Detection of the UV-vis silent biomarker trimethylamine-N-oxide via outer-sphere interactions in a lanthanide metal-organic framework.

ABSTRACT: Trimethylamine-N-oxide (TMAO) is a biomarker of the cardiovascular disease that is one of the leading causes of worldwide death. Facile detection of TMAO can significantly improve the survival rate of this disease by allowing early prevention. However, the UV-vis silent nature of TMAO makes it intricated to be detected by conventional sensing materials or analytical instruments. Here we show a bilanthanide metal-organic framework functionalized by borono group for the recognition of TMAO. Superior sensitivity, selectivity and anti-interference ability were achieved by the inverse emission intensity changes of the two lanthanide centers. The limit of detection is 15.6 μM, covering the clinical urinary concentration range of TMAO. A smartphone application was developed based on the change in R-G-B chromaticity. The sensing mechanism via a well-matched outer-sphere interaction governing the sensing function was studied in detail, providing fundamentals in molecular level for the design of advanced sensing materials for UV-Vis silent molecules.

Project description:Two-dimensional materials have been at the forefront of chemistry and materials science research for the past decade owing to promising applications across many fields. Improvements in exfoliation processes continually give access to new two-dimensional material compositions, demanding a deeper understanding of the defect structure and exfoliation mechanisms. Chemical exfoliation processes allow for both the fabrication of new, and the production of industrial-scale quantities of two-dimensional materials. For this reason, we report a rapid, efficient, and simple method for evaluating the exfoliation behavior of protonated lithium cobalt oxide. Using a two-step chemical exfoliation method, first by proton-cation exchange, then by treatment with an organo-ammonium hydroxide, the exfoliation yield of lithium cobalt oxide was quantified with a quick and powerful technique, ultraviolet-vis spectroscopy characterization. This method provided an in-depth analysis of the exfoliation of lithium cobalt oxide confirming and discovering many key aspects of its soft-chemical exfoliation relating to layered transition metal oxides. It was determined that the exfoliation yield has a strong dependence on multiple factors, such as the concentration of protons in the powder, the presence of water and hydroxide groups in solution, and the ionic radius and concentration of the intercalating cation. Both morphological changes occurring as a function of reaction conditions and the two-dimensional nature of the final sheets were revealed through scanning electron microscopy and atomic force microscopy. Relative proton concentration of acid-treated lithium cobalt oxide was determined as a function of acid concentration using time of flight secondary ion mass spectrometry after deuterated acid treatment. These experiments led to an improved understanding of the soft-chemical exfoliation of lithium cobalt oxide and can be applied to many layered transition metal oxides.

Project description:IntroductionTrimethylamine-N-oxide (TMAO) is a circulating biomarker associated with cardiovascular disease (CVD). Production of TMAO is facilitated by gut microbiota and dependent on micronutrients such as choline, betaine, and L-carnitine, present in foods such as red meat and eggs.HypothesisWe sought to predict serum TMAO quartile levels among healthy individuals at increased risk of CVD using clinical data via an ordinal logistic model.MethodsData from participants (n = 127) enrolled in a longitudinal observational study on CVD were used to build a predictive model for TMAO using ordinal logistic regression with demographic variables and 40 other variables considered related to CVD risk. First, univariate models for each covariate were tested (with serum TMAO quartiles as the dependent variable), and only variables with P < 0.30 were evaluated further. Second, demographic variables (age, gender, white vs. non-white race) were included in a multivariable model with each previously identified independent variable controlling for potential confounding. Last, the final model included fixed demographics and candidates from the confounder-adjusted model with P < 0.10.ResultsEight candidate variables were included in the final model, with only transferrin, high-density lipoprotein cholesterol (HDL-C) and race (white vs. non-white) showing significant associations with TMAO. Participants had 0.16 (Q2), 0.31 (Q3), and 0.20 (Q4) odds of being in a higher TMAO quartile compared with participants in the lowest transferrin quartile. Non-white participants had 2.92 times higher odds of being in the highest TMAO quartile compared to white individuals. Participants in the second quartile of HDL-C had 2.68 times higher odds of being in a higher TMAO quartile compared with participants in the lowest HDL-C quartile.ConclusionsTransferrin demonstrated a significant predictive association with TMAO and may represent a novel potential biomarker of increased CVD risk worthy of further study. These results warrant further examination of iron, metabolism, homeostasis, and gut microbiome to better understand and mitigate known increased CVD risk.
