Project description:Lanthanide-doped (nano)crystals are an important class of materials in luminescence thermometry. The working mechanism of these thermometers is diverse but most often relies on variation of the ratio of emission intensities from two thermally coupled excited states with temperature. At low temperatures, nonradiative coupling between the states can be slow compared to radiative decay, but, at higher temperatures, the two states reach thermal equilibrium due to faster nonradiative coupling. In thermal equilibrium, the intensity ratio follows Boltzmann statistics, which gives a convenient model to calibrate the thermometer. Here, we investigate multiple strategies to shift the onset of thermal equilibrium to lower temperatures, which enables Boltzmann thermometry in a wider dynamic range. We use Eu3+-doped microcrystals as a model system and find that the nonradiative coupling rates increase for host lattices with higher vibrational energies and shorter lanthanide-ligand distances, which reduces the onset temperature of thermal equilibrium by more than 400 K. We additionally reveal that thermometers with excited states coupled by electric-dipole transitions have lower onset temperatures than those with magnetic-dipole-coupled states due to selection rules. These insights provide essential guidelines for the optimization of Boltzmann thermometers to operate in an extended temperature range.

Project description:Similar to other cropping systems, few walnut cultivars are used as scion in commercial production. Germplasm collections can be used to diversify cultivar options and hold potential for improving crop productivity, disease resistance and stress tolerance. In this study, we explored the anatomical and biochemical bases of photosynthetic capacity and response to water stress in 11 Juglans regia accessions in the U.S. department of agriculture, agricultural research service (USDA-ARS) National Clonal Germplasm. Net assimilation rate (An ) differed significantly among accessions and was greater in lower latitudes coincident with higher stomatal and mesophyll conductances, leaf thickness, mesophyll porosity, gas-phase diffusion, leaf nitrogen and lower leaf mass and stomatal density. High CO2 -saturated assimilation rates led to increases in An under diffusional and biochemical limitations. Greater An was found in lower-latitude accessions native to climates with more frost-free days, greater precipitation seasonality and lower temperature seasonality. As expected, water stress consistently impaired photosynthesis with the highest % reductions in lower-latitude accessions (A3, A5 and A9), which had the highest An under well-watered conditions. However, An for A3 and A5 remained among the highest under dehydration. J. regia accessions, which have leaf structural traits and biochemistry that enhance photosynthesis, could be used as commercial scions or breeding parents to enhance productivity.

Project description:Extensive research shows temperature to be the primary environmental factor controlling the phyllochron, or rate of leaf appearance, of wheat (Triticum aestivum L.). Experimental results suggest that soil temperature at crown depth, rather than air temperature above the canopy, would better predict wheat leaf appearance rates. To test this hypothesis, leaf appearance in spring wheat ('Nordic') was measured in a 2-year field experiment (Nunn clay loam soil; fine, smectitic, mesic Aridic, Argiustoll) with three planting dates and two soil temperature treatments. One temperature treatment (denoted +3C) consisted of heating the soil at crown depth to 3 degrees C above the ambient soil temperature (denoted +0C). Main stem cumulative leaf number was measured at least weekly until flag leaf emergence. Leaf appearance was essentially linear with both air and soil growing degree-days (GDD), although there was a stronger linear relationship with soil GDD in the +0C plants than in +3C plants. A weak positive relationship between planting date and the phyllochron was observed. Unexpectedly, we found that heating the soil did not increase the rate of leaf appearance, as the paradigm would predict. To explain these results, we propose extending the paradigm in two ways. First, three processes are involved in leaf appearance: (1) cell division at the shoot apex forms the primordium; (2) cell division in the intercalary meristem forms the cells that then (3) expand to produce the leaf. Cell division is predominantly controlled by temperature, but cell expansion is considerably more affected by factors other than temperature, explaining the influence of other factors on the phyllochron. Secondly, the vertical distribution of the two meristems and region of cell expansion occur over a significant distance, where temperature varies considerably, and temperature at a specific point (e.g. crown depth) does not account for the entire temperature regime under which leaves are developing.

Project description:The pattern of cell division, growth and separation during leaf development determines the pattern and volume of airspace in a leaf. The resulting balance of cellular material and airspace is expected to significantly influence the primary function of the leaf, photosynthesis, and yet the manner and degree to which cell division patterns affect airspace networks and photosynthesis remains largely unexplored. In this paper we investigate the relationship of cell size and patterning, airspace and photosynthesis by promoting and repressing the expression of cell cycle genes in the leaf mesophyll. Using microCT imaging to quantify leaf cellular architecture and fluorescence/gas exchange analysis to measure leaf function, we show that increased cell density in the mesophyll of Arabidopsis can be used to increase leaf photosynthetic capacity. Our analysis suggests that this occurs both by increasing tissue density (decreasing the relative volume of airspace) and by altering the pattern of airspace distribution within the leaf. Our results indicate that cell division patterns influence the photosynthetic performance of a leaf, and that it is possible to engineer improved photosynthesis via this approach.

Project description:The aim of this study was to systematically analyze the potential and limitations of using plant functional trait observations from global databases versus in situ data to improve our understanding of vegetation impacts on ecosystem functional properties (EFPs). Using ecosystem photosynthetic capacity as an example, we first provide an objective approach to derive robust EFP estimates from gross primary productivity (GPP) obtained from eddy covariance flux measurements. Second, we investigate the impact of synchronizing EFPs and plant functional traits in time and space to evaluate their relationships, and the extent to which we can benefit from global plant trait databases to explain the variability of ecosystem photosynthetic capacity. Finally, we identify a set of plant functional traits controlling ecosystem photosynthetic capacity at selected sites. Suitable estimates of the ecosystem photosynthetic capacity can be derived from light response curve of GPP responding to radiation (photosynthetically active radiation or absorbed photosynthetically active radiation). Although the effect of climate is minimized in these calculations, the estimates indicate substantial interannual variation of the photosynthetic capacity, even after removing site-years with confounding factors like disturbance such as fire events. The relationships between foliar nitrogen concentration and ecosystem photosynthetic capacity are tighter when both of the measurements are synchronized in space and time. When using multiple plant traits simultaneously as predictors for ecosystem photosynthetic capacity variation, the combination of leaf carbon to nitrogen ratio with leaf phosphorus content explains the variance of ecosystem photosynthetic capacity best (adjusted R2 = 0.55). Overall, this study provides an objective approach to identify links between leaf level traits and canopy level processes and highlights the relevance of the dynamic nature of ecosystems. Synchronizing measurements of eddy covariance fluxes and plant traits in time and space is shown to be highly relevant to better understand the importance of intra- and interspecific trait variation on ecosystem functioning.

Project description:Storage nitrogen (N) is a buffer pool for maintaining leaf growth and synthesizing photosynthetic proteins, but the dynamics of its forms within the life cycle of a single leaf and how it is influenced by N supply remain poorly understood. A field experiment was conducted to estimate the influence of N supply on leaf growth, photosynthetic characteristics, and N partitioning inthe sixth leaf of winter oilseed rape (Brassica napus L.) from emergence through senescence. Storage N content (Nstore) decreased gradually along with leaf expansion. The relative growth rate based on leaf area (RGRa) was positively correlated with Nstore during leaf expansion. The water-soluble protein form of storage N was the main N source for leaf expansion. After the leaves fully expanded, the net photosynthetic rate (An) followed a linear-plateau response to Nstore, with An stabilizing at the highest value above a threshold and declining below the threshold. Non-protein and SDS (detergent)-soluble protein forms of storage N were the main N sources for maintaining photosynthesis. For the leaf N economy, storage N is used for co-ordinating leaf expansion and photosynthetic capacity. N supply can improve Nstore, thereby promoting leaf growth and biomass.

Project description:Ferns usually have relatively lower photosynthetic potential than angiosperms. However, it is unclear whether low photosynthetic potential of ferns is linked to leaf water supply. We hypothesized that there is an evolutionary association of leaf water transport capacity with photosynthesis and stomatal density in ferns. In the present study, a series of functional traits relating to leaf anatomy, hydraulics and physiology were assessed in 19 terrestrial and 11 epiphytic ferns in a common garden, and analyzed by a comparative phylogenetics method. Compared with epiphytic ferns, terrestrial ferns had higher vein density (Dvein), stomatal density (SD), stomatal conductance (gs), and photosynthetic capacity (Amax), but lower values for lower epidermal thickness (LET) and leaf thickness (LT). Across species, all traits varied significantly, but only stomatal length (SL) showed strong phylogenetic conservatism. Amax was positively correlated with Dvein and gs with and without phylogenetic corrections. SD correlated positively with Amax, Dvein and gs, with the correlation between SD and Dvein being significant after phylogenetic correction. Leaf water content showed significant correlations with LET, LT, and mesophyll thickness. Our results provide evidence that Amax of the studied ferns is linked to leaf water transport capacity, and there was an evolutionary association between water supply and demand in ferns. These findings add new insights into the evolutionary correlations among traits involving carbon and water economy in ferns.

Project description:Researchers from a number of disciplines have long sought the ability to estimate the functional attributes of plant canopies, such as photosynthetic capacity, using remotely sensed data. To date, however, this goal has not been fully realized. In this study, fresh-leaf reflectance spectroscopy (λ=450-2500 nm) and a partial least-squares regression (PLSR) analysis were used to estimate key determinants of photosynthetic capacity-namely the maximum rates of RuBP carboxylation (V(cmax)) and regeneration (J(max))-measured with standard gas exchange techniques on leaves of trembling aspen and eastern cottonwood trees. The trees were grown across an array of glasshouse temperature regimes. The PLSR models yielded accurate and precise estimates of V(cmax) and J(max) within and across species and glasshouse temperatures. These predictions were developed using unique contributions from different spectral regions. Most of the wavelengths selected were correlated with known absorption features related to leaf water content, nitrogen concentration, internal structure, and/or photosynthetic enzymes. In a field application of our PLSR models, spectral reflectance data effectively captured the short-term temperature sensitivities of V(cmax) and J(max) in aspen foliage. These findings highlight a promising strategy for developing remote sensing methods to characterize dynamic, environmentally sensitive aspects of canopy photosynthetic metabolism at broad scales.
