EDES1.0 Eindhoven Diabetes Simulator

ABSTRACT: First version (1.0) of the Eindhoven Diabetes Simulator (EDES) model, describing postprandial glucose and insulin dynamics for a healthy human. Model can also be used to simulate insulin resistance (pre-diabetes, Metabolic Syndrome) and Type 2 Diabetes Mellitus (T2DM). Next to simulating a meal, the model can simulate oral glucose tolerance tests (OGTT's).

Gene expression Fulminant type 1 diabetes vs classical type 1A diabetes

Project description:We determined the genes that were differentially expressed between fulminant type 1 diabetes and classical type 1A diabetes using gene expression microarray in peripheral blood cells. Ten patients with type 1 diabetes (5 fulminant and 5 classical type 1A) were enrolled in this study. All patients had recovered from ketosis or ketoacidosis at disease onset and had been receiving intensive insulin injection therapy for at least 1 month, which means that all patients and healthy controls were free of metabolic derangements at the time of blood sampling. To determine the genes that were differentially expressed between classical type 1A and fulminant type 1 diabetes, a volcano plot analysis (fold change >1.5 and p-value <0.05) was performed.

Project description:Due to the insidious onset and the difficulty to diagnose and intervene, it is particularly important to find the diagnostic markers of diabetic coronary microcirculation disorder (CMD) and therapeutic targets. Circulating exosomes, as endogenous exosomes derived from multiple tissue cells, mediate communication between cells and tissues, may serve as a key point for future diagnosis, prognosis and personalized treatment of CMD in diabetes. Therefore, we isolated serum exosomes from patients with diabetes, as well as patients with CMD or coronary artery disease (CAD) respectively. We explored the differences in characteristics between three circulating exosomes.

Aiming to analyze such hypothesis, patients submitted to gastric bypass for morbid obesity, gastrectomy for gastric cancer and colectomy for colo-rectal cancer will be compared. The end point will be changes in fasting blood glucose and hemoglobin A1c concentration.
