On the interplay of hierarchies, conflicts, and cooperation: An experimental approach.

ABSTRACT: Previous research suggests that it is difficult to maintain cooperation in a large society when there is a strong hierarchical structure. In this study, we implement online human experiments to study the effects of exogenous variation in a particular notion of hierarchy on cooperation and conflict within groups. We demonstrate how cooperation can be maintained when collective action is accompanied by dyadic conflicts whose outcome feeds back on the hierarchical rank of the contestants. We find that the majority of individuals take part in conflicts and that highly ranked individuals mostly cooperate and engage in conflicts as a way to punish noncooperators. As a consequence, stable hierarchical groups can arise and maintain high levels of cooperation. Our results are in agreement with the prediction of earlier theoretical models on hierarchical societies and are relevant to understanding the interplay of hierarchy, cooperation, and conflict.

Project description:Protein promiscuity is of considerable interest due its role in adaptive metabolic plasticity, its fundamental connection with molecular evolution and also because of its biotechnological applications. Current views on the relation between primary and promiscuous protein activities stem largely from laboratory evolution experiments aimed at increasing promiscuous activity levels. Here, on the other hand, we attempt to assess the main features of the simultaneous modulation of the primary and promiscuous functions during the course of natural evolution. The computational/experimental approach we propose for this task involves the following steps: a function-targeted, statistical coupling analysis of evolutionary data is used to determine a set of positions likely linked to the recruitment of a promiscuous activity for a new function; a combinatorial library of mutations on this set of positions is prepared and screened for both, the primary and the promiscuous activities; a partial-least-squares reconstruction of the full combinatorial space is carried out; finally, an approximation to the Pareto set of variants with optimal primary/promiscuous activities is derived. Application of the approach to the emergence of folding catalysis in thioredoxin scaffolds reveals an unanticipated scenario: diverse patterns of primary/promiscuous activity modulation are possible, including a moderate (but likely significant in a biological context) simultaneous enhancement of both activities. We show that this scenario can be most simply explained on the basis of the conformational diversity hypothesis, although alternative interpretations cannot be ruled out. Overall, the results reported may help clarify the mechanisms of the evolution of new functions. From a different viewpoint, the partial-least-squares-reconstruction/Pareto-set-prediction approach we have introduced provides the computational basis for an efficient directed-evolution protocol aimed at the simultaneous enhancement of several protein features and should therefore open new possibilities in the engineering of multi-functional enzymes.

Project description:In this study, we propose a visualization technique to explore and visualize concept hierarchies generated from a textbook in the legal domain. Through a human-centered design process, we developed a tool that allows users to effectively navigate through and explore complex hierarchical concepts in three kinds of traversal techniques: top-down, middle-out, and bottom-up. Our concept hierarchies offer an overview over a given domain, with increasing level of detail toward the bottom of the hierarchy which is consisting of entities. In the legal use case we considered, the concepts were adapted from section headings in a legal textbook, whereas references to law or legal cases inside the textbook became entities. The design of this tool is refined following various steps such as gathering user needs, pain points of an existing visualization, prototyping, testing, and refining. The resulting interface offers users several key features such as dynamic search and filter, explorable concept nodes, and a preview of leaf nodes at every stage. A high-fidelity prototype was created to test our theory and design. To test our concept, we used the System Usability Scale as a way to measure the prototype's usability, a task-based survey to asses the tool's ability in assisting users in gathering information and interacting with the prototype, and finally mouse tracking to understand user interaction patterns. Along with this, we gathered audio and video footage of users when participating in the study. This footage also helped us in getting feedback when the survey responses required further information. The data collected provided valuable insights to set the directions for extending this study. As a result, we have accounted for varying hierarchy depths, longer text spans than only one to two words in the elements of the hierarchy, searchability, and exploration of the hierarchies. At the same time, we aimed for minimizing visual clutter and cognitive overload. We show that existing approaches are not suitable to visualize the type of data which we support with our visualization.
