Molecular Fingerprinting and Phytochemical Investigation of Syzygium cumini L. from Different Agro-Ecological Zones of India.

ABSTRACT: Syzygium cumini L. (ver Jamun; BlackBerry) is a native, evergreen multipurpose tree species of India. Besides being a fruit tree and for agroforestry in different regions, it is medicinally important too. This study aimed to determine genetic diversity using molecular and phytochemical markers in sixteen genotypes of Indian S. cumini from different agro-ecological zones. The present study used a combination of ISSR markers and the HPLC technique to explore these genotypes. The results showed a wide genetic diversity range based on the similarity coefficient values observed in S. cumini sixteen accessions from different sites. Four primary phenolic acids were discovered in all the accessions; caffeic acid (CA) was found in high concentrations. The intraspecific association between molecular and phytochemical characteristics was the primary goal of this investigation. By employing gene-specific markers for the route of secondary metabolites (polyphenols) production, it further investigated the progressive research of diversity analysis of polyphenol content in S. cumini accessions, which may also expand its nutraceutical and pharmaceutical utilization.

Project description:Genotype by environment interaction (GxE) complicates the process of selecting genotypes suitable for quantitative traits like seed yield in beans, hence slows down the development and release of varieties by breeding programs. GxE study on seed yield in beans enables identification of stable genotypes across sites and best site(s) for discriminating the tested genotypes in terms of seed yield. The purpose of this study was to evaluate the influence of the environment, genotype, and genotype by environment interaction on seed yield stability and adaptability of common bean landraces, lines, and improved varieties across three different agro-ecologies in Tanzania. The 99 common bean genotypes (Landraces, lines, and improved varieties) were planted following alpha lattice design in three replications each contained five blocks with 20 plots. Soil properties from the experimental sites, days to 75% flowering, Seed yield, 100 seed weight, number of seeds/pod, and number of pods/plant were recorded. Data on seed yield and its components were analyzed using Additive main effect and multiplicative interaction (AMMI), genotype main effects plus genotype × environment interaction (GGE), and yield stability index (YSI). The AMMI revealed very highly significant (P ≤ 0.001) effects of genotypes, environmental, and genotype × environment interaction on all the traits. AMMI analysis revealed that genotype main effects accounted for 39.3% of the total sum square of seed yield, whereas the environment and genotype × environmental interaction accounted for 31.4% and 26.8 % respectively. Genotype main effects largely influenced the variation in days to 75% flowering (55.5%), number of pods/plant (49.2%), number of seeds/pod (73.3%), and 100 seed weight (71.2%). Among soil properties recorded, available soil phosphorus, soil pH, soil exchangeable K, Ca, and Na had a strong positive association with common bean seed yield, while soil organic carbon and total nitrogen exhibited a strong negative association with seed yield. GGE revealed that E1 (TARI-Selian) was the most discriminative and representative site for common bean genotypes seed yield. Based on the yield stability index, the most stable and high seed yielding genotypes were ACC 714, Selian 14, Selian 9, Katuku, and Msolini. The identified high seed yielding and stable genotypes can be further tested in participatory variety selection involving farmers and later on released as varieties and can also be used for different breeding purposes in different agro-ecologies of Tanzania.

Project description:BACKGROUND:Ticks and tick-borne diseases are a major impediment to livestock production worldwide. Cattle trade and transnational transhumance create risks for the spread of ticks and tick-borne diseases and threaten cattle production in the absence of an effective tick control program. Few studies have been undertaken on cattle ticks in the Central African region; therefore, the need to assess the occurrence and the spatial distribution of tick vectors with the aim of establishing a baseline for monitoring future spread of tick borne-diseases in the region is urgent. RESULTS:A total of 7091 ixodid ticks were collected during a countrywide cross-sectional field survey and identified using morphological criteria. Of these, 4210 (59.4%) ticks were Amblyomma variegatum, 1112 (15.6%) Rhipicephalus (Boophilus) microplus, 708 (10.0%) Rhipicephalus (Boophilus) decoloratus, 28 (0.4%) Rhipicephalus (Boophilus) annulatus, 210 (3.0%) Hyalomma rufipes, 768 (10.8%) Hyalomma truncatum, and 19 (0.3%) Rhipicephalus sanguineus. Three ticks of the genus Hyalomma spp. and 33 of the genus Rhipicephalus spp. were not identified to the species level. Cytochrome c oxidase subunit 1 (cox1) gene sequencing supported the data from morphological examination and led to identification of three additional species, namely Hyalomma dromedarii, Rhipicephalus sulcatus and Rhipicephalus pusillus. The finding of the invasive tick species R. microplus in such large numbers and the apparent displacement of the indigenous R. decoloratus is highly significant since R. microplus is a highly efficient vector of Babesia bovis. CONCLUSIONS:This study reports the occurrence and current geographical distribution of important tick vectors associated with cattle in Cameroon. It appears that R. microplus is now well established and may be displacing native Rhipicephalus (Boophilus) species, such as R. decoloratus. This calls for an urgent response to safeguard the livestock sector in western central Africa.
