Bioengineered intestinal tubules as a tool to test intestinal biological efficacy of lettuce species.

ABSTRACT: Lettuce (Lactuca sativa) is one of the most consumed and cultivated vegetables globally. Its breeding is focused on the improvement of yield and disease resistance. However, potential detrimental or beneficial health effects for the consumer are often not targeted in the breeding programs. Here, a bioengineered intestinal tubule was used to assess the intestinal efficacy of extracts from five plant accessions belonging to four Lactuca species. These four species include the domesticated L. sativa, closely related wild species L. serriola, and phylogenetically more distant wild relatives L. saligna and L. virosa. We assessed the epithelial barrier integrity, cell viability, cell attachment, brush border enzyme activity, and immune markers. Extracts from L. sativa cv. Salinas decreased cell attachment and brush border enzyme activity. However, extracts from the non-edible wild species L. saligna and L. virosa reduced the epithelial barrier functions, cell attachment, cell viability, and brush border enzyme activity. Since wild species represent a valuable germplasm pool, the bioengineered intestinal tubules could open ways to evaluate the safety and nutritional properties of the lettuce breeding material originating from crosses with wild Lactuca species.

Project description:Objectives:Spermatogenesis is a regular and lengthy process in which the function of testicular cells may potentially be influenced by several extrinsic and intrinsic stressors, including environmental factors such as magnetic resonance imaging (MRI) waves and radiation. Our study aimed to investigate the effects of MRI waves and fields on the testicular histology and morphometry of seminiferous tubules in mice. Methods:The experiment was conducted on 40 adult Naval Medical Research Institute mice. The control group was located in the center of the MRI bore while it was turned off, while the exposed group was exposed to the active scanner for 36 minutes once a week for three weeks. Our study included four groups: group I (control group at one hour after last exposure), group II (experimental group at one hour after last exposure), group III (control group at 35 days after last virtual exposure), and group IV (experimental group at 35 days after last exposure). We then assessed the tube and lumen diameters, as well as epithelium thickness of the seminiferous tubules. Results:Our data showed that MRI waves partially reduced testicular weight one hour after the last exposure (group II) compared to group I (p = 0.240). On the other hand, in group II the Johnson's score (score 10, complete spermatogenesis and perfect tubules) was 87.5% which was slightly less than recorded in groups I, III, and IV (91.4%, 92.2%, and 90.5%, respectively). Furthermore, the MRI in group II revealed induces vacuolization in the epithelium, arrest in primary spermatocytes in the pachytene stage as well as disruption in the testicular parenchyma. Conclusions:Long-term exposure to MRI waves has deleterious effects on the male reproductive system, fertility parameters, and the quantity of germ cells in the seminiferous tubules with the exception of the number of round spermatid cells and epithelial thickness. All these effects were reversible after a new period of spermatogenesis.

Project description:BackgroundSeed germination is a critical stage in plant life, and recent practices use nanomaterials for the improvement of plant seed germination indices. This study was conducted to assess the effect of laboratory prepared zinc oxide nanoparticles on the physiological and biochemical changes of lettuce seeds.MethodsLettuce seeds were soaked in a suspension of moderately polydisperse zinc oxide nanoparticles at two different concentrations (25 ppm or 50 ppm) and shaken for 3 h at 25 °C. Seeds treatment was followed subsequently by two to three days drying at ambient conditions. Treated seeds were stored for 3-4 weeks, at ambient conditions and then tested for germination in petri dishes. Germination was observed on daily basis and seedling length was measured. After imbibition and before the start of the visible germination, seeds were examined for topography and surface analysis using the scanning electron microscope and zinc uptake was measured by using the atomic absorption spectrometry and the energy dispersive X-ray. The pattern of mobilization of biomolecules was analyzed to detect any differences among different seed groups.ResultsThere was no loss of viability for the nanoparticles treated seeds. Indeed their germination was enhanced and their biomass increased. The activated performance of the nanoparticles imbibed seeds has been found to be correlated with an increased level of Zn inside lettuce seeds. The recorded measurements show a significant enhancement of seedling length. Interaction of zinc oxide nanoparticles with lettuce seeds mediates a variation in the biochemical processes. Changes detected in treated seeds were as following: reduced levels of the total carbohydrates (including simple saccharides and polysaccharides), higher capacity of protein synthesis, an elevated level of starch as well as an increased activity of antioxidant enzymes.Discussion and conclusionLettuce seeds primed with ZnO nanoparticles were found not only to maintain seed viability but even to exhibit a detectable level of germination enhancement compared to the control seeds. Overall, the promoted response of lettuce seeds during early stages of seed growth is encouraging for the application of ZnO NPs for seed priming for better germination indices.
