Effective simulations of interacting active droplets.

ABSTRACT: Droplets form a cornerstone of the spatiotemporal organization of biomolecules in cells. These droplets are controlled using physical processes like chemical reactions and imposed gradients, which are costly to simulate using traditional approaches, like solving the Cahn-Hilliard equation. To overcome this challenge, we here present an alternative, efficient method. The main idea is to focus on the relevant degrees of freedom, like droplet positions and sizes. We derive dynamical equations for these quantities using approximate analytical solutions obtained from a sharp interface limit and linearized equations in the bulk phases. We verify our method against fully-resolved simulations and show that it can describe interacting droplets under the influence of chemical reactions and external gradients using only a fraction of the computational costs of traditional methods. Our method can be extended to include other processes in the future and will thus serve as a relevant platform for understanding the dynamics of droplets in cells.

Immersed Boundary Simulations of Active Fluid Droplets.

Project description:We present numerical simulations of active fluid droplets immersed in an external fluid in 2-dimensions using an Immersed Boundary method to simulate the fluid droplet interface as a Lagrangian mesh. We present results from two example systems, firstly an active isotropic fluid boundary consisting of particles that can bind and unbind from the interface and generate surface tension gradients through active contractility. Secondly, a droplet filled with an active polar fluid with homeotropic anchoring at the droplet interface. These two systems demonstrate spontaneous symmetry breaking and steady state dynamics resembling cell motility and division and show complex feedback mechanisms with minimal degrees of freedom. The simulations outlined here will be useful for quantifying the wide range of dynamics observable in these active systems and modelling the effects of confinement in a consistent and adaptable way.

Self-organizing motors divide active liquid droplets.

Project description:The cytoskeleton is a collection of protein assemblies that dynamically impose spatial structure in cells and coordinate processes such as cell division and mechanical regulation. Biopolymer filaments, cross-linking proteins, and enzymatically active motor proteins collectively self-organize into various precise cytoskeletal assemblies critical for specific biological functions. An outstanding question is how the precise spatial organization arises from the component macromolecules. We develop a system to investigate simple physical mechanisms of self-organization in biological assemblies. Using a minimal set of purified proteins, we create droplets of cross-linked biopolymer filaments. Through the addition of enzymatically active motor proteins, we construct composite assemblies, evocative of cellular structures such as spindles, where the inherent anisotropy drives motor self-organization, droplet deformation, and division into two droplets. These results suggest that simple physical principles underlie self-organization in complex biological assemblies and inform bioinspired materials design.

Self-Solidifying Active Droplets Showing Memory-Induced Chirality.

Project description:Most synthetic microswimmers do not reach the autonomy of their biological counterparts in terms of energy supply and diversity of motions. Here, this work reports the first all-aqueous droplet swimmer powered by self-generated polyelectrolyte gradients, which shows memory-induced chirality while self-solidifying. An aqueous solution of surface tension-lowering polyelectrolytes self-solidifies on the surface of acidic water, during which polyelectrolytes are gradually emitted into the surrounding water and induce linear self-propulsion via spontaneous symmetry breaking. The low diffusion coefficient of the polyelectrolytes leads to long-lived chemical trails which cause memory effects that drive a transition from linear to chiral motion without requiring any imposed symmetry breaking. The droplet swimmer is capable of highly efficient removal (up to 85%) of uranium from aqueous solutions within 90 min, benefiting from self-propulsion and flow-induced mixing. These results provide a route to fueling self-propelled agents which can autonomously perform chiral motion and collect toxins.

Rotation and propulsion in 3D active chiral droplets.

Project description:Chirality is a recurrent theme in the study of biological systems, in which active processes are driven by the internal conversion of chemical energy into work. Bacterial flagella, actomyosin filaments, and microtubule bundles are active systems that are also intrinsically chiral. Despite some exploratory attempt to capture the relations between chirality and motility, many features of intrinsically chiral systems still need to be explored and explained. To address this gap in knowledge, here we study the effects of internal active forces and torques on a 3-dimensional (3D) droplet of cholesteric liquid crystal (CLC) embedded in an isotropic liquid. We consider tangential anchoring of the liquid crystal director at the droplet surface. Contrary to what happens in nematics, where moderate extensile activity leads to droplet rotation, cholesteric active droplets exhibit more complex and variegated behaviors. We find that extensile force dipole activity stabilizes complex defect configurations, in which orbiting dynamics couples to thermodynamic chirality to propel screw-like droplet motion. Instead, dipolar torque activity may either tighten or unwind the cholesteric helix and if tuned, can power rotations with an oscillatory angular velocity of 0 mean.

Mesoscopic simulations of active nematics.

Project description:Coarse-grained, mesoscale simulations are invaluable for studying soft condensed matter because of their ability to model systems in which a background solvent plays a substantial role but is not the primary interest. Such methods generally model passive solvents; however, far-from-equilibrium systems may also be composed of complex solutes suspended in an active fluid. Yet, few coarse-grained simulation methods exist to model an active medium. We introduce an algorithm to simulate active nematics, which builds on multiparticle collision dynamics (MPCD) for passive fluctuating nematohydrodynamics by introducing dipolar activity in the local collision operator. Active nematic MPCD (AN-MPCD) simulations not only exhibit the key characteristics of active nematic turbulence but, as a particle-based algorithm, also reproduce crucial attributes of active particle models. Thus, mesoscopic AN-MPCD is an approach that bridges microscopic and continuum descriptions, allowing simulations of composite active-passive systems.

Shaping Liquid Droplets on an Active Air-Ferrofluid Interface.

Project description:An air-liquid interface is important in many biological and industrial applications, where the manipulation of liquids on the air-liquid interface can have a significant impact. However, current manipulation techniques on the interface are mostly limited to transportation and trapping. Here, we report a magnetic liquid shaping method that can squeeze, rotate, and shape nonmagnetic liquids on an air-ferrofluid interface with programmable deformation. We can control the aspect ratio of the ellipse and generate repeatable quasi-static shapes of a hexadecane oil droplet. We can rotate droplets and stir liquids into spiral-like structures. We can also shape phase-changing liquids and fabricate shape-programmed thin films at the air-ferrofluid interface. The proposed method may potentially open up new possibilities for film fabrication, tissue engineering, and biological experiments that can be carried out at an air-liquid interface.

Accurate Measurements and Simulations of the Evaporation and Trajectories of Individual Solution Droplets.

Project description:A refined numerical model for the evaporation and transport of droplets of binary solutions is introduced. Benchmarking is performed against other models found in the literature and experimental measurements of both electrodynamically trapped and freefalling droplets. The model presented represents the microphysical behavior of solutions droplets in the continuum and transition regimes, accounting for the unique hygroscopic behavior of different solutions, including the Fuchs-Sutugin and Cunningham slip correction factors, and accounting for the Kelvin effect. Simulations of pure water evaporation are experimentally validated for temperatures between 290 K and 298 K and between relative humidity values of approximately 0% and 85%. Measurements and simulations of the spatial trajectories and evaporative behavior of aqueous sodium chloride droplets are compared for relative humidity values between 0 and 40%. Simulations are shown to represent experimental data within experimental uncertainty in initial conditions. Calculations of a time-dependent Péclet number, including the temperature dependence of solute diffusion, are related to morphologies of sodium chloride particles dried at different rates. For sodium chloride solutions, dried particles are composed of collections of reproducibly shaped crystals, with higher evaporation rates resulting in higher numbers of crystals, which are smaller.

Topologies of Nanoscale Droplets upon Head-On Collision from Large Molecular Dynamics Simulations.

Project description:The binary collision of nanoscale droplets is studied with molecular dynamics simulation for droplets consisting of up to 2 × 107 molecules interacting via a truncated and shifted form of the Lennard-Jones potential. Considering head-on collisions of droplets with a temperature near the triple point that occur in a saturated vapor of the same fluid, this work explores a range of collision topologies. Four droplet sizes, with a radius ranging from 30 to 120 molecule diameters, are simulated with a varying initial relative collision velocity, covering 36 cases in total. Due to the relatively large size of the droplets, this study aims to resolve the differences in the collision behavior between droplets on the micro- and on the macroscale. By analyzing various metrics of the impact, four distinct collision regimes are found: coalescence, stable collision, holes and shattering. Coalescence, observed at low Weber and Reynolds numbers, is the formation of a stable droplet without significant deformations of the merging objects. Stable collisions, characterized by the formation of one stable droplet with notable deformations during collision, occur within a Weber number range between 10 and 505. The holes regime is only observed for droplet radii greater than 30 molecule diameters and a Weber number between 505 to 750, while collision cases surpassing this Weber number fall into the shattering regime, resulting in the breakup into satellite structures.

Size control and oscillations of active droplets in synthetic cells.

Project description:Oscillations in the formation and dissolution of molecular assemblies inside living cells are pivotal in orchestrating various cellular functions and processes. However, designing such rhythmic patterns in synthetic cells remains a challenge. Here, we demonstrate the spontaneous emergence of spatio-temporal oscillations in the number of droplets, size, and their spatial distribution within a synthetic cell. The coacervate-based droplets in these synthetic cells sediment and fuse at the cell's bottom. Through a size control mechanism, the sedimented, large droplets shrink by expelling droplet material. The expelled molecules nucleate new droplets at the top of the synthetic cell, which grow and sediment again. These oscillations are sustained by converting chemical fuel into waste and can continue for hundreds of periods without evidence of fatigue. Strikingly, the period of the oscillation is in the minute's regime and tunable. The design of oscillating artificial organelles in synthetic cells brings us closer to creating more life-like materials and de novo life.

Entanglement Renyi negativity of interacting fermions from quantum Monte Carlo simulations.

Project description:Many-body entanglement unveils additional aspects of quantum matter and offers insights into strongly correlated physics. While ground-state entanglement has received much attention in the past decade, the study of mixed-state quantum entanglement using negativity in interacting fermionic systems remains largely unexplored. We demonstrate that the partially transposed density matrix of interacting fermions, similar to their reduced density matrix, can be expressed as a weighted sum of Gaussian states describing free fermions, enabling the calculation of rank-n Rényi negativity within the determinant quantum Monte Carlo framework. We calculate the rank-two Rényi negativity for the half-filled Hubbard model and the spinless t-V model. Our calculation reveals that the area law coefficient of the Rényi negativity for the spinless t-V model has a logarithmic finite-size scaling at the finite-temperature transition point. Our work contributes to the calculation of entanglement and sets the stage for future studies on quantum entanglement in various fermionic many-body mixed states.
