Controlling extrudate volume fraction through poroelastic extrusion of entangled looped fibers.

ABSTRACT: When a suspension of spherical or near-spherical particles passes through a constriction the particle volume fraction either remains the same or decreases. In contrast to these particulate suspensions, here we observe that an entangled fiber suspension increases its volume fraction up to 14-fold after passing through a constriction. We attribute this response to the entanglements among the fibers that allows the network to move faster than the liquid. By changing the fiber geometry, we find that the entanglements originate from interlocking shapes or high fiber flexibility. A quantitative poroelastic model is used to explain the increase in velocity and extrudate volume fraction. These results provide a new strategy to use fiber volume fraction, flexibility, and shape to tune soft material properties, e.g., suspension concentration and porosity, during delivery, as occurs in healthcare, three-dimensional printing, and material repair.

Loop extrusion driven volume phase transition of entangled chromosomes.

Project description:Experiments on reconstituted chromosomes have revealed that mitotic chromosomes are assembled even without nucleosomes. When topoisomerase II (topo II) is depleted from such reconstituted chromosomes, these chromosomes are not disentangled and form "sparklers," where DNA and linker histone are condensed in the core and condensin is localized at the periphery. To understand the mechanism of the assembly of sparklers, we here take into account the loop extrusion by condensin in an extension of the theory of entangled polymer gels. The loop extrusion stiffens an entangled DNA network because DNA segments in the elastically effective chains are translocated to loops, which are elastically ineffective. Our theory predicts that the loop extrusion by condensin drives the volume phase transition that collapses a swollen entangled DNA gel because the stiffening of the network destabilizes the swollen phase. This may be an important piece to understand the mechanism of the assembly of mitotic chromosomes.

The impact of vascular volume fraction and compressibility of the interstitial matrix on vascularised poroelastic tissues.

Project description:In this work we address the role of the microstructural properties of a vascularised poroelastic material, characterised by the coupling between a poroelastic matrix and a viscous fluid vessels network, on its overall response in terms of pressures, velocities and stress maps. We embrace the recently developed model (Penta and Merodio in Meccanica 52(14):3321-3343, 2017) as a theoretical starting point and present the results obtained by solving the full interplay between the microscale, represented by the intervessels' distance, and the macroscale, representing the size of the overall tissue. We encode the influence of the vessels' density and the poroelastic matrix compressibility in the poroelastic coefficients of the model, which are obtained by solving appropriate periodic cell problem at the microscale. The double-poroelastic model (Penta and Merodio 2017) is then solved at the macroscale in the context of vascular tumours, for different values of vessels' walls permeability. The results clearly indicate that improving the compressibility of the matrix and decreasing the vessels' density enhances the transvascular pressure difference and hence transport of fluid and drug within a tumour mass after a transient time. Our results suggest to combine vessel and interstitial normalization in tumours to allow for better drug delivery into the lesions.

Poroelastic osmoregulation of living cell volume.

Project description:Cells maintain their volume through fine intracellular osmolarity regulation. Osmotic challenges drive fluid into or out of cells causing swelling or shrinkage, respectively. The dynamics of cell volume changes depending on the rheology of the cellular constituents and on how fast the fluid permeates through the membrane and cytoplasm. We investigated whether and how poroelasticity can describe volume dynamics in response to osmotic shocks. We exposed cells to osmotic perturbations and used defocusing epifluorescence microscopy on membrane-attached fluorescent nanospheres to track volume dynamics with high spatiotemporal resolution. We found that a poroelastic model that considers both geometrical and pressurization rates captures fluid-cytoskeleton interactions, which are rate-limiting factors in controlling volume changes at short timescales. Linking cellular responses to osmotic shocks and cell mechanics through poroelasticity can predict the cell state in health, disease, or in response to novel therapeutics.

Influence of extrusion cooking on phytochemical, physical and sorption isotherm properties of rice extrudate infused with microencapsulated anthocyanin.

Project description:The effect of extrusion cooking on the quality of rice extrudate with infused microencapsulated anthocyanin was investigated. The moisture sorption isotherm of the extrudate was also studied. The rotatable central composite design was used to optimize the extrusion process and the optimized conditions were: screw speed, 121 rpm; barrel temperature, 91.89 °C; and moisture content, 22.03%. The extrudate showed anthocyanin content of 0.218 mg/L; true density, 1.48 g/cc; water activity 0.51, water solubility index, 7.49%; and specific mechanical energy, 31.39 kJ/kg. The antioxidant activity and solubility of the extrudate were higher as compared with native extrudate. The moisture sorption isotherm of the extrudate was found to follow type III isotherm behavior according to the Brunauer-Emmett-Teller classification. The sorption isotherm was analyzed using several models and the Caurie and Peleg models were best fitted with the extrudate isotherm data. The present work manifested a way to develop antioxidant rich extrudate.

Entangled ZnO on Ultrathin Hollow Fibers for UV-Aided Pollutant Decomposition.

Project description:Zinc oxide (ZnO), a widely used ultraviolet (UV) degrading substance, offers high selectivity for wastewater treatment, but the leaching of ZnO into water could cause secondary contamination. Using porous substrates to fix and load ZnO is a promising technical method to improve the water purification efficiency and recycling durability of ZnO. However, limited by the slow kinetics and shielding effects, it is challenging to use traditional techniques to introduce ZnO into the interior of a hollow structure. Here, inspired by an ancient dyeing procedure, we formed a unique single-molecule bio-interfacial entanglement as an absorption layer to capture the catalyst for ZnO electroless deposition (ELD) on the surface of natural ultrathin hollow-structured Kapok fibers. With curcumin serving as a linking bridge, ELD allowed the spontaneous formation of intensive ZnO nanocrystals on both the outer and inner walls. ZnO-kapok as the catalyst for ultraviolet photodecomposition of organic pollutants (methylene blue (MB) and phenol as model pollutants) delivered a decomposition efficiency of 80% and outstanding durability. Further modification of the ZnO-kapok catalyst by doping with reduced graphene oxide (rGO) showed an improvement in photodegradation performance of 90% degradation under 2-h irradiation with 21.85 W/dm2 light power. Moreover, to the best of our knowledge, this is the first report featuring ZnO loading on both the outer and inner walls of a fiber-structured hollow kapok material, which provides inspiration for immobilization of metallic oxides on hollow-structured materials for further applications in renewable catalysis, chemical engineering, and energy storage fields.

Entropic Competition between Supercoiled and Torsionally Relaxed Chromatin Fibers Drives Loop Extrusion through Pseudo-Topologically Bound Cohesin.

Project description:We propose a model for cohesin-mediated loop extrusion, where the loop extrusion is driven entropically by the energy difference between supercoiled and torsionally relaxed chromatin fibers. Different levels of negative supercoiling are controlled by varying imposed friction between the cohesin ring and the chromatin fiber. The speed of generation of negative supercoiling by RNA polymerase associated with TOP1 is kept constant and corresponds to 10 rotations per second. The model was tested by coarse-grained molecular simulations for a wide range of frictions between 2 to 200 folds of that of generic fiber and the surrounding medium. The higher friction allowed for the accumulation of higher levels of supercoiling, while the resulting extrusion rate also increased. The obtained extrusion rates for the given range of investigated frictions were between 1 and 10 kbps, but also a saturation of the rate at high frictions was observed. The calculated contact maps indicate a qualitative improvement obtained at lower levels of supercoiling. The fits of mathematical equations qualitatively reproduce the loop sizes and levels of supercoiling obtained from simulations and support the proposed mechanism of entropically driven extrusion. The cohesin ring is bound on the fibers pseudo-topologically, and the model suggests that the topological binding is not necessary.

Segmented, Side-Emitting Hydrogel Optical Fibers for Multimaterial Extrusion Printing.

Project description:Side-emitting optical fibers allow light to be deliberately outcoupled along the fiber. Introducing a customized side-emission profile requires modulation of the guiding and emitting properties along the fiber length, which is a particular challenge in continuous processing of soft waveguides. In this work, it is demonstrated that multimaterial extrusion printing can generate hydrogel optical fibers with tailored segments for light-side emission. The fibers are based on diacrylated Pluronic F-127 (PluDA). 1 mm diameter fibers are printed with segments of different optical properties by switching between a PluDA waveguiding ink and a PluDA scattering ink containing nanoparticles. The method allows the fabrication of fibers with segment lengths below 500 microns in a continuous process. The length of the segments is tailored by varying the switching time between inks during printing. Fibers with customized side-emission profiles along their length are presented. The functionality of the printed fibers is demonstrated by exciting fluorescence inside a surrounding 3D hydrogel. The presented technology and material combination allow unprecedented flexibility for designing soft optical fibers with customizable optical properties using simple processes and a medical material. This approach can be of interest to improve illumination inside tissues for photodynamic therapy (PDT).

Myelin volume fraction imaging with MRI.

Project description:MRI is a valuable tool to assess myelin during development and demyelinating disease processes. While multiexponential T2 and quantitative magnetization transfer measures correlate with myelin content, neither provides the total myelin volume fraction. In many cases correlative measures are adequate; but to assess microstructure of myelin, (e.g. calculate the g-ratio using MRI), an accurate measure of myelin volume fraction is imperative. Using a volumetric model of white matter, we relate MRI measures of myelin to absolute measures of myelin volume fraction and compare them to quantitative histology. We assess our approach in control mice along with two models of hypomyelination and one model of hypermyelination and find strong agreement between MRI and histology amongst models. This work investigates the sensitivities of MRI myelin measures to changes in axon geometry and displays promise for estimating g-ratio from MRI.

Void Volume Fraction of Granular Scaffolds.

Project description:Void volume fraction (VVF) is a global measurement frequently used to characterize the void space of granular scaffolds, yet there is no gold standard by which to measure VVF in practice. To study the relationship between VVF and particles of varying size, form, and composition, a library of 3D simulated scaffolds is used. Results reveal that relative to particle count, VVF is a less predictable metric across replicate scaffolds. Simulated scaffolds are used to explores the relationship between microscope magnification and VVF, and recommendations are offered for optimizing the accuracy of approximating VVF using 2D microscope images. Lastly, VVF of hydrogel granular scaffolds is measured while varying four input parameters: image quality, magnification, analysis software, and intensity threshold. Results show that VVF is highly sensitive to these parameters. Overall, random packing produces variation in VVF among granular scaffolds comprising the same particle populations. Furthermore, while VVF is used to compare the porosity of granular materials within a study, VVF is a less reliable metric across studies that use different input parameters. VVF, a global measurement, cannot describe the dimensions of porosity within granular scaffolds, and the work supports the notion that more descriptors are necessary to sufficiently characterize void space.

Grasping through dynamic weaving with entangled closed loops.

Project description:Pick-and-place is essential in diverse robotic applications for industries including manufacturing, and assembly. Soft grippers offer a cost-effective, and low-maintenance alternative for secure object grasping without complex sensing and control systems. However, their inherent softness normally limits payload capabilities and robustness to external disturbances, constraining their applications and hindering reliable performance. In this study, we propose a weaving-inspired grasping mechanism that substantially increases payload capacity while maintaining the use of soft and flexible materials. Drawing from weaving principles, we designed a flexible continuum structure featuring multiple closed-loop strips and employing a kirigami-inspired approach to enable the instantaneous and reversible creation of a woven configuration. The mechanical stability of the woven configuration offers exceptional loading capacity, while the softness of the gripper material ensures safe and adaptive interactions with objects. Experimental results show that the 130 g·f gripper can support up to 100 kg·f. Outperforming competitors in similar weight and softness domains, this breakthrough, enabled by the weaving principle, will broaden the scope of gripper applications to previously inaccessible or barely accessible fields, such as agriculture and logistics.
