Methyl 1-[(6-meth-oxy-5-methyl-pyrimidin-4-yl)meth-yl]-1H-benzo[d]imidazole-7-carboxyl-ate: a combined X-ray and DFT study.

ABSTRACT: The title compound, C₁₆H₁₆N₄O₃, was obtained as a side product during the synthesis of the previously reported anti-tubercular agent N-(2-fluoro-eth-yl)-1-[(6-meth-oxy-5-methyl-pyrimidin-4-yl)meth-yl]-1H-benzo[d]imidazole-4-carboxamide and structurally characterized by X-ray crystallography and computational methods. In the crystal (space group P2₁/n, Z = 4), the title compound adopts a twisted conformation with a dihedral angle between the benzimidazole and pyrimidine mean planes of 84.11 (3)°. The carboxyl-ate group and the 5-methyl group on the pyrimidine ring exhibit partial disorder. The DFT-optimized mol-ecular structure resembles the structure of the minor component in the crystal.

Project description:Herein, a series of N-1-sulfonyl substituted derivatives of 2-substituted benzimidazoles (2a-2e) were designed and synthesized via structural tailoring of the acceptor part of donor-π-acceptor schemes, and their nonlinear optic (NLO) characteristics were reported. The structures of 2a-2e were investigated and their characterization was accomplished by employing spectroscopic procedures, i.e., UV-vis, FT-IR, and 1H and 13C NMR. Further, a density functional theory (DFT) approach was used to calculate UV-vis, vibrational, and 1H and 13C NMR techniques; frontier molecular orbitals (FMOs); global reactivity parameters (GRPs); natural bond orbitals (NBOs); optical and vibrational analysis; and nonlinear optics (NLO). The most promising results were obtained for 6-nitro-2-(4-nitrophenyl)-1-(4-nitrophenylsulfonyl)-1H-benzo[d]imidazole among entitled compounds, as it exhibited the highest ⟨α⟩ and βtot values, showing it is an eye-catching NLO material. This DFT study evokes the interest of researchers regarding the development of benzimidazole-based tempting NLO compounds that could be beneficial in modern hi-tech applications.

Project description:The title compound, C(22)H(20)N(2), (Ib), forms along with 2-[(cyclo-penta-1,3-dien-1-yl)diphenyl-meth-yl]-1-methyl-1H-imid-azole, (Ia), which differs with respect to the position of the double-bonds in the C(5)H(5) ring, in an approximately 3:7 ratio (Ia:Ib; NMR spectroscopy data). However, in a single crystal, only compound (Ib) is present. H atoms of the CH(2) group (C(5)H(5) ring) were found from the difference Fourier synthesis and refined isotropically using the riding model. Hypothesis on possible presence of the (Ia) isomer in crystal lattice (model with a C(5)H(5) ring disordered between two positions) was especially checked and rejected due to its inconsistency. In the crystal structure, no significant hydrogen-bonding inter-actions between the CH(2) groups of the C(5)H(5) rings and nonsubstituted N-atoms of the imidazole rings were observed. Despite the fact that the chemically achiral compound (I) crystallizes in a chiral space group P2(1)2(1)2(1), neither the absolute structure determination nor assignment of the inversion twinning was possible in the absence of a heavy atom.
