Characterization of the mitochondrial genome of the lawn cutworm Spodoptera depravata (Lepidoptera: Noctuidae).

ABSTRACT: The lawn cutworm, Spodoptera depravata, is one of the most important pests that causes economic damage to grass crops. This study reports the complete mitochondrial genome of an S. depravata sample collected in China. The genome is a circular molecule 15,460 bp in length with an overall A + T content of 81.6%. It contains 13 protein-coding genes, 22 transfer RNA genes, and two ribosomal RNA genes. The gene content and organization of the mitogenome of S. depravata are identical to those of other Spodoptera species. Maximum-likelihood phylogenetic analysis based on mitogenomes showed a close evolutionary relationship between S. depravata and S. exempta. This study provides new molecular data for the identification and further phylogenetic analyses of Spodoptera species.

Project description:The complete 15,553 bp mitochondrial genome of the western bean cutworm, Stricosta albicosta, (Lepidoptera: Noctuidae) was assembled from paired end Illumina HiSeq2500 read data. Annotation showed 13 predicted protein coding genes (PCGs), 22 tRNAs and 2 rRNAs have an order and orientation typical of insect mitochondrial genomes, and the derived rearrangement of tRNA-Met, -Ile and -Gln upstream of nad2 as in other Lepidoptera. A 79.3% A + T content resulted in a bias for codons with A or T in the 3rd position, and prevalence of synonymous substitutions suggest the effects of purifying selection on the mitochondrial genome sequence. Two microsatellite repeat motifs, (CA)10(AT)19 and (AT)12, are respectively located in intergenic spaces between tRNA-Glu and -Phe and tRNA-Leu and 16S rRNA. Mitochondrial phylogenomics was able to resolve sub-families within the Noctuidae, and suggest analogous analyses may be applicable across other lepidopteran Families.

Complete mitochondrial genome of Spodoptera littoralis (Lepidoptera: Noctuidae) from Egypt.

Project description:The polyphagous cotton leafworm (Spodoptera littoralis) is one of the most destructive herbivorous insects worldwide. The present study reports the complete mitochondrial genome of S. littoralis collected from Egypt. The circular-mapping mitogenome was 15,408 bp in length with an overall A + T content of 81.1%, encoding a common set of 37 genes, including 13 protein-coding genes (PCGs), 22 tRNA genes, and two rRNA genes. Most PCGs were found to use conventional ATN as the start codon and TAN as the stop codon. The phylogenetic tree based on the nucleic acid sequences of 13 shared PCGs of 29 Noctuidae species revealed that S. littoralis and Spodoptera litura are sister species. The data in this study will be helpful to understand geographical genetic variations, phylogenetic relationships, and species identification of S. littoralis.

Responses of the cutworm Spodoptera litura (Lepidoptera: Noctuidae) to two Bt corn hybrids expressing Cry1Ab.

Project description:To examine the responses of the secondary lepidopteran pest Spodoptera litura to two Bacillus thuringiensis (Bt) corn hybrids [5422Bt1 (Event Bt11), 5422CBCL (MON810)] expressing Cry1Ab, larval bioassays with Cry1Ab toxin, corn leaves or kernels and bagging on corn plants were conducted. The results showed that larvae displayed a similar performance when fed kernels, but not leaves of 5422Bt1, 5422CBCL and their near-isogenic non-Bt corn (5422). Significantly higher Cry1Ab amounts were detected in larvae fed leaves than kernels of both Bt hybrids, with different molecular weights of protein band in plants (72 and 90 kDa for 5422Bt1 and 5422CBCL, respectively), gut contents (65 kDa), feces (50 kDa), which indicated that larvae had lower ingestion, higher degradation and excretion of Cry1Ab when fed kernels not leaves of both Bt hybrids. Significantly higher levels of cadherin-like receptors and alkaline phosphatase transcripts were detected in larvae fed leaves than kernels of two Bt hybrids. Catalase, superoxide dismutase and glutathione-S-transferase activities in larvae fed 5422Bt1 leaves were significantly higher than that of 5422 treatments. Therefore, S. litura had low susceptibility to 5422Bt1 and 5422CBCL when larvae fed kernels not leaves of Bt corn. Additionally, S. litura presented a much stronger tolerance to 5422CBCL than 5422Bt1.

Effects of Pyriproxyfen on Female Reproduction in the Common Cutworm, Spodoptera litura (F.) (Lepidoptera: Noctuidae).

Project description:The common cutworm, Spodoptera litura, is a rapidly reproducing pest of numerous agricultural ecosystems worldwide. The use of pesticides remains the primary means for controlling S. litura, despite their negative ecological impact and potential threat to human health. The use of exogenous hormone analogs may represent an alternative to insecticides. Juvenile hormones (JHs) play an important role in the reproductive systems of female insects, but the effects of pyriproxyfen, a JH analog, on reproduction in S. litura were poorly understood. In this paper, we topically treated the newly emerged females with 20, 60, or 100 μg of pyriproxyfen to determine its effects on reproduction. Then, we examined the expression of vitellogenin (Vg) and three hormone receptors, USP, HR3, and EcR, using quantitative reverse transcription and real-time polymerase chain reaction (qRT-PCR), and found that pyriproxyfen up-regulated the expression of Vg, USP, and HR3, whereas the expression of EcR was unaffected. An analysis of fecundity showed that the peak oviposition day, lifespan, and oviposition period were progressively shortened as the pyriproxyfen dosage increased. We also found that pyriproxyfen decreased egg laying amount, whereas the number of mature eggs that remained in the ovarioles of dead females increased as the pyriproxyfen dosage increased. We examined oocytes using transmission electron microscopy and found that treatment with 100 μg of pyriproxyfen increased the metabolism by increasing the amount of rough endoplasmic reticulum and mitochondria in the primary oocytes. Our results suggest that the topical application of pyriproxyfen on newly emerged females can efficiently reduce reproduction in S. litura and may represent an alternative to the use of insecticides for controlling the agricultural pest.

The complete mitochondrial genome of the fall armyworm, Spodoptera frugiperda Smith, 1797 (Lepidoptera; Noctuidae), firstly collected in Korea.

Project description:The fall armyworm, Spodoptera frugiperda, is a serious pest in large numbers on more than 350 plant species in the world. We have determined a 15,388 bp mitogenome of S. frugiperda which includes 13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNAs. The base composition was AT-biased (81.3%). Phylogenetic trees present that Korean S. frugiperda placed in basal position of S. frugiperda clade. S. frugiperda mitochondrial genome can be used for understanding recent active migration of S. frugiperda.

Comparison of gene expression in three strains of Spodoptera Frugiperda (Insecta: Lepidoptera: Noctuidae)

Project description:The aim of this experiment was to compare the transciptome of the fall armyworm (Spodoptera frugiperda) strain SUS (a laboratory insecticide-susceptible standard) with organophosphate (OP) and pyrethroid (PYR) resistant strains were collected in cornfields located in Minas Gerais and Mato Grosso States, Brazil, in 2008 and maintained in the laboratory under selection with chlorpyrifos (at increasing discriminating doses from 100 up to 400 µg of insecticide/g of insect in a topical bioassay) or lambda-cyhalothrin (from 8.4 up to 27 µg of insecticide/g of insect) respectively The custom microarray used in this study was designed using the Agilent eArray platform (Agilent Technologies). A SurePrint HD (8×15k) expression array was designed using the base composition and the best probe methodologies to design sense orientation 60-mer probes with a 3′ bias. The FAW EST database (SPODOBASE) was used as the reference transcriptome (Negre, Hotelier et al. 2006). These sequences are derived from 8 cDNA libraries as follows: Sf1F: Fat body, Sf1H: Hemocyte, Sf1M: Midgut, Sf1P: Pool of various tissues, Sf2H: Immune Challenged hemocytes, Sf2L: Sf21 Cell lines sequences, Sf2M: Xenobiotic Induced Midgut and Sf9L: Sf9 cell lines sequences. All assembled contigs and singlets were kindly sent by the website maintainers. The BLAST2GO software v.2.3.1 (http://www.blast2go.org) was used to annotate the EST database. 60-mer probes were designed for all 7,552 assembled contigs and 5,519 annotated singlets (BlastX), totaling 13,071 sequences. For contigs encoding detoxification enzymes (P450s, GSTs and CEs) three probes were designed. Additional probe groups for 15 control genes were also included. Establishment and characterization of three embryonic cell lines of beet armyworm, Spodoptera exigua (Lepidoptera: Noctuidae).

Project description:Three cell lines (QAU-Se-E-1, -2 and -3, or Se-1, -2 and -3 for short) were established from eggs of beet armyworm (Spodoptera exigua) that have been passaged stably for more than 60 times in TNM-FH medium supplemented with 10 % fetal bovine serum. The cell lines consisted of round and spindle-shaped cells. The round cells accounted for 96.82, 84.34 and 83.16 % of the cells in the three cell lines, respectively, with cell diameters of 16.21 ± 0.72, 15.63 ± 0.58 and 13.06 ± 0.44 μm. Random amplified polymorphic DNA and analysis of the CO I gene showed that the three cell lines were all derived from S. exigua. Growth curves at passage 30 were determined and the results showed that the cell population doubling times were 59.03, 49.08 and 49.91 h, respectively. The three cell lines can be infected by S. exigua multiple nucleopolyhedrovirus (SeMNPV). Se-3 was extremely susceptible to the virus with an infection rate of 97.52 % 4 days after the inoculation and produced 2.02 × 10(6) OBs per mL of culture. Flow cytometry analysis showed that some of Se-1 and Se-2 cells had apoptosis after infection, whereas Se-3 cells did not. Bioassays showed that the virulence of the SeMNPV proliferated from Se-3 was similar to that from the insect with LC50 of 5.55 × 10(5) and 2.64 × 10(5) OBs/mL. Therefore, the cell lines can be used to study the SeMNPV-host interactions and mechanisms underlying the interactions.

Mortality Dynamics of Spodoptera frugiperda (Lepidoptera: Noctuidae) Immatures in Maize.

Project description:We characterized the dynamics of mortality factors affecting immature developmental stages of the fall armyworm, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae). Multiple decrement life tables for egg and early larval stages of S. frugiperda in maize (Zea mays L.) fields were developed with and without augmentative releases of Telenomus remus Nixon (Hymenoptera: Platygastridae) from 2009 to 2011. Total egg mortality ranged from 73 to 81% and the greatest egg mortality was due to inviability, dislodgement, and predation. Parasitoids did not cause significant mortality in egg or early larval stages and the releases of T. remus did not increase egg mortality. Greater than 95% of early larvae died from predation, drowning, and dislodgment by rainfall. Total mortality due to these factors was largely irreplaceable. Results indicate that a greater effect in reducing generational survival may be achieved by adding mortality to the early larval stage of S. frugiperda.

Characterization and phylogenetic analysis of the mitochondrial genome of Macdunnoughia hybrida (Lepidoptera: Noctuidae: Plusiinae).

Project description:Macdunnoughia hybrida is a polyphagous herbivorous moth within the family Noctuidae. In this study, we sequenced and analyzed the complete mitochondrial genome (mitogenome) of M. hybrida. This mitogenome was 15,382 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA unit genes (rRNAs). Except for cox1 started with CGA, all other PCGs started with the standard ATN codons. Most of the PCGs terminated with the stop codon TAA, whereas cox1, cox2 and nad4 end with the incomplete codon T--. The whole mitogenome exhibited heavy AT nucleotide bias (80.7%). Gene order was conserved and identical to most other previously sequenced Noctuidae. Phylogenetic analysis positioned M. hybrida in a well-supported clade with Diachrysia nadeja and three Ctenoplusia species. The relationships (Catocalinae + (Plusiinae + (Acronictinae + (Heliothinae + (Amphipyrinae + (Hadeninae + Noctuinae)))))) were supported within Noctuidae.

Transcriptome Analysis of Detoxification-Related Genes in Spodoptera frugiperda (Lepidoptera: Noctuidae).

Project description:Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) is an important pest on maize, and it can cause large yield losses. As S. frugiperda has invaded many developing countries in Africa and Asia in recent years, it could impact food security. Pesticides remain the main method to control S. frugiperda in the field, and this pest has developed resistance to some pesticides. In this study, we used second-generation sequencing technology to detect the gene expression change of S. frugiperda after treatment by LC20 of three pesticides, lufenuron, spinetoram, and tetrachloroamide, which have different modes of actions. The sequence data were first assembled into a 60,236 unigenes database, and then the differential expression unigenes (DEUs) after pesticide treatment were identified. The DEU numbers, Gene Ontology catalog, and Kyoto Encyclopedia of Genes and Genomes pathway catalog were analyzed. Finally, 11 types of unigenes related to detoxification and DEUs after pesticide treatment were listed, and Cytochrome P450, Glutathione S-transferase, and ATP-binding cassette transporter were analyzed. This study provides a foundation for molecular research on S. frugiperda pesticide detoxification.
