Microbial community functional genes under 20-year warming and harvesting


ABSTRACT: The study was conducted at the Kessler Atmospheric and Ecological Field Station (KAEFS) in central Oklahoma in the Great Plains, U.S. (34° 58′ 31.8″ N, 97° 31′ 19.6″W). The experiment began in 1999 after the site was not used for cultivation and grazing for 40 years. There are four treatments (6 replicates for each treatment): Control (Ctr), Warmed (W), Harvested (H), Warmed and Harvested (W+H). We sampled soil from each plot in 2019. Soil total DNA was extracted from 1.5 g soil using cryogenic grinding and SDS-based lysis, as described previously (77), and purified with a PowerSoil DNA Isolation Kit (MO BIO Laboratories, Carlsbad, CA). For soils from 2019, the functional capacity of microbial communities was determined by GeoChip 5.0
