Project description:Differences in boldness are common between populations or between related species and are discussed as part of individual coping style, personality, or behavioral syndrome. Boldness has been found to be dependent on experience, social, and environmental contexts. The major aim of the present study was to establish an experimental environment that would allow analyzing the risk-taking behavior of 2 competing invasive goby species. Neogobius melanostomus was more active in the absence of a predator Sander lucioperca than N. fluviatilis and clearly spent more time "swimming" and "feeding" than N. fluviatilis. In addition, N. melanostomus was always faster than N. fluviatilis both when leaving the shelter and reaching offered food. Based on the different behaviors recorded, species-specific boldness scores were established using a principal component analysis. Although there was no overall difference in boldness scores between the 2 species, both competitive conditions and the effect of the predator played significant roles as factors influencing boldness. Neogobius melanostomus was more affected by the presence/absence of the predator than the social circumstances. Neogobius fluviatilis, on the other hand, was more active and bolder in competitive situations. However, when alone, N. fluviatilis was rather inactive and displayed altogether shy behavior, independent of the presence/absence of the predator. Thus, the study confirms the prediction that there are differences in behavior and behavioral plasticity, and therein predator-avoidance strategies, between ecologically similar species of goby living in sympatry. We argue that these differences may be related to differential habitat use of both invasive species that presently dominate the fish community in the Lower Rhine.

Project description:The "pace-of-life" syndrome (POLS) framework can encompass multiple personality axes that drive important functional behaviors (e.g., foraging behavior) and that co-vary with multiple life history traits. Food hoarding is an adaptive behavior important for an animal's ability to adapt to seasonal fluctuations in food availability. However, the empirical evidence for the relationships between animal personality and hoarding behavior remains unclear, including its fitness consequences in the POLS framework. In this study, the Mongolian gerbil (Meriones unguiculatus), a social rodent, was used as a model system to investigate how boldness or shyness is associated with food hoarding strategies during the food hoarding season and their impact on over-winter survival and reproduction at both individual and group levels. The results of this study showed that, compared with shy gerbils, bold gerbils had a lower effort foraging strategy during the food hoarding season and exhibited lower over-winter survival rates. However, bold-shy personality differences had no effect on over-winter reproduction. These findings suggest that the personality is a crucial factor influencing the foraging strategy during the food hoarding season in Mongolian gerbils. Personality may be related to energy states or the reaction to environmental changes (e.g., predation risk and food availability) in bold or shy social animals. These results reflect animal life history trade-offs between current versus future reproduction and reproduction versus self-maintenance, thereby helping Mongolian gerbils in adapting to seasonal fluctuations in their habitat.

Project description:This study explored if boldness could be used to predict social status. First, boldness was assessed by monitoring individual zebrafish behaviour in (1) an unfamiliar barren environment with no shelter (open field), (2) the same environment when a roof was introduced as a shelter, and (3) when the roof was removed and an unfamiliar object (Lego® brick) was introduced. Next, after a resting period of minimum one week, social status of the fish was determined in a dyadic contest and dominant/subordinate individuals were determined as the winner/loser of two consecutive contests. Multivariate data analyses showed that males were bolder than females and that the behaviours expressed by the fish during the boldness tests could be used to predict which fish would later become dominant and subordinate in the ensuing dyadic contest. We conclude that bold behaviour is positively correlated to dominance in zebrafish and that boldness is not solely a consequence of social dominance.

Project description:P-glycoprotein (Pgp) is a multidrug transporter that uses the energy from ATP binding and hydrolysis to export from cells a wide variety of hydrophobic compounds including anticancer drugs, and mediates the bioavailability and pharmacokinetics of many drugs. Lipids and cholesterol have been shown to modulate the substrate-stimulated ATPase activity of purified Pgp in detergent solution and the substrate transport activity after reconstitution into proteoliposomes. While lipid extracts from E. coli, liver or brain tissues generally support well Pgp's functionality, their ill-defined composition and high UV absorbance make them less suitable for optical biophysical assays. On the other hand, studies with defined synthetic lipids, usually the bilayer-forming phosphatidylcholine with or without cholesterol, are often plagued by low ATPase activity and low binding affinity of Pgp for drugs. Drawing from the lipid composition of mammalian plasma membranes, we here investigate how different head groups modulate the verapamil-stimulated ATPase activity of purified Pgp in detergent-lipid micelles and compare them with components of E. coli lipids. Our general approach was to assay modulation of verapamil-stimulation of ATPase activity by artificial lipid mixtures starting with the bilayer-forming palmitoyloyl-phosphatidylcholine (POPC) and -phosphatidylethanolamine (POPE). We show that POPC/POPE supplemented with sphingomyelin (SM), cardiolipin, or phosphatidic acid enhanced the verapamil-stimulated activity (Vmax) and decreased the concentration required for half-maximal activity (EC50). Cholesterol (Chol) and more so its soluble hemisuccinate derivative cholesteryl hemisuccinate substantially decreased EC50, perhaps by supporting the functional integrity of the drug binding sites. High concentrations of CHS (>15%) resulted in a significantly increased basal activity which could be due to binding of CHS to the drug binding site as transport substrate or as activator, maybe acting cooperatively with verapamil. Lastly, Pgp reconstituted into liposomes or nanodiscs displayed higher basal activity and sustained high levels of verapamil stimulated activity. The findings establish a stable source of artificial lipid mixtures containing either SM and cholesterol or CHS that restore Pgp functionality with activities and affinities similar to those in the natural plasma membrane environment and will pave the way for future functional and biophysical studies.

Project description:AbstractBoldness, the tendency to be explorative, risk prone and proactive, often varies consistently between individuals. An individual's position on the boldness-shyness continuum has many implications. Bold individuals may outperform shyer conspecifics during foraging as they cover more ground, accumulate information more rapidly and make more frequent food discoveries. Individual variation in boldness may also affect behavioural plasticity across environmental contexts, as the time to process new information, the ability to locate and memorise resources and the time and ability to apply prior information in a novel context all differ between individuals. The primary aim of the current study was to examine plasticity in, and covariation between, boldness, foraging speed and foraging accuracy across social foraging contexts. We showed that the stickleback that were shyest when foraging alone became relatively boldest when foraging in a social context and also delayed their entry to a known food patch the most in the presence of conspecifics. These results support the assertion that shyer foragers are more reactive to social cues and add to current knowledge of how an individual's position on the boldness-shyness continuum may correlate to foraging task performance and behavioural plasticity. We conclude that the correlation between boldness and behavioural plasticity may have broad relevance as the ability to adjust or retain behaviours in changing social environments could often have consequences for fitness.Significance statementAnimal personality may affect how much individuals change their behaviour to suit different environments. We studied the link between threespine stickleback personality (boldness or shyness), foraging performance and change in foraging performance when either alone or in the presence of other stickleback. We found that shyer threespine stickleback were more reactive to the presence of other fish when foraging. When observed or joined by other fish, shy stickleback started exploring earlier, but entered a known food patch later, than when alone. Bolder stickleback changed their foraging behaviour much less in the presence of other fish. Our results suggest that how bold or shy individuals are may have important consequences on how well they adjust their foraging behaviour to environmental change.

Project description:Within populations, some individuals tend to exhibit a bold or shy social behavior phenotype relative to the mean. The neural underpinnings of these differing phenotypes - also described as syndromes, personalities, and coping styles - is an area of ongoing investigation. Although a social decision-making network has been described across vertebrate taxa, most studies examining activity within this network do so in relation to exhibited differences in behavioral expression. Our study instead focuses on constitutive gene expression in bold and shy individuals by isolating baseline gene expression profiles that influence social boldness predisposition, rather than those reflecting the results of social interaction and behavioral execution. We performed this study on male green anole lizards (Anolis carolinensis), an established model organism for behavioral research, which provides a crucial comparison group to investigations of birds and mammals. After identifying subjects as bold or shy through repeated reproductive and agonistic behavior testing, we used RNA sequencing to compare gene expression profiles between these groups within various forebrain, midbrain, and hindbrain regions. The ventromedial hypothalamus had the largest group differences in gene expression, with bold males having increased expression of neuroendocrine and neurotransmitter receptor and calcium channel genes compared to shy males. Conversely, shy males express more integrin alpha-10 in the majority of examined regions. There were no significant group differences in physiology or hormone levels. Our results highlight the ventromedial hypothalamus as an important center of behavioral differences across individuals and provide novel candidates for investigations into the regulation of individual variation in social behavior phenotype.

Project description:The movement of ammonium across biologic membranes is a fundamental process in all living organisms and is mediated by the ubiquitous ammonium transporter/methylammonium permease/rhesus protein (Amt/Mep/Rh) family of transporters. Recent structural analysis and coupled mass spectrometry studies have shown that the Escherichia coli ammonium transporter AmtB specifically binds 1-palmitoyl-2-oleoyl phosphatidylglycerol (POPG). Upon POPG binding, several residues of AmtB undergo a small conformational change, which stabilizes the protein against unfolding. However, no studies have so far been conducted, to our knowledge, to explore whether POPG binding to AmtB has functional consequences. Here, we used an in vitro experimental assay with purified components, together with molecular dynamics simulations, to characterize the relation between POPG binding and AmtB activity. We show that the AmtB activity is electrogenic. Our results indicate that the activity, at the molecular level, of Amt in archaebacteria and eubacteria may differ. We also show that POPG is an important cofactor for AmtB activity and that, in the absence of POPG, AmtB cannot complete the full translocation cycle. Furthermore, our simulations reveal previously undiscovered POPG binding sites on the intracellular side of the lipid bilayer between the AmtB subunits. Project description:This study tested the hypothesis that time spent on social activities (i.e., in direct interaction with others) and time spent in social contexts (i.e., while others are present) is associated with survival in persons aged 70 and older. An observational study with mortality follow-ups was carried out in the former West Berlin, Germany (Berlin Aging Study). The sample was stratified by age and sex, and consisted of 473 persons aged 70 to 103 years. Social activity and social context measures were assessed in 1990-1993 by structured interviews in the participants' homes. Cox regression was used to model survival from time of interview. The main outcome measure was survival on 1 August 2003. Time spent on social activities was revealed as a predictor of survival only in analyses that did not control for confounding factors. In contrast, time spent in context "with friends" was significantly related to increased survival (relative risk=0.76, 95% confidence interval 0.59 to 0.99) even after several confounding factors were controlled for. This study suggests that time spent with friends affords a survival advantage among older adults, above and beyond the effects of other leisure activities. Future research on social participation and survival may benefit from an examination of the interaction between activity content and social context.

Project description:Novel or changing environments expose animals to diverse stressors that likely require coordinated hormonal and behavioral adaptations. Predicted adaptations to urban environments include attenuated physiological responses to stressors and bolder exploratory behaviors, but few studies to date have evaluated the impact of urban life on codivergence of these hormonal and behavioral traits in natural systems. Here, we demonstrate rapid adaptive shifts in both stress physiology and correlated boldness behaviors in a songbird, the dark-eyed junco, following its colonization of a novel urban environment. We compared elevation in corticosterone (CORT) in response to handling and flight initiation distances in birds from a recently established urban population in San Diego, California to birds from a nearby wildland population in the species' ancestral montane breeding range. We also measured CORT and exploratory behavior in birds raised from early life in a captive common garden study. We found persistent population differences for both reduced CORT responses and bolder exploratory behavior in birds from the colonist population, as well as significant negative covariation between maximum CORT and exploratory behavior. Although early developmental effects cannot be ruled out, these results suggest contemporary adaptive evolution of correlated hormonal and behavioral traits associated with colonization of an urban habitat.
