First Report of Aphelenchoides Bicaudatus (Imamura, 1931) Filipjev and Schuurmans Stekhoven, 1941 Associated with Grass in South Africa.

ABSTRACT: Aphelenchoides bicaudatus associated with grass in South Africa was identified morphologically and molecularly. This population is characterized by a body length of 409 - 529 μm, a stylet length of 9.5 - 13 μm, a post-vulval uterine sac of 45 - 50 μm, and the characteristic tail bifurcated at the end with one prong longer than the other. Molecular analyses based on the 18S and ITS rDNA data confirmed the primary morphological identification of the A. bicaudatus species. The obtained phylogenetic trees revealed a close positioning of the South African population to other representatives of A. bicaudatus with the maximum (1.00) posterior probability value. Principal component analysis (PCA) also indicated a variation within the populations of A. bicaudatus. This is the first report of A. bicaudatus from South Africa.

First Report of Tripylina Zhejiangensis Associated with Grassland in South Africa.

Project description:Specimens of Tripylina zhejiangensis, collected from natural grass in the Free State Province of South Africa, were identified morphologically and molecularly. This species characterizes by body length (1037 - 1128 μm), a = 27.3 - 35.3, b = 5.1 - 5.6, c = 16.1 - 19.8, c' = 2.5 - 3.4, V = 62 - 65. Molecular analysis was also undertaken, based on the 28S rDNA regions, and confirmed this population as T. zhejiangensis. Phylogenetic analysis using the Bayesian inference method, places this population in a clade close to T. zhejiangensis with a 1.00 posterior probability value. According to the knowledge, this is the first report of the genus Tripylina and its species from South Africa. Besides, this is the second report of T. zhejiangensis worldwide after the original description from China.

First report of Meloidogyne hapla on kiwifruit in South Africa.

Project description:Kiwi is becoming one of the most important fruit in subtropical regions of South Africa with altitudes that confer sufficient chilling requirements. During a survey on biodiversity of plant-parasitic nematodes of kiwi in Magoebaskloof in Limpopo Province, several plant-parasitic nematodes were discovered, with Meloidogyne species occurring at the highest frequency. Nematodes were sampled from roots and the rhizosphere of one stunted Kiwi tree, extracted using the tray method and then fixed. The morphological characters fit well with those of M. hapla. The molecular approach using ITS and 28S rDNA, along with the related phylogenetic analysis, placed the examined population in a group with other populations of M. hapla. Kiwi is being reported as a new host for M. hapla in South Africa.

First report of Bitylenchus ventrosignatus () associated with wild grass in Botswana.

Project description:During a survey on the biodiversity of plant-parasitic nematodes of natural areas in Botswana, Bitylenchus ventrosignatus was discovered around the rhizosphere of wild grass. The nematodes were extracted using the tray method and then fixed according to the available protocols. The morphological characters fit well with the description of B. ventrosignatus. In addition, molecular analysis using 18 S and 28 S rDNA indicated 98% (KJ461617) and 95% (KJ461567) similarity with the Spanish population of B. ventrosignatus. The phylogenetic analysis of 18 S and 28 S rDNA placed the examined population with other populations of B. ventrosignatus in a group with a posterior probability support value of 100. According to published literature, this is the first report of B. ventrosignatus from Botswana.

Observation on Hemicriconemoides Brachyurus () Chitwood & Birchfield, 1957 Associated with Grass in South Africa.

Project description:Hemicriconemoides brachyurus was identified morphologically, using body annuli number (103105), stylet length (49-52 μm), tail conical with small rounded lobe, spermatheca with round sperm, and VL/VB (1.3). Molecular analysis was also undertaken, based on the 18S rDNA region, and the obtained DNA sequence data confirmed the present population from South Africa as Hemicriconemoides. Phylogenetic analysis using the Bayesian inference method places this population close to H. fujianensis from China (MH444620-21; MH444626-28). To the best of our knowledge, this is the first report of the 18S rDNA sequence for H. brachyurus.

X-linked hyper IgM (HIGM1) in an African kindred: the first report from South Africa.

Project description:BackgroundThe objective of this study was to describe the clinical and molecular features of the first South African family with X-linked hyper-IgM syndrome (HIGM1).MethodsDiagnoses were based on immunoglobulin results and the absence of CD40 ligand (CD40L) expression on activated T-cells. Complete molecular characterisation involved CD40L cDNA sequencing, and genomic DNA analysis by polymerase chain reaction amplification, restriction enzyme digestion and sequencing. A PCR-based diagnostic assay was established for carrier detection and prenatal diagnosis in this family.ResultsThere were originally six children, three males and three females. The eldest boy died after being diagnosed with hypogammaglobulinaemia, before HIGM1 was considered. This disorder was diagnosed in the second eldest boy at the age of 5 years, after failing to detect CD40L expression on his activated T-cells. A deficiency of CD40L was also confirmed in the youngest male at the age of 5 years. Both younger brothers have since died of infections relating to HIGM1. Molecular investigation showed that exon 3 was deleted from the CD40L mRNA of the affected males. Genomic DNA analysis identified a 1.5 kilobase deletion, spanning exon 3 and including extended flanking intronic sequence. Carrier status in the mother was confirmed by RT-PCR of her CD40L mRNA. Genetic analysis of the three female children was deferred because they were below the legal consenting age of 18 years. A PCR-based assay for genomic DNA was established for easy identification of female carriers and affected males in the future.ConclusionsThis study confirmed the diagnosis of HIGM1 in the first South African family to be investigated and identified a novel mutation in the CD40L gene.

Morphological and molecular characterization of Butlerius butleri Goodey, 1929 (Nematoda: Diplogastridae) from South Africa: First report.

Project description:Two populations of a Butlerius species were recovered from compost in two gardens in Potchefstroom, North-West Province, South Africa. Although the genus has previously been reported from South Africa, no species of the genus has ever been identified in the country. Based on morphological, morphometric, and molecular studies, the specimens were identified as Butlerius butleri and are herein reported for the first time from South Africa. The South African specimens are 1,082 to 1,423 µm long, a = 40.8 to 47.6; b = 4.7 to 5.8; c = 4.0 to 6.0; c' = 117 to 16.3; V = 44 to 47%. Cuticle with evenly spaced punctations. Reproductive system didelphic, amphidelphic, both branches equal in length. Four large glands opening into proximal part of uterus. Males with prominent sphincter present in mid-region of vas deferens. Spicules 36 to 43 μm long, gubernaculum 23 to 31 µm long, nine pairs of genital papillae, three pre-cloacal and six post-cloacal, formula: v1, v2, v3d/v4, ad, ph, v5, 6, 7, pd. The v5, 6, 7 clusters greatly separated, left subventral group at level of phasmid, right subventral group at level of posterior dorsal papilla. Although there were some differences, the South African populations of the species compare well to all know descriptions of the species. Phylogenetic analysis based on partial small subunit (SSU) rDNA sequences showed that both South African populations of B. butleri are in a maximally supported sister relation with an Iranian population of this species. Based on large subunit (LSU) rDNA sequences, the two populations of B. butleri clustered together in a well-supported clade.
