Demonstration of a single-visit algorithm to improve breast care efficiency in rural Zambia

ABSTRACT: Simulating Zambia's “screen and treat” approach to cervical cancer prevention, we designed and implemented a single-visit algorithm to improve breast care efficiency. Two breast care outreach camps were implemented, each of one-week duration, during which breast self-awareness, psychosocial counseling, clinical breast examination, ultrasound, ultrasound-guided biopsy, imprint cytology of biopsy specimens, surgical treatment or referral, were offered to participants within a single-visit format.Similar to the “screen and treat” approach for cervical cancer prevention, a single-visit algorithm has the potential to vastly improve breast care efficiency in low-resource environments.

Modified loop2 epegRNA improve prime editing efficiency.

Project description:Prime editor (PE) has wide application prospects in disease treatment due to its diversity of editing outcomes. However, the editing efficiency of PE still needs to be further improved for therapeutic applications. Here, we increase the probability of hybridization of the pegRNA primer binding site (PBS) to single-stranded DNA flap by adding additional PBS sequence and reverse transcription template (RTT) to the loop region of pegRNA. The selection of loop and the design of loop length are optimized. The resulting modified loop2 epegRNA (ML-epegRNA) showed higher prime editing efficiency than that of the epegRNA in HEK293T cells. In addition, we used ML-epegRNA for PE editing of multiple editing types in multiple cell lines, and the results showed a general improvement in editing efficiency. Overall, the ML-epegRNA expands the capabilities of genome editing tools.

Dnmt3l-knockout donor cells improve somatic cell nuclear transfer reprogramming efficiency

Project description:We found that Dnmt3l-KO donor cells display decreased levels of H3K9me3 and H3K27me3 accumulation, higher level of active histone mark H3K27ac and increased cytoplasmic localization of HDAC1, implicating a permissive epigenetic state beneficial for nuclear reprogramming. To investigate the gene expression profiles of MEFs and to find out the potential genes responsible for the improvement of Dnmt3l-KO cloned embryos.

Two Supporting Factors Greatly Improve the Efficiency of Human iPS Cell Generation

Project description:Human fibroblasts can be induced into pluripotent stem cells (iPS cells), but the reprogramming efficiency is quite low. Here, we screened a panel of candidate factors in the presence of OCT4, SOX2, KLF4 and c-MYC in an effort to improve the reprogramming efficiency from human adult fibroblasts. We found that p53 siRNA and UTF1 enhanced the efficiency of iPS cell generation up to 100-fold, even when the oncogene c-MYC was removed from the combinations. We further demonstrated that by using a novel combination of the four factors OCT4, SOX2, KLF4 and UTF1, iPS cells could be generated at a frequency at least 10 times higher than using the original four reprogramming factors without c-MYC. The iPS cells generated in this work have a similar gene expression profile and differentiation potential as human embryonic stem (hES) cells. In conclusion, two novel supporting factors that increase the efficiency of direct reprogramming have been identified, and a more-efficient method for the generation of human iPS cells has been developed in the absence of the oncogene c-MYC. Keywords: cell type comparison Total RNA from hFSF, hAFF, hES cells (H1, H7) and 7 established iPS cell lines were labeled with Cy5, hybridized to a human Oligo Microarray (Phalanx Human Whole Genome OneArray™, Phalanx Biotech) according to the manufacturer's protocol. Three technical repetitions were performed. After hybridization, arrays were scanned using GenePix 4000B scanner (Molecular Devices) and processed using the GenePix Pro 6.0 software (Molecular Devices). After removing control probes, a 14/33 presence call (SNR>=5 and foreground-background>0) was used to filter probes for the 33 microarrays, resulting in 12311 probes for further quantile normalization.

Optimizing sgRNA structure to improve CRISPR-Cas9 knockout efficiency

Project description:The current commonly used single-guide RNA (sgRNA) structure has a shortened duplex compared with the native bacterial clustered regularly interspaced short palindromic repeats RNA (crRNA)–transactivating crRNA (tracrRNA) duplex. Here we show that modifying the sgRNA structure by extending the duplex length and mutating the fourth T of the continuous sequence of Ts (which is the pause signal for RNA polymerase III [pol III]) to C or G significantly, and sometimes dramatically, improves knockout efficiency in cells. In addition, the new sgRNA structure also significantly increases the efficiency of more challenging genome-editing procedures, such as gene deletion, which is important for inducing a loss-of-function in non-coding genes.
