Single-cell RNA sequencing reveals the suppressive effect of PPP1R15A inhibitor Sephin1 in antitumor immunity.

ABSTRACT: Protein phosphatase 1 regulatory subunit 15A (PPP1R15A) is an important factor in the integrated stress response (ISR) in mammals and may play a crucial role in tumorigenesis. In our studies, we found an inhibitor of PPP1R15A, Sephin1, plays a protumorigenic role in mouse tumor models. By analyzing the single-cell transcriptome data of the mouse tumor models, we found that in C57BL/6 mice, Sephin1 treatment could lead to higher levels of ISR activity and lower levels of antitumor immune activities. Specifically, Sephin1 treatment caused reductions in antitumor immune cell types and lower expression levels of cytotoxicity-related genes. In addition, T cell receptor (TCR) repertoire analysis demonstrated that the clonal expansion of tumor-specific T cells was inhibited by Sephin1. A special TCR + macrophage subtype in tumor was identified to be significantly depleted upon Sephin1 treatment, implying its key antitumor role. These results suggest that PPP1R15A has the potential to be an effective target for tumor therapy.

Project description:Sephin1 is the selective inhibitor of Protein Phosphatase 1 Regulatory Subunit 15A (PPP1R15A), which is an important factor in the integrated stress response (ISR) of mammalian cells. Sephin1 can promote ISR by inhibiting the dephosphorylation of eIF2α, and preventing neurodegenerative disease occurrence in mice. However, we found that Sephin1 plays a protumorigenic role in mouse tumor models by suppressing the antitumor immune activities, which was highly relevant to the function of PPP1R15A. To fully understand the mechanism of Sephin1 to the antitumor immunity, we designed this experiment to explore the function of PPP1R15A and Sephin1 in the single-cell expression level. To understand effect of injection of Sephin1 to the immune cells in blood and tumor microenvironment in mice, we injected Sephin1 to mice intraperitoneally, and then subcutaneously inoculated with B16F1 cells. Blood before/after the tumor development and tumor tissues were collected and immune cells were isolated for single-cell sequencing and bioinformatics analysis afterwards. We found that in C57BL/6 mice, Sephin1 treatment could lead to higher levels of ISR activity and lower levels of antitumor immune activities. Specifically, Sephin1 treatment caused reductions in several tumor-killing immune cell populations, including CD8+ T cells, NK cells and NKT cells. The expression levels of cytotoxicity-related genes in lymphocytes were altered upon Sephin1 treatment. Additionally, TCR repertoire analysis demonstrated that the clonal expansion of tumor-specific T cells was inhibited by Sephin1, resulting in a compromised antitumor immune response. A special TCR+ macrophage subtype was identified to be significantly depleted upon Sephin1 treatment, implying that this type of macrophage plays a key antitumor role in the tumor microenvironment. The effects of Sephin1 were also verified in in vitro experiments and studies of other tumor cell lines. Taken together, these data suggest that inhibiting PPP1R15A promotes tumorigenesis by inducing adverse immune suppression. PPP1R15A has the potential to be an effective target for tumor therapy. Besides, a macrophage subtype, the TCR+ macrophages, played an important role in the anti-tumor immunity and can also be significantly inhibited by Sephin1.

Project description:BackgroundImmune checkpoint inhibitors (ICIs) have altered the clinical management of non-small cell lung cancer (NSCLC). However, the low response rate, severe immune-related adverse events (irAEs), and hyperprogressive disease following ICIs monotherapy require attention. Combination therapy may overcome these limitations and traditional Chinese medicine with immunomodulatory effects provides a promising approach. Shenmai injection (SMI) is a clinically effective adjuvant treatment for cancer with chemotherapy and radiotherapy. Therefore, the combined effects and mechanisms of SMI and programmed death-1 (PD-1) inhibitor against NSCLC was focused on this study.MethodsA Lewis lung carcinoma mouse model and a lung squamous cell carcinoma humanized mouse model were used to investigate the combined efficacy and safety of SMI and PD-1 inhibitor. The synergistic mechanisms of the combination therapy against NSCLC were explored using single-cell RNA sequencing. Validation experiments were performed using immunofluorescence analysis, in vitro experiment, and bulk transcriptomic datasets.ResultsIn both models, combination therapy alleviated tumor growth and prolonged survival without increasing irAEs. The GZMAhigh and XCL1high natural killer (NK) cell subclusters with cytotoxic and chemokine signatures increased in the combination therapy, while malignant cells from combination therapy were mainly in the apoptotic state, suggesting that mediating tumor cell apoptosis through NK cells is the main synergistic mechanisms of combination therapy. In vitro experiment confirmed that combination therapy increased secretion of Granzyme A by NK cells. Moreover, we discovered that PD-1 inhibitor and SMI combination blocked inhibitory receptors on NK and T cells and restores their antitumoral activity in NSCLC better than PD-1 inhibitor monotherapy, and immune and stromal cells exhibited a decrease of angiogenic features and attenuated cancer metabolism reprogramming in microenvironment of combination therapy.ConclusionsThis study demonstrated that SMI reprograms tumor immune microenvironment mainly by inducing NK cells infiltration and synergizes with PD-1 inhibitor against NSCLC, suggested that targeting NK cells may be an important strategy for combining with ICIs.
