Benthic biota of Chilean fjords and channels in 25 years of cruises of the National Oceanographic Committee.

ABSTRACT: The CIMAR program (Marine Research Cruises to Remote Areas) run by the Chilean Navy through the National Oceanographic Committee has been developed since 1995, focused on Chilean fjord and channel zones (~41-56°S; "CIMAR-Fjords") and Chilean remote islands ("CIMAR-Islands"). Samples and data was collected on biotic and abiotic variables on all these cruises, both from the water column and benthos. Our work standardizes, compiles, and summarizes the published information on benthic organisms for twenty-one CIMAR-Fjords cruises developed in the first 25 years of the program, plus the Southern Ice Fields Cruise 1995 (precursor of the CIMAR program), which includes the distribution, abundance and geographic location of cruises sampling stations. The data set includes 8,854 records from 880 different localities, corresponding to 1,225 species from 24 different phyla (four kingdoms) and more than 150,000 individuals. Only two cruises did not record any benthic sampling. The fjords and channels of Chilean Patagonia have high biodiversity, so we hope that our data set will serve as a baseline for ecological studies and ecosystem conservation.

Project description:To protect and restore ecosystems and biodiversity is one of the 10 challenges identified by the United Nations's Decade of the Ocean Science. In this study we used eDNA from sediments collected in two fjords of the Svalbard archipelago and compared the taxonomic composition with traditional methods through metabarcoding, targeting mitochondrial CO1, to survey benthos. Clustering of 21.6 mill sequence reads with a d value of 13 in swarm, returned about 25 K OTU reads. An identification search with the BOLD database returned 12,000 taxonomy annotated sequences spanning a similarity range of 50% to 100%. Using an acceptance filter of minimum 90% similarity to the CO1 reference sequence, we found that 74% of the ca 100 taxon identified sequence reads were Polychaeta and 22% Nematoda. Relatively few other benthic invertebrate species were detected. Many of the identified sequence reads were extra-organismal DNA from terrestrial, planktonic, and photic zone sources. For the species rich Polychaeta, we found that, on average, only 20.6% of the species identified from morphology were also detected with DNA. This discrepancy was not due to missing reference sequences in the search database, because 90-100% (mean 96.7%) of the visually identified species at each station were represented with barcodes in Boldsystems. The volume of DNA samples is small compared with the volume searched in visual sorting, and the replicate DNA-samples in sum covered only about 2% of the surface area of a grab. This may considerably reduce the detection rate of species that are not uniformly distributed in the sediments. Along with PCR amplification bias and primer mismatch, this may be an important reason for the limited congruence of species identified with the two approaches. However, metabarcoding also identified 69 additional species that are usually overlooked in visual sample sorting, demonstrating how metabarcoding can complement traditional methodology by detecting additional, less conspicuous groups of organisms.

Project description:Fjord ecosystems cycle and export significant amounts of carbon and appear to be extremely sensitive to climate change and anthropogenic perturbations. To identify patterns of microbial responses to ongoing natural and human-derived changes in the fjords of Chilean Patagonia, we examined the effect of organic enrichment associated with salmon aquaculture and freshening produced by glacial melting on bacterial production (BP), extracellular enzymatic activity (EEA), and community diversity of free-living bacterioplankton. We assayed the effects of salmon food-derived dissolved organic matter (SF-DOM) and meltwaters through microcosm experiments containing waters from Puyuhuapi Fjord and the proglacial fjords of the Southern Patagonia Icefield, respectively. Rates of BP and EEA were 2 times higher in the presence of SF-DOM than in controls, whereas the addition of autochthonous organic matter derived from diatoms (D-DOM) resulted in rates of BP and EEA similar to those measured in the controls. The addition of SF-DOM also reduced species richness and abundance of a significant fraction of the representative taxa of bacterioplankton of Puyuhuapi Fjord. In the proglacial fjords, bacterioplankton diversity was reduced in areas more heavily influenced by meltwaters and was accompanied by moderate positive changes in BP and EEA. Our findings strongly suggest that SF-DOM is highly reactive, promoting enhanced rates of microbial activity while could be influencing the diversity of bacterioplankton communities in Patagonian fjords with a strong salmon farming activity. These findings challenge the traditional view of phytoplankton production as the primary source of labile DOM that fuels heterotrophic activity in coastal ecosystems impacted by anthropogenic organic enrichment. Given the intensive local production of salmon, we analyze the significance of this emerging source of rich "allochthonous" organic substrates for autotrophic/heterotrophic balance, carbon exportation, and hypoxia in Patagonian fjords. The effect of human DOM enrichment can be enhanced in proglacial fjords, where progressive glacial melting exerts additional selective pressure on bacterioplankton diversity.

Project description:Greenhouse gas emissions from land-use change, fossil fuel, agriculture, transportation, and electricity sectors expose marine ecosystems to overlapping environmental stressors. Existing climate vulnerability assessment methods analyze the frequency of extreme conditions but often minimally consider how environmental data gaps hinder assessments. Here, we show an approach that assesses vulnerability and the uncertainty introduced by monitoring data gaps, using a case study of ocean acidification and deoxygenation in coastal California. We employ 5 million publicly available oceanographic observations and existing studies on species responses to low pH, low oxygen conditions to calculate vulnerability for six ecologically and economically valuable benthic invertebrate species: red sea urchin (Mesocentrotus franciscanus), purple sea urchin (Strongylocentrotus purpurpatus), warty sea cucumber (Apostichopus parvimensis), pink shrimp (Pandalus jordani), California spiny lobster (Panulirus interruptus), and Dungeness crab (Metacarncinus magister). Further, we evaluate the efficacy of current monitoring programs by examining how data gaps heighten associated uncertainty. We find that most organisms experience low oxygen (<35% saturation) conditions less frequently than low pH ( < 7.6) conditions. It is only deeper dwelling (>75 m depth) life stages such as Dungeness crab adults and pink shrimp embryos, juveniles, and adults that experience more frequent exposure to low oxygen conditions. Adult Dungeness crabs experience the strongest seasonal variation in exposure. Though these trends are intriguing, exposure remains low for most species despite well-documented pH and oxygen declines and strengthening upwelling in the central portions of the California Current. Seasonal biases of data collection and sparse observations near the benthos and at depths where organisms most frequently experience stressful conditions undermine exposure estimates. Herein we provide concrete examples of how pink shrimp and Dungeness crab fisheries may be impacted by our findings, and provide suggestions for incorporating oceanographic data into management plans. By limiting our scope to California waters and assessing the limitations presented by current monitoring coverage, this study aims to provide a granular, actionable framework that policymakers and managers can build from to prioritize targeted enhancements and sustained funding of oceanographic monitoring recommendations.
