Correlation Study of Biological Activity with Quercetin and Phenolics Content in Onion Extracts.

ABSTRACT: In this study it was shown that the fungistatic and antioxidant activities of onion extracts are related to the type of liquid used as the extractant and the technique of its preparation. A change in the antioxidant properties of white and red onion extracts was demonstrated with the change of the temperature of the pressurized hot water extraction process, which can be easily related to the changes accompanying the process of thermal processing of vegetables and fruits during cooking. Owing to the experimental and mathematical approaches concerning both the main and characteristic components of onions, i.e., quercetin and phenols, respectively, with the biological activity of the extracts, it was possible to demonstrate the significant share of these compounds in the antifungal and antioxidant properties of the extracts. Considering that the research was carried out, inter alia, on onion husks, demonstrating a very high potential of biological properties of this waste material from agricultural production, the research results presented in the paper should encourage the popularization of the use of this so far underestimated raw material for the production of various functional materials.

Project description:Quorum sensing (QS) regulates bacterial gene expression and studies suggest quercetin, a flavonol found in onion, as a QS inhibitor. There are no studies showing the anti-QS activity of plants containing quercetin in its native glycosylated forms. This study aimed to evaluate the antimicrobial and anti-QS potential of organic extracts of onion varieties and its representative phenolic compounds quercetin aglycone and quercetin 3-β-D-glucoside in the QS model bacteria Chromobacterium violaceum ATCC 12472, Pseudomonas aeruginosa PAO1, and Serratia marcescens MG1. Three phenolic extracts were obtained: red onion extract in methanol acidified with 2.5% acetic acid (RO-1), white onion extract in methanol (WO-1) and white onion extract in methanol ammonium (WO-2). Quercetin 4-O-glucoside and quercetin 3,4-O-diglucoside were identified as the predominant compounds in both onion varieties using HPLC-DAD and LC-ESI-MS/MS. However, quercetin aglycone, cyanidin 3-O-glucoside and quercetin glycoside were identified only in RO-1. The three extracts showed minimum inhibitory concentration (MIC) values equal to or above 125 μg/ml of dried extract. Violacein production was significantly reduced by RO-1 and quercetin aglycone, but not by quercetin 3-β-D-glucoside. Motility in P. aeruginosa PAO1 was inhibited by RO-1, while WO-2 inhibited S. marcescens MG1 motility only in high concentration. Quercetin aglycone and quercetin 3-β-D-glucoside were effective at inhibiting motility in P. aeruginosa PAO1 and S. marcescens MG1. Surprisingly, biofilm formation was not affected by any extracts or the quercetins tested at sub-MIC concentrations. In silico studies suggested a better interaction and placement of quercetin aglycone in the structures of the CviR protein of C. violaceum ATCC 12472 than the glycosylated compound which corroborates the better inhibitory effect of the former over violacein production. On the other hand, the two quercetins were well placed in the AHLs binding pockets of the LasR protein of P. aeruginosa PAO1. Overall onion extracts and quercetin presented antimicrobial activity, and interference on QS regulated production of violacein and swarming motility.

Project description:Seaweeds are potentially excellent sources of bioactive metabolites that could represent useful leads in the development of new functional ingredients in pharmaceutical and cosmetic industries. In the last decade, new marine bioprocess technologies have allowed the isolation of substances with biological properties. The brown alga Sargassum muticum (Yendo) Fensholt (Ochrophyta) was enzymatically hydrolyzed to prepare water-soluble extracts by using six different commercially available carbohydrate-degrading enzymes and two proteases. Evaluation of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) antioxidant, tyrosinase, elastase, and biofilm inhibition, antibacterial and antiviral activities as well as evaluation of cytotoxicity were realized for each extract. Total phenolic content was measured for extract characterization, and solid-phase extraction was useful to purify the enzymatic extract. Soluble total phenolic content of S. muticum Viscozyme extract was highest with 6.4% of dry weight. Enzymatic Celluclast and Viscozyme extracts had the lowest value of DPPH IC50 indicating a strong antiradical activity, 0.6 mg mL-1, in comparison with other enzymes. The ferric reducing antioxidant power ranged between 48.7 ?M Fe2+ Eq, digested with Viscozyme, and 60.8 ?M Fe2+ Eq, digested with Amyloglucosidase. Tyrosinase inhibition activity of S. muticum Neutrase extract was 41.3% higher compared to other enzymes. Elastase inhibition activity of S. muticum Shearzyme extract had highest activity (32.8%). All enzymatic extracts showed no cytotoxic effect towards the kidney Vero cells. Meanwhile, only S. muticum Neutrase and Alcalase extracts exhibited potential antiviral activity. In addition, S. muticum Viscozyme and Shearzyme extracts showed promising activity in suppressing the biofilm formation against Pseudomonas aeruginosa and Escherichia coli, respectively. Purification of S. muticum Viscozyme extracts by solid-phase extraction managed to concentrate the phenolic content and improve the bioactivity. These results indicate the promising potential of enzyme-assisted followed by solid-phase extraction in recovering phenolic content and in improving its bioactivity.
