ABSTRACT: The genome of an antibiotic-producing bacterium, Bacillus velezensis H208, was sequenced. Strain H208 was isolated from ginger rhizosphere in Laifeng County, China. The genome consisted of 3,929,792 bp, with a GC content of 46.5%, and contained 3,773 protein-coding genes and 118 noncoding RNA genes.

The complete genome sequence of Bacillus velezensis LPL061, an exopolysaccharide-producing bacterium.

Project description:Bacillus velezensis LPL061, which shows strong exopolysaccharide (EPS) producing capacity, was isolated from carnations in Beijing, China. The complete genome of LPL061 comprised a single circular chromosome (3,907,268 bp; G+C content of 46.7%) with 3,737 coding DNA sequences, 26 rRNA, and 89 tRNA. According to genome analysis, 12 protein-coding genes which related to polysaccharide biosynthesis in LPL061 were identified. Comparative genome analysis revealed that the EPS biosynthetic gene cluster was relatively conserved among Bacillus species. EPS showed approximately 60% inhibitory activity on the α-glucosidase at 100 μg/mL. The results of quantitative reverse transcription PCR further demonstrated that compared to insulin-resistant model with insulin (500 μg/mL) (without EPS treatment), the insulin-resistant HepG2 cells treated with EPS decreased the expression of phosphoenolpyruvate carboxykinase (PEPCK) from 4.425 to 0.1587, glucose-6-phosphatase (G6Pase) decreased from 4.272 to 0.1929, and glycogen synthase kinase3β (GSK(3)β) decreased from 2.451 to 0.993, respectively. Meanwhile, EPS treatment increased GS expression and resulted in intracellular glycogen concentration increased from 28.30% to 86.48%, which further supported that EPS form LPL061 could reduce the concentration of blood glucose effectively. These results could be beneficial for better understanding of the hypoglycemic mechanism of B. velezensis LPL061 EPS and developing an EPS-based anti-diabetic agent in the future.

Bacillus paralicheniformis SYN-191 isolated from ginger rhizosphere soil and its growth-promoting effects in ginger farming.

Project description:BackgroundThe use of chemical fertilizers and pesticides and the farming without crop rotation may negatively impact the microbial community and the quality of the soils in ginger farm. It is important to improve soil properties to promote the healthy growth of ginger in ginger farm.ResultsWe isolated and identified the pathogenic Fusarium ramigenum from infected ginger roots. We then isolated a new Bacillus paralicheniformis strain SYN-191 from the rhizosphere soil around healthy ginger roots, and showed B. paralicheniformis SYN-91 could inhibit F. ramigenum growth, degrade proteins, dissolve silicate, and decompose cellulose. SYN-191 treatment significantly improved the agronomic traits of ginger seedlings in healthy soil and continuous cropping soil. Furthermore, SYN-191 treatment restructured the microbial microbiomes in rhizosphere soil, including reducing the number of harmful fungi, such as Fusarium, and increasing the beneficial bacterial populations such as Bacillus and Pseudomonas. Field experiments showed that SYN-191 application increased ginger yield by 26.47% (P < 0.01). Whole-genome sequencing of strain SYN-191 revealed the relevant genes for antibiotic synthesis, potassium dissolution, and cellulose decomposition.ConclusionsA new plant-growth-promoting B. paralicheniformis SYN-191 was obtained. This strain could antagonize ginger root rot pathogenic fungus, improve agronomic traits and ginger yield in field, and improve the microbial community structure in the ginger rhizosphere soil. This study provides a valuable bacterial resource for overcoming obstacles in the continuous cropping of ginger.

Here, we present a complete circular genome sequence and a plasmid of an antimicrobial-producing isolate, Bacillus velezensis strain Sam8H1, from the Makgadikgadi saltpans in Botswana. Bioinformatic analyses revealed 12 putative secondary metabolite biosynthetic gene clusters important for genome-guided drug discovery studies.

Project description:We report the complete genome sequence and annotation of Bacillus velezensis GMEKP1, which was isolated from a hive of stingless bees (Trigona laeviceps). This bacterium has a circular 4,014,839-nucleotide chromosome and two circular plasmids. Genome-mining analysis of the whole-genome sequence revealed that GMEKP1 has 12 biosynthetic gene clusters, dominated by genes encoding polyketide synthase hybrids.
