Three new species of Macrostomum (Platyhelminthes, Macrostomorpha) from China and Australia, with notes on taxonomy and phylogenetics.

ABSTRACT: In this paper, three species of the macrostomid free-living flatworm genus Macrostomum are described. Two species, Macrostomumlittorale Wang & Shi, sp. nov. and M.shekouense Wang & Shi, sp. nov., were collected from coastal water at Shenzhen, Guangdong Province, China. One species, M.brandi Wang & Shi, sp. nov., was collected from Perth, Western Australia and Queenscliff, Victoria, Australia. Macrostomumlittorale sp. nov. differs from congeneric species within the genus in the length of the stylet, diameter of stylet opening, and the bend of the stylet. Macrostomumshekouense sp. nov. and M.brandi sp. nov. differ from similar species within the genus in the stylet morphology, position of the female antrum and developing eggs, or presence or absence of the false seminal vesicle. Phylogenetic analysis based on cytochrome c oxidase subunit I (COI) gene shows that M.littorale sp. nov. and M.hystrix are sister clades on two well-separated branch, M.shekouense sp. nov. and M.brandi sp. nov. are sister clades on two well-separated branches. Accordingly, both morphological and molecular evidence support M.littorale sp. nov., M.shekouense sp. nov., and M.brandi sp. nov. as three new species.

Project description:BackgroundFreshwater shrimp of the family Atyidae De Haan, 1849 have been studied in Vietnam for more than a century. A total of 24 species of atyid shrimps from the genera Caridina H. Milne Edwards, 1837, Neocaridina Kubo, 1938, Atyopsis Chace, 1983 have been recorded from Vietnam. With 22 species, the majority are from the genus Caridina. In 2013, Karge and Klotz mentioned the occurrence of four yet undescribed species belonging to Paracaridina Liang, Guo & Tang, 1999 in Vietnam without taxonomic details.In general, studies of freshwater atyids in Vietnam are limited and most Vietnamese taxa await a taxonomic revision. The available data do not fully reflect their estimated species diversity in the country and distribution data are deficient. Here, we focus on four species of atyid shrimps from two genera, viz. Caridinacantonensis Yu, 1938, C.lanceifrons Yu, 1936, C.serrata Stimpson, 1860 and Neocaridinapalmata (Shen, 1948), all described from China and have been reported to occur in Vietnam. The previous reports on the occurrence of these species in Vietnam are largely unreliable due to taxonomic confusion. To contribute to the knowledge of these taxa, we provide the first verified distribution records of the four species in the country with some taxonomic remarks.New informationThis study shows the first taxonomically-verified distribution data of four atyid shrimp species originally described from China, but also reported from Vietnam, albeit under various species names and, in some cases, erroneously. These data allow the first meaningful discussion of the distribution in light of the reproductive strategy of these four species and, in conjunction with the taxonomic remarks, will contribute to the knowledge of these taxa. As a result of this research and data from previous studies, we now consider all four species as widespread and non-endemic, but land-locked (with a complete freshwater life cycle). In Vietnam, all four species are confined to the northern half of the country. Beyond Vietnam, we provide the first records for Caridinalanceifrons from southern Thailand, which suggests a major sampling gap in Indochina.

Project description:Knowledge of pathogen biology and genetic diversity is a cornerstone of effective disease management, and accurate identification of the pathogen is a foundation of pathogen biology. Species names provide an ideal framework for storage and retrieval of relevant information, a system that is contingent on a clear understanding of species boundaries and consistent species identification. Verticillium, a genus of ascomycete fungi, contains important plant pathogens whose species boundaries have been ill defined. Using phylogenetic analyses, morphological investigations and comparisons to herbarium material and the literature, we established a taxonomic framework for Verticillium comprising ten species, five of which are new to science. We used a collection of 74 isolates representing much of the diversity of Verticillium, and phylogenetic analyses based on the ribosomal internal transcribed spacer region (ITS), partial sequences of the protein coding genes actin (ACT), elongation factor 1-alpha (EF), glyceraldehyde-3-phosphate dehydrogenase (GPD) and tryptophan synthase (TS). Combined analyses of the ACT, EF, GPD and TS datasets recognized two major groups within Verticillium, Clade Flavexudans and Clade Flavnonexudans, reflecting the respective production and absence of yellow hyphal pigments. Clade Flavexudans comprised V. albo-atrum and V. tricorpus as well as the new species V. zaregamsianum, V. isaacii and V. klebahnii, of which the latter two were morphologically indistinguishable from V. tricorpus but may differ in pathogenicity. Clade Flavnonexudans comprised V. nubilum, V. dahliae and V. longisporum, as well as the two new species V. alfalfae and V. nonalfalfae, which resembled the distantly related V. albo-atrum in morphology. Apart from the diploid hybrid V. longisporum, each of the ten species corresponded to a single clade in the phylogenetic tree comprising just one ex-type strain, thereby establishing a direct link to a name tied to a herbarium specimen. A morphology-based key is provided for identification to species or species groups.
