Dynamic Roles of Insect Carboxyl/Cholinesterases in Chemical Adaptation.

ABSTRACT: Insects have evolved several intricate defense mechanisms to adapt to their chemical environment. Due to their versatile capabilities in hydrolytic biotransformation, insect carboxyl/cholinesterases (CCEs) play vital roles in the development of pesticide resistance, facilitating the adaptation of insects to their host plants, and manipulating insect behaviors through the olfaction system. CCEs confer insecticide resistance through the mechanisms of qualitative or quantitative changes of CCE-mediated enhanced metabolism or target-site insensitivity, and may contribute to the host plant adaptation. CCEs represent the first odorant-degrading enzymes (ODEs) discovered to degrade insect pheromones and plant odors and remain the most promising ODE candidates. Here, we summarize insect CCE classification, currently characterized insect CCE protein structure characteristics, and the dynamic roles of insect CCEs in chemical adaptation.

Roles of insect odorant binding proteins in communication and xenobiotic adaptation.

Project description:Odorant binding proteins (OBPs) are small water-soluble proteins mainly associated with olfaction, facilitating the transport of odorant molecules to their relevant receptors in the sensillum lymph. While traditionally considered essential for olfaction, recent research has revealed that OBPs are engaged in a diverse range of physiological functions in modulating chemical communication and defense. Over the past 10 years, emerging evidence suggests that OBPs play vital roles in purifying the perireceptor space from unwanted xenobiotics including plant volatiles and pesticides, potentially facilitating xenobiotic adaptation, such as host location, adaptation, and pesticide resistance. This multifunctionality can be attributed, in part, to their structural variability and effectiveness in transporting, sequestering, and concealing numerous hydrophobic molecules. Here, we firstly overviewed the classification and structural properties of OBPs in diverse insect orders. Subsequently, we discussed the myriad of functional roles of insect OBPs in communication and their adaptation to xenobiotics. By synthesizing the current knowledge in this field, our review paper contributes to a comprehensive understanding of the significance of insect OBPs in chemical ecology, xenobiotic adaptation, paving the way for future research in this fascinating area of study.

Evidence for inhibition of cholinesterases in insect and mammalian nervous systems by the insect repellent deet.

Project description:BACKGROUND: N,N-Diethyl-3-methylbenzamide (deet) remains the gold standard for insect repellents. About 200 million people use it every year and over 8 billion doses have been applied over the past 50 years. Despite the widespread and increased interest in the use of deet in public health programmes, controversies remain concerning both the identification of its target sites at the olfactory system and its mechanism of toxicity in insects, mammals and humans. Here, we investigated the molecular target site for deet and the consequences of its interactions with carbamate insecticides on the cholinergic system. RESULTS: By using toxicological, biochemical and electrophysiological techniques, we show that deet is not simply a behaviour-modifying chemical but that it also inhibits cholinesterase activity, in both insect and mammalian neuronal preparations. Deet is commonly used in combination with insecticides and we show that deet has the capacity to strengthen the toxicity of carbamates, a class of insecticides known to block acetylcholinesterase. CONCLUSION: These findings question the safety of deet, particularly in combination with other chemicals, and they highlight the importance of a multidisciplinary approach to the development of safer insect repellents for use in public health.

Dynamic interchange between two protonation states is characteristic of active sites of cholinesterases.

Project description:Cholinesterases are well-known and widely studied enzymes crucial to human health and involved in neurology, Alzheimer's, and lipid metabolism. The protonation pattern of active sites of cholinesterases influences all the chemical processes within, including reaction, covalent inhibition by nerve agents, and reactivation. Despite its significance, our comprehension of the fine structure of cholinesterases remains limited. In this study, we employed enhanced-sampling quantum-mechanical/molecular-mechanical calculations to show that cholinesterases predominantly operate as dynamic mixtures of two protonation states. The proton transfer between two non-catalytic glutamate residues follows the Grotthuss mechanism facilitated by a mediator water molecule. We show that this uncovered complexity of active sites presents a challenge for classical molecular dynamics simulations and calls for special treatment. The calculated proton transfer barrier of 1.65 kcal/mol initiates a discussion on the potential existence of two coupled low-barrier hydrogen bonds in the inhibited form of butyrylcholinesterase. These findings expand our understanding of structural features expressed by highly evolved enzymes and guide future advances in cholinesterase-related protein and drug design studies.

Environmental adaptation in an asexual invasive insect.

Project description:Parthenogenetic reproduction is generally associated with low genetic variance and therefore reduced ability for environmental adaptation, and this could limit the potential invasiveness of introduced species that reproduce asexually. However, the hemlock woolly adelgid is an asexual invasive insect that has spread across a large geographic temperature gradient in its introduced range. Consequently, this insect has shown significant variation in cold hardiness among populations. We hypothesized that the increased cold hardiness of northern populations represents an adaptation to the colder temperatures. To test this, we collected individual adelgid from populations spanning their invaded range and inoculated them into a common thermal environment. We then experimentally sampled the supercooling point of the progeny of these adelgids and compared these results with tests of the supercooling point of adelgid sampled directly from their source populations. The results showed that the same significant differences in supercooling that was found among geographically distinct populations existed even when the adelgid was reared in a common environment, indicating a genetic basis for the variation in cold hardiness. These findings support the hypothesis that the adelgid has adapted to the colder environment as it has expanded its distribution in its invaded range.

Chemical composition: Hearing insect defensive volatiles.

Project description:Chemical signals mediate major ecological interactions in insects. However, using bioassays only, it is difficult to quantify the bioactivity of complex mixtures, such as volatile defensive secretions emitted by prey insects, and to assess the impact of single compounds on the repellence of the entire mixture. To represent chemical data in a different perceptive mode, we used a process of sonification by parameter mapping of single molecules, which translated chemical signals into acoustic signals. These sounds were then mixed at dB levels reflecting the relative concentrations of the molecules within species-specific secretions. Repellence of single volatiles, as well as mixtures of volatiles, against predators were significantly correlated with the repulsiveness of their respective auditory translates against humans, who mainly reacted to sound pressure. Furthermore, sound pressure and predator response were associated with the number of different molecules in a secretion. Our transmodal approach, from olfactory to auditory perception, offers further prospects for chemo-ecological research and data representation.

Semen adaptation to microbes in an insect.

Project description:Sperm function is suggested to evolve by sexual selection but is also reduced by microbial damage. Here, we provide experimental evidence that male fertility can adapt to microbes. We found that in vivo, male fertility was reduced by one-fifth if sperm encountered microbes in the females that they had not previously been exposed to, compared to sperm from males that coevolved with these microbes. The female immune system activation reduced male fertility by an additional 13 percentage points. For noncoevolved males, fertility was larger if microbes were injected into females after they had stored away the sperm, indicating microbial protection as a previously unrecognized benefit of female sperm storage. Both medical and evolutionary research on reproductive health and fertility will benefit from considering our findings that the impact of microbes on sperm depends on their joint evolutionary history. Our results may assist in reconciling contradictory results of sexually transmitted disease effects on sperm and bring empirical realism to a recently proposed role of locally adapted reproductive microbiomes to speciation.

Roles of carboxyl groups in the transmembrane insertion of peptides.

Project description:We have used pHLIP® [pH (low) insertion peptide] to study the roles of carboxyl groups in transmembrane (TM) peptide insertion. pHLIP binds to the surface of a lipid bilayer as a disordered peptide at neutral pH; when the pH is lowered, it inserts across the membrane to form a TM helix. Peptide insertion is reversed when the pH is raised above the characteristic pK(a) (6.0). A key event that facilitates membrane insertion is the protonation of aspartic acid (Asp) and/or glutamic acid (Glu) residues, since their negatively charged side chains hinder membrane insertion at neutral pH. In order to gain mechanistic understanding, we studied the membrane insertion and exit of a series of pHLIP variants where the four Asp residues were sequentially mutated to nonacidic residues, including histidine (His). Our results show that the presence of His residues does not prevent the pH-dependent peptide membrane insertion at ~pH 4 driven by the protonation of carboxyl groups at the inserting end of the peptide. A further pH drop leads to the protonation of His residues in the TM part of the peptide, which induces peptide exit from the bilayer. We also find that the number of ionizable residues that undergo a change in protonation during membrane insertion correlates with the pH-dependent insertion into the lipid bilayer and exit from the lipid bilayer, and that cooperativity increases with their number. We expect that our understanding will be used to improve the targeting of acidic diseased tissue by pHLIP.

Insect-plant relationships predict the speed of insecticide adaptation.

Project description:Herbivorous insects must circumvent the chemical defenses of their host plants and, in cropping systems, must also circumvent synthetic insecticides. The pre-adaptation hypothesis posits that when herbivorous insects evolve resistance to insecticides, they co-opt adaptations against host plant defenses. Despite its intuitive appeal, few predictions of this hypothesis have been tested systematically. Here, with survival analysis of more than 17,000 herbivore-insecticide interactions, we show that resistance evolution tends to be faster when herbivorous insect diets are broad (but not too broad) and when insecticides and plant defensive chemicals are similar (but not too similar). These general relations suggest a complex interplay between macro-evolutionary contingencies and contemporary population genetic processes, and provide a predictive framework to forecast which pest species are most likely to develop resistance to particular insecticide chemistries.

Reduced Genome of the Gut Symbiotic Bacterium "Candidatus Benitsuchiphilus tojoi" Provides Insight Into Its Possible Roles in Ecology and Adaptation of the Host Insect.

Project description:Diverse animals, including insects, harbor microbial symbionts within their gut, body cavity, or cells. The subsocial parastrachiid stinkbug Parastrachia japonensis is well-known for its peculiar ecological and behavioral traits, including its prolonged non-feeding diapause period and maternal care of eggs/nymphs in an underground nest. P. japonensis harbors a specific bacterial symbiont within the gut cavity extracellularly, which is vertically inherited through maternal excretion of symbiont-containing white mucus. Thus far, biological roles of the symbiont in the host lifecycle has been little understood. Here we sequenced the genome of the uncultivable gut symbiont "Candidatus Benitsuchiphilus tojoi." The symbiont has an 804 kb circular chromosome encoding 606 proteins and a 14.5 kb plasmid encoding 13 proteins. Phylogenetic analysis indicated that the bacterium is closely related to other obligate insect symbionts belonging to the Gammaproteobacteria, including Buchnera of aphids and Blochmannia of ants, and the most closely related to Ishikawaella, an extracellular gut symbiont of plataspid stinkbugs. These data suggested that the symbiont genome has evolved like highly reduced gamma-proteobacterial symbiont genomes reported from a variety of insects. The presence of genes involved in biosynthesis pathways for amino acids, vitamins, and cofactors in the genome implicated the symbiont as a nutritional mutualist, supplementing essential nutrients to the host. Interestingly, the symbiont's plasmid encoded genes for thiamine and carotenoid synthesis pathways, suggesting the possibility of additional functions of the symbiont for protecting the host against oxidative stress and DNA damage. Finally, possible involvement of the symbiont in uric acid metabolism during diapause is discussed.

A chemical signal that promotes insect survival via thermogenesis.

Project description:Cold-activated thermogenesis of brown adipose tissues (BAT) is vital for the survival of animals under cold stress and also inhibits the development of tumours. The development of small-molecule tools that target thermogenesis pathways could lead to novel therapies against cold, obesity, and even cancer. Here, we identify a chemical signal that is produced in beetles in the winter to activate fat thermogenesis. This hormone elevates the basal body temperature by increasing cellular mitochondrial density and uncoupling in order to promote beetle survival. We demonstrate that this hormone activates UCP4- mediated uncoupled respiration through adipokinetic hormone receptor (AKHR). This signal serves as a novel fat-burning activator that utilizes a conserved mechanism to promote thermogenesis not only in beetles, nematode and flies, but also in mice, protecting the mice against cold and tumor growth. This hormone represents a new strategy to manipulate fat thermogenesis.
