Respiratory complex I (NADH:ubiquinone oxidoreductase) is essential for cellular energy production and NAD<sup>+</sup> homeostasis. Complex I mutations cause neuromuscular, mitochondrial diseases, such as Leigh Syndrome, but their molecular-level consequences remain poorly understood.
