Chemical composition of the lipophilic compounds from the rind and pith of papyrus (Cyperus papyrus L.) stems.

ABSTRACT: Papyrus (Cyperus papyrus L.) is a sedge plant with a high rate of biomass productivity that represents an interesting raw material to produce chemicals, materials and fuels, which are currently still obtained from fossil resources, in the context of a lignocellulosic biorefinery. In this work, the content and chemical composition of the lipids present in papyrus stems were thoroughly studied. For this, the papyrus stems were separated into the rind and the pith. The lipid content accounted for 4.1% in the rind and 4.9% in the pith (based on dry matter). The main compounds identified in both parts of the papyrus stem were hydrocarbons, n-fatty acids, 2-hydroxyfatty acids, alcohols, alkylamides, mono- and diglycerides, steroids (sterols, ketones, hydrocarbons, esters and glycosides), tocopherols, tocopherol esters, phytol, phytol esters, alkyl ferulates, ω-carboxyalkyl ferulates (and their monoglycerides), and acylglycerol glycosides. The rind presented a predominance of n-fatty acids (6790 mg/kg; that represented 28.6% of all identified compounds), steroid compounds (6255 mg/kg; 26.3%), phytol and phytol esters (4985 mg/kg; 21.0%), and isoprenoid hydrocarbons, namely phytadiene and squalene (2660 mg/kg; 11.2%), while the most abundant lipids in the pith were steroids (8600 mg/kg; 44.4% of all identified compounds) and fatty acids (6245 mg/kg; 32.2%). Due to the great diversity and significant abundance of the compounds identified in papyrus, it can be considered as a potential raw material for biorefineries to obtain valuable phytochemicals of interest to various industrial sectors.

Project description:In rice, internode elongation is a critical aspect of plant development and agricultural productivity. Previous morphological and histochemical studies using [3H]thymidine have visualized the cell division zone (intercalary meristem) in internodes. However, it has remained unclear how the intercalary meristem forms during stem development. In addition, while a pith cavity forms in the central part of the rice stem, the spatiotemporal relationship between pith cavity formation and intercalary meristem development is not well understood. Therefore, we performed histological analysis of intercalary meristem and pith cavity development using C9285, a deepwater rice variety that shows internode elongation from the vegetative growth stage. We classified the developmental stages of the stem into four stages based on the analysis of pith cavity formation using Trypan blue, Calcein-AM, and MitoRed staining, and visualized dividing cells using the Click-iT EdU imaging assay. In Stage 1, no pith cavity was formed. Vertical cell rows were observed between above the axillary bud attachment and the upper node, suggesting anticlinal divisions that lead to internode formation in the early stage of stem development. In Stage 2, the first pith cavity formed in the pith of the foot, which is the region of axillary bud attachment. Compared to cell division in the internode, that in the foot was significantly activated resulting in slight elongation from Stage 1 to Stage 2. In Stage 3, cell division in the foot ceased, while active cell division at the base of the internode led to significant vertical elongation. The second pith cavity formed due to cell death in the pith of the internode. In Stage 4, the two pith cavities connected to form a single large pith cavity. Although the intercalary meristem maintained cell division activity, the number of cell divisions decreased. Based on these results, we propose a model for stem development that involves two phases of elongation regulation: primary elongation involving slight elongation in the foot, and secondary elongation involving significant internode elongation due to the activation of cell division and cell elongation in the intercalary meristem. This is the first study to anatomically elucidate the spatiotemporal relationship between intercalary meristem development and pith cavity formation in rice stem development. It provides new insights for future research on rice stem development and studies of other grass species.

Project description:BackgroundModern computational modeling could provide the key to obtaining new insights into the mechanisms of maize stalk failure as well as suggesting new ways to improve stalk strength. However, a complete set of mechanical properties of maize tissues is required to enable computational modeling of maize stems. This study developed two compression test methods for obtaining the longitudinal modulus of elasticity of both rind and pith tissues, assessed the influence of water content on tissue properties, and investigated the relationship between rind modulus and pith modulus. These methods involved uniform 5-7 cm segments of maize stems which were scanned using a flatbed scanner then tested in compression using a universal testing machine in both intact and dissected (rind-only and pith-only) states.ResultsThe modulus of elasticity of pith tissues was highest for fully turgid specimens and decreased as water was removed from the specimens. Water content was negatively correlated with the modulus of elasticity of the rind. Rind and pith tissues were found to be weakly correlated. The median ratio of rind modulus to pith modulus was found to be 17. Of the two methods investigated, the pith-only specimen preparation was found to be simple reliable while the rind-only method was found to be adversely affected by lateral bowing of the specimen.ConclusionsResearchers can use the information in this paper to improve computational models of maize stems in three ways: (1) by incorporating realistic values of the longitudinal modulus of elasticity of pith and rind tissues; (2) by selecting pith and rind properties that match empirically observed ratios; and (3) by incorporating appropriate dependencies between these material properties and water content. From an experimental perspective, the intact/pith-only experimental method outlined in this paper is simpler than previously reported methods and provides reliable estimates of both pith and rind modulus of elasticity values. Further research using this measurement method is recommended to more clearly understand the influence of water content and turgor pressure on tissue properties.
