, with the greatest decline occurring in the check cultivar (Sakha-93) and the least decline in DHL2. The genotypes DHL21 and DHL5 exhibited increased expression rate of salt-stress-related genes (TaNHX1, TaHKT1, and TaCAT1) compared with DHL2 and Sakha93 under salt stress conditions. Principle component analysis detection of the first two components explains 70.78% of the overall variation of all traits (28 out of 32 traits). A multiple linear regression model and path coefficient analysis showed a coefficient of determination (R2) of 0.93. The models identified two interpretive variables, number of spikelets, and/or number of kernels, which can be unbiased traits for assessing wheat DHLs under salinity stress conditions, given their contribution and direct impact on the grain yield.

Project description:Global environmental changes with more extreme episodes of heat waves are major threats to agricultural productivity. Heat stress in spring affects the reproductive stage of maize, resulting in tassel blast, pollen abortion, poor pollination, reduced seed set, barren ears and ultimately yield loss. As an aneamophelous crop, maize has a propensity for pollen abortion under heat stress conditions. To overcome the existing challenges of heat stress and pollen abortion, this study utilized a broad genetic base of maize germplasm to identify superior alleles to be utilized in breeding programs. A panel of 375 inbred lines was morpho-physiologically screened under normal and heat stress conditions in two locations across two consecutive planting seasons, 2017 and 2018. The exposure of pollen to high temperature showed drastic decline in pollen germination percentage. The average pollen germination percentage (PGP) at 35 and 45°C was 40.3% and 9.7%, respectively, an average decline of 30.6%. A subset of 275 inbred lines were sequenced using tunable genotyping by sequencing, resulting in 170,098 single nucleotide polymorphisms (SNPs) after filtration. Genome wide association of PGP in a subset of 122 inbred lines resulted in ten SNPs associated with PGP35°C (p ≤ 10-5), nine with PGP45°C (p ≤ 10-6-10-8) and ten SNPs associated with PGP ratio (p ≤ 10-5). No SNPs were found to be in common across PGP traits. The number of favorable alleles possessed by each inbred line for PGP35°C, PGP45°C, and the PGP ratio ranged between 4 and 10, 3-13 and 5-13, respectively. In contrast, the number of negative alleles for these traits ranged between 2 and 8, 3-13 and 3-13, respectively. Genetic mapping of yield (adjusted weight per plant, AWP-1) and flowering time (anthesis-silking interval, ASI) in 275 lines revealed five common SNPs: three shared for AWP-1 between normal and heat stress conditions, one for ASI between conditions, and one SNP, CM007648.1-86615409, was associated with both ASI and AWP-1. Variety selection can be performed based on these favorable alleles for various traits. These marker trait associations identified in the diversity panel can be utilized in breeding programs to improve heat stress tolerance in maize.
